Todays requirements as applied to manufacturers of<br />
pharmaceuticals are higher than ever before. Safety of<br />
course has absolute priority; however, the shortening<br />
of development time required for the research and development<br />
process, the consequent reduction of costs<br />
and the lengthening of the time during which the<br />
developed drug can be marketed are also important.<br />

Merck has been active in chromatography for more than 100 years.<br />
Based on this long tradition of highest quality silica-based resins,<br />
Merck today is the world’s largest producer of silica gels for chromatographic<br />
applications.<br />

Amino Acids for <strong>Processing</strong><br />
Amino acids are the building blocks of life, the substances<br />
that proteins are made of. L-Amino acids and their derivatives<br />
are key compounds in the life sciences. They are used<br />
in large quantities as raw materials, excipients and active<br />
ingredients in the pharmaceutical and chemical industries.<br />
In the area of biotechnology, they are essential components<br />
in practically all fermentation and cell culture processes.<br />
The quality demanded of such amino acids for use in<br />
pharmaceutical applications is prescribed by procedures<br />
contained in the most common used pharmacopoeias (DAB,<br />
Ph Eur, USP, etc.). In addition, when used in biotechnological<br />
applications, particularly for the manufacture of pharmaceutical<br />
active ingredients, further requirements have to<br />
be fulfilled with respect to product properties. Our stringent<br />
analytical control procedures ensure high and uniform<br />
quality and excellent batch-to-batch consistency.<br />
Amino acids for pharmaceutical applications<br />
These are tested according to the relevant current<br />
pharmacopoeias Ph Eur, BP and USP as mentioned in the<br />
product name.<br />
Amino acids for biochemistry<br />
These are mainly used in research and development. The<br />
quality and specifications of these products is therefore<br />
performed according to this application. Essential contents<br />
of the specifications are purity, optical purity, the absence<br />
of biological inhibitors (heavy metals, salts, etc.) and<br />

Biological Buffers for <strong>Processing</strong><br />
Working with biologically active<br />
molecules invariably involves the use<br />
of buffer solutions. The task of the<br />
buffer is to create a defined environment<br />
for the biomolecules so that they<br />
can be kept in a stable and native<br />
form and stays fully active. The buffer<br />
itself, however, should not exert any<br />
influence on the compound. Equally<br />
important, the buffer should not<br />
contain any impurities that may have<br />
a negative effect on the activity or<br />
stability of the molecule. Hence, only<br />
highly-purified substances of defined<br />
specifications should be used to ensure<br />
reproducible processes on a lab or<br />
production scale.<br />
Merck supplies buffers in various<br />
grades. The differences between them<br />
rarely involve purity itself but more<br />
the additional parameters that are<br />
checked prior to delivery. For<br />
example, whilst the nucleases may be<br />
an important quality criterion for<br />
biotechnological applications, the<br />
fulfilling of the requirements of a<br />
particular pharmacopoeia may well be<br />
critical when it comes to manufacturing<br />
a pharmaceutical active ingredient.<br />
However, for certain applications,<br />
it may be desirable to specify<br />
additional quality parameters not<br />
included in our “Certificate of<br />
Analysis”. Should you require any<br />

Downstream processing of proteins<br />
and other biomolecules represents a<br />
key role in the production of therapeutic<br />
substances. The major requirement<br />
is to maintain the biological activity of<br />
these biomolecules during purification,<br />
i.e. maintaining the intact native<br />
structure through-out the separation<br />
process. Therefore, to avoid protein<br />
denaturation, most biochromatography<br />
unit operations employ aqueous<br />
buffer systems. After the complete<br />
purification process has been developed<br />
on a small scale in the lab, the<br />
protocols, transferred to large-scale<br />
production, must give the same results.<br />
Normally, the large-scale production<br />
of bio-molecules is a multi-step process<br />
where column chromatography<br />
plays a crucial role. The overall separation<br />
and purification sequence for a<br />
protein typically involves 5 -10 unit<br />
operations. Each step must be operated<br />
with the highest possible efficiency in<br />
order to optimize product yield, ensure<br />
feed stream<br />
centrifugation, filtration<br />
capture<br />
high throughput; reduction of volume<br />
intermediate purification<br />
higher resolution; removal of host cell proteins<br />
final polishing<br />
high resolution; removal of aggregates and DNA<br />
process reproducibility and minimize<br />
process economics. The purification<br />
strategy is to determine experimentally<br />
the shortest route from the starting<br />
material to the final product that meets<br />
the required specifications for contaminants<br />
and impurities. A schematic<br />
overview is given in Figure 1.<br />
Fractogel® <strong>EMD</strong> media enable the user<br />
to meet all relevant requirements.<br />
Capture steps require high flow rates<br />
with no loss of capacity. All Fractogel®<br />
<strong>EMD</strong> media provide excellent results<br />
for ion exchange, hydrophobic interaction,<br />
affinity and size exclusion<br />
chromatography. High capacities, high<br />
recoveries and high chemical and<br />
mechanical stability of Fractogel® <strong>EMD</strong><br />
provide the downstream specialist a<br />
powerful tool for protein separations.<br />
At least five times the number of cycles<br />
can be run on Fractogel® <strong>EMD</strong> resins<br />
using the same column packing as<br />
with soft gels based on carbohydrate<br />

New Robust Tentacle Media<br />
Tentacle surface technology, based on the grafting<br />
of functionally substituted acrylamides to<br />
the surface of porous particles, has now been<br />
transferred to a novel hydrophilic vinyl polymer<br />
resin. The resulting Fractoprep® ion exchange<br />
media show excellent protein binding capacities<br />
at high flow rates, with much improved mechanical<br />
and chemical stability. The resins are extremely<br />
resistant to sodium hydroxide treatment,<br />
the most important clean-in-place step in almost<br />
every production process and, in addition, pro-<br />
Ion Exchange Chromatography on Fractogel® <strong>EMD</strong> Tentacle Resins<br />
In ion exchange chromatography, charged molecules<br />
are separated on column materials that<br />
carry an opposite charge. The ionic groups of ion<br />
exchange resins are covalently bound to the gel<br />
matrix and are partially neutralized by counter<br />
ions present in the buffer. Because proteins are<br />
multivalent anions or cations, at low salt concentrations<br />
they will bind to stationary phases<br />
that carry the opposite charge. The binding is<br />
strongly influenced by the pH of the buffer. For<br />
efficient use of ion exchange chromatography, it<br />
is important to operate at pH values where the<br />
exchangers are mostly ionised and the biopolymers<br />
have a net charge (Table 1). Excessively<br />
high salt concentrations, however, cause<br />
shielding of the charges on the protein surface<br />
and effective binding of protein to an exchanger<br />
can no longer take place. Once bound to the<br />
matrix, biomolecules are in turn displaced by an<br />
increasing salt gradient or pH changes. Proteins<br />
with different net charges can be separated.<br />
Desorption of proteins from the column begins<br />
only with increasing salt concentrations or pH<br />
changes, when the protein no longer binds. The<br />
compounds that have a higher charge density are<br />
bound more firmly to the column while the<br />
others elute more rapidly. In contrast to strong<br />
ion exchangers, which can be used over a wide<br />
pH range, weak ion exchangers have changing<br />
degrees of ionization depending on the pH. Thus,<br />
the pH range over which you can use the ion<br />
exchangers is limited by these properties., e.g.<br />
Fractogel® <strong>EMD</strong> COO - can be used above pH 5,<br />
Fractogel® <strong>EMD</strong> DEAE below pH 8.5. The weak<br />
vide other features essential for application in<br />
bio-pharmaceutical production. Due to their<br />
excellent mechanical rigidity, Fractoprep® media<br />
are easy to handle and can be packed in large<br />
production columns without any problems. For a<br />

ion exchangers are used, in general, in situations<br />
where the pH of the load is consistent and doesn't<br />
change from run to run. Strong ion exchangers<br />
that are charged regardless of pH are generally<br />
used in production were the feed streams<br />
sometimes change.<br />
Recently, a new generation of Fractoprep®<br />
tentacle media was launched. Fractoprep® beads<br />
have a particle size range between 30 and 150 µm<br />
and were designed for capture steps. The new<br />
Fractoprep® ion exchangers provide high productivity<br />
especially in the capture step. Large<br />
pores enable a free diffusion of proteins into the<br />
beads and allow high throughput at very low<br />
back pressure. Three different types of ion<br />
exchangers, Fractoprep® DEAE, Fractoprep®<br />
TMAE and Fractoprep® SO 3 - , extend the product<br />
range of the well-known Fractogel® tentacle<br />

Advantages of the Tentacle Ion Exchangers<br />
With most ion exchange gels the functional<br />
groups are fixed directly, or with<br />
short linkers, onto the surface of the<br />
matrix. On any given gel matrix, only<br />
a certain number of ionic groups can<br />
be attached because of the chemistries<br />
used and the limited inner surface of<br />
the matrix. Once attached, the functional<br />
groups must be accessible in<br />
order for biopolymers to bind. The<br />
spatial requirements for this binding<br />
dictates the limit of the protein binding<br />
capacity for these kinds of ion exchangers.<br />
With tentacle ion exchangers,<br />
however, the functional groups are<br />
covalently anchored to the matrix via<br />
flexible linear polymer chains. Therefore,<br />
the binding capacity for proteins<br />
functional groups<br />

can be increased significantly. The<br />
tentacles allow biopolymers greater<br />
accessibility to the functional groups<br />
due to less steric hindrance (Figure 2).<br />
Especially for large proteins, tentacle<br />
media provide higher binding capacities<br />
compared to conventional<br />
supports. Due to the good selectivity<br />
and high efficiency, columns packed<br />
with tentacle media provide good<br />
chromatographic results and highly<br />
concentrated fractions can be obtained.<br />
The mechanical stability of all tentacle<br />
media also allows the use of high flow<br />
rates without a significant loss of<br />
capacity.<br />
Fractogel® <strong>EMD</strong> media as well as<br />
Fractoprep® media are pressure-stable<br />
and can be easily regenerated with<br />
sodium hydroxide.<br />

High performance hydrophobic interaction chromatography<br />
is a powerful technique for the isolation<br />
of proteins on an analytical and preparative<br />
scale. The method of hydrophobic interaction<br />
chromatography (HIC) is based on the interaction<br />
between hydrophobic ligands on the chromatographic<br />
support and hydrophobic areas located on<br />
the surface of proteins. The hydrophobic feature<br />
of a given protein is caused by certain amino acid<br />
residues like isoleucine, valine, leucine, phenylalanine<br />
and others.<br />
Although most of the hydrophobic amino acid<br />
residues are located in the center of the molecule,<br />
recent structure analyses have shown<br />
hydrophobic patches on the protein’s surface.<br />
These areas are often correlated with the biological<br />
function of the protein. The hydrophobic<br />
character of a protein is promoted by high salt<br />
and increased temperatures.<br />

affinity chromatography<br />
The high efficiency of the method is based on the<br />
interaction of a covalently bound chelating ligand<br />
immobilized on a chromatographic support with<br />
proteins containing a large number of histidine<br />
residues. Some recombinant proteins have His<br />
tags engineered at the end of the protein sequence<br />
to facilitate purification on IMAC resins. Iminodiacetic<br />
acid has been chosen as the metal-chelating<br />
ligand for Fractogel® <strong>EMD</strong> Chelate. The<br />
iminodiacetic acid residue is suitable as a chelating<br />
compound because a bidentate chelating<br />
moiety remains free after immobilization, onto<br />
which a metal ion can be co-ordinated. Various<br />
metal ions can be immobilized on the stationary<br />
phase (e.g. Cu 2+ ; Ni 2+ ; Co 2+ ; Zn 2+ ). Free coordination<br />
sites of the metal ions are used to bind<br />
different proteins and peptides.<br />
Compared to conventional affinity gels synthesised<br />
with spacer molecules, Fractogel® <strong>EMD</strong><br />
and Fractogel® <strong>EMD</strong> Phenyl (S) allow efficient<br />
purification steps. Both phases, with a particle<br />
size of 20-40 µm, are mechanically stable and<br />
resistant to biological and chemical degradation.<br />
The functional groups are on linear polymer<br />
chains. The ligands differ with respect to their<br />
hydrophobicity. Ligand density of the functional<br />
groups bound to the matrix is optimised by the<br />
tentacle technology so that excessive hydrophobicity<br />
is avoided. Strong hydrophobic resins<br />
inhibit recovery and resolution.<br />
Application areas<br />
Both hydrophobic phases are suitable for a wide<br />
range of applications. The main application areas<br />
for HIC is the purification of cytosolic proteins,<br />
antibodies, and recombinant proteins. Membrane<br />
proteins can also be isolated on Fractogel® <strong>EMD</strong><br />
Propyl (due to its weak hydrophobicity this resin<br />
is more suitable). For the elution of strong<br />
hydrophobic proteins, a detergent should be<br />
used. In addition HIC is suitable for the removal<br />
or exchange of non-ionic detergents.<br />
Chelate allows the binding of larger amounts of<br />
proteins. Because the immobilized metal ion can<br />
be arranged in the best position relative to the<br />
binding sites on the protein’s surface, proteins<br />
can be bound more tightly to Fractogel® <strong>EMD</strong><br />
Chelate than comparable conventional gels.<br />
Application areas<br />
The existence of surface-accessible histidine<br />
residues is important for the binding of a native<br />
protein. As recombinant proteins become more<br />
important, expression systems using a polyhistidine<br />
tag for the protein enable rapid isolation<br />
of those proteins by metal chelating affinity<br />
chromatography. With peptides, the alpha amino<br />
group also plays a role, so that peptides can be<br />
retained even if no histidine residues are present.<br />
In the case of peptides, other metal binding amino<br />
acids like cysteine and tryptophan contribute to<br />
the binding.

Advantages of tentacle-mediated<br />
thiophilic adsorption<br />
The binding mechanism of thiophilic adsorption<br />
chromatography is based on the specific interaction<br />
of a covalently bound sulphur-containing<br />
ligand immobilized on a chromatographic support<br />
with proteins or peptides. An electron<br />
donor-acceptor mechanism between the affinity<br />
ligand and tryptophan residues on the surface of<br />
the target molecule is responsible for the selective<br />
binding. Fractogel® <strong>EMD</strong> TA is designed specifically<br />
for the efficient isolation of antibodies. The<br />
synthesis is performed according to the tentacle<br />
technology where the group-specific ligands are<br />
present in high density. Thus, the thiophilic tentacle<br />
material has a high protein-binding capacity<br />
and is suitable for antibody purification on<br />
an analytical as well as a preparative scale. Due to<br />
the steric accessibility of the ligand, the resin provides<br />
minimized non-specific interactions with<br />
proteins combined with high protein binding capacities.<br />
The chemistry of the 3-S type ligand that<br />
is used for Fractogel® <strong>EMD</strong> TA allows significantly<br />
better thiophilic binding compared to the 2-S<br />
types which have more hydrophobic interaction.<br />
Application areas<br />
Thiophilic adsorption chromatography is specifically<br />
designed for the purification of immunoglobulins<br />
(monoclonal and polyclonal antibodies)<br />
including single-chain antibodies and chicken<br />
antibodies. Albumins are not adsorbed on thiophilic<br />
media, which often simplifies the effective<br />
separation of antibodies. The binding of the protein<br />
takes place mainly via accessible tryptophan<br />
and/or phenylalanine residues. Corresponding<br />
motifs can be observed within conserved regions<br />
of various antibodies. Antibodies of the IgM subclass<br />
can also bind to a Fractogel® <strong>EMD</strong> TA<br />
column. The binding of antibodies from different<br />
species to thiophilic adsorption supports offers<br />
advantages over protein A. Elution conditions at<br />
physiological pH values enable high recoveries of<br />
biologically active antibodies.<br />

Molecular Size Exclusion Chromatography on Fractogel® <strong>EMD</strong> BioSEC (S)<br />
This method, also known as gel filtration<br />
(GPC) or molecular sieve chromatography<br />
is based on the different size<br />
and shape of proteins. Proteins of<br />
different sizes penetrate into the internal<br />
pores of the beads to different<br />
degrees. Small protein molecules are<br />
retarded by the column while large<br />
molecules pass through more rapidly.<br />
A column packed with Fractogel® <strong>EMD</strong><br />
BioSEC can be used for the separation<br />
of proteins in the range between 5 kDa<br />
and 1,000 kDa (Figure 3). The mechanical<br />
stability of the resin allows higher<br />
flow rates compared to conventional<br />
soft gels. This is very helpful during<br />
regeneration and equilibration of the<br />
column. The pressure stability also enables<br />
the packing of larger columns<br />
that are needed for polishing steps in<br />
the production of bio-molecules.<br />

Application areas<br />
SEC is very easy to perform up through<br />
production scale and the results are<br />
very reproducible. SEC is generally<br />
performed late in the process when the<br />
number of protein contaminants are<br />
small, as in a “polishing” step. The<br />
goal is the final removal of possible<br />
impurities such as structurally similar<br />
or closely related forms of the product<br />
(aggregates, deamidated or oxidised<br />
product, etc.), host proteins, reagents<br />
and buffer substances, leachables from<br />
chromatographic supports, endotoxins,<br />
nucleic acids (DNA, RNA) or<br />
viruses. However, if a target protein is<br />
extremely large or small, SEC can be<br />
utilized earlier in the purification process.<br />
Principal applications of SEC on Fractogel®<br />
<strong>EMD</strong> BioSEC are purification of<br />
cytosolic proteins as well as recombinant<br />
proteins and, simultaneously, the<br />
determination of their molecular mass<br />
after the column has been calibrated.<br />
Membrane proteins can also be isolated<br />
by this technique and SEC has been<br />
used in blood plasma fractionation.<br />
With the introduction of recombinant<br />
proteins there has been an emphasis<br />
on the determination of aggregates<br />
and other by-products that co-purify<br />
with the target protein. Size exclusion<br />
chromatography is also very useful for<br />
the purification of monoclonal and<br />
polyclonal antibodies. Albumins with<br />
a molecular weight of about 60 kDa<br />
elute earlier than IgG or IgM molecules.<br />
Since no gradient elution has to be<br />
applied, a programmable gradient<br />
mixing system is not necessary and<br />
comparatively simple equipment is<br />
needed for operation. In addition, the<br />
method can be easily integrated into<br />
purification schemes and most of the<br />
operational steps can be automated.

Preparative column chromatography<br />
is a separation and purification method<br />
that plays an important role in purifying<br />
valuable compounds in research,<br />
pilot plant operation and production.<br />
This method can be used not only to<br />
purify substances to a very high degree,<br />
but to do this in a rapid and<br />
economical way.<br />
In practice, standardized sorbents are<br />
required. These provide a high degree<br />
of method reliability, direct transfer<br />
from analytical scale and optimised<br />
throughput per unit of time. Only silica<br />
gels with a defined pore structure fulfill<br />
the requirements of process chromatography.<br />
Merck has specialized over decades in<br />
the manufacture of standardised silica<br />
gel packing materials for preparative<br />
chromatography and offers a wide<br />
range of customized sorbents. The product<br />
range is completed with several<br />
special types, e.g. silica gels of the<br />
type LiChroprep® or highly purified<br />
acid-washed silica gels.<br />
Such gels provide the user with unique<br />
method reliability. Regardless of which<br />
chromatography process is used<br />
(HPTLC, HPLC, preparative chromatography),<br />
direct and rapid transfer to a<br />
production scale is guaranteed and<br />
without risk. This is ensured by the<br />
fact that identical raw materials are<br />

These particle size ranges are selected<br />
in such a way that an optimum in separating<br />
performance and permeability<br />
results for the chromatographic<br />
process used (DC, HPTLC, HPLC, preparative<br />
chromatography).<br />
Thus, once a successful configuration<br />
has been found on an analytical scale,<br />
it can be transferred rapidly to processes<br />
on a pilot plant or production<br />

The selectivity of Merck’s silica gels<br />

is identical. Resolution, however,<br />
decreases, according to theory,<br />

PharmPrep®<br />
PharmPrep® media are a new line<br />
of spherical silica sorbents designed<br />
for process chromatography in the<br />
pharmaceutical market.<br />
The first product to be made available,<br />
PharmPrep® CC for preparative<br />
chromatography, is a spherical, totally<br />
porous Silica Gel manufactured by a<br />
large-scale process from water glass<br />
and sulphuric acid to produce a large<br />
particle spherical product.<br />
The spherical materials have the<br />
follwing advantages for classical open<br />
column chromatography:<br />
Aluminium Oxide for Preparative Chromatography<br />
Aluminium oxide is one of the most<br />
versatile sorbents for preparative<br />
chromatography. Due to its weaker<br />
adsorption than silica gel, standard<br />
aluminium oxide can be used in<br />
specifically defined pH-ranges.<br />
The aluminium oxide matrix<br />
comprises octahedral and tetrahedral<br />
aluminium atoms coupled with<br />
oxygen, whereby the surface<br />
aluminium atoms are saturated with<br />
free hydroxyl groups. The different<br />
acidic centres (Broenstedt acid, Lewis<br />
acid and Lewis basic groups) result in<br />
the fact that, in contact to ionic salt<br />
solutions, aluminium oxides possess<br />
both anion and cation exchange<br />
properties. Its better pH-stability,<br />
especially in the alkaline range, in<br />
comparison with silica gels is worth<br />
mentioning.<br />
They are easily packed into columns;<br />
they have high bulk densities, which<br />
result in good loadability; and they<br />
exhibit low back pressures, allowing<br />
for fast elutions.<br />
PharmPrep® CC can be used in conjunction<br />
with spherical sorbents for<br />
performing process chromatography<br />
at any scale and at a reasonable cost.<br />
PharmPrep® CC is the ideal media for<br />
high-quality, high-performance separations<br />
at a true process scale.<br />
Aluminium oxide occurs in various<br />
crystal forms. For chromatography, the<br />
γ-structure in the form of aluminium<br />
oxide 90 with a mean pore diameter of<br />
90 Å is mostly used. Further structural<br />
types are aluminium oxide 60 (ε-type)<br />
of 60 Å pore diameter and aluminium<br />
oxide 150 (α-type) of 150 Å pore<br />
diameter, each of which possesses<br />
specific adsorption properties.<br />
Standardized aluminium oxide 90 for<br />
chromatographic adsorption analysis<br />
according to Brockmann is a sorbent<br />
of medium polarity and it is frequently<br />
used when the cationic exchange<br />
properties of the basic aluminium<br />
oxide are required. In addition, it is an<br />
alternative material to activated charcoal<br />
where this product cannot be<br />
used because of its organic character.

Column Packing Stands NW25, NW50 and NW100: Innovative Column Packing Technology<br />
Conventional packing methods for preparative<br />
columns use rapid filtration under pressure and<br />
at high linear flow rates. This technology often<br />
leads to bridging effects between the particles in<br />
the stationary phase that have a negative impact<br />
on the column performance. Our studies have<br />
shown that this undesirable side-effect can be<br />
overcome by employing a pulling instead of a<br />
pushing mechanism.<br />
The new column packing technology is based on<br />
a combination of vacuum suction with subsequent<br />
column bed compression; this stabilizes<br />
the packing. A common water-jet pump is used<br />
to generate the vacuum. The gel slurry is sucked<br />
through a packing reservoir into the column by<br />
applying low pressure at the bottom end of the<br />
column.<br />
This method allows precise adjustment of the desired<br />
packing volume or gel bed height. In the<br />
production environment, the new technology<br />

Packing and Unpacking the Packing Stands<br />
The required volume of gel slurry is prepared<br />
according to the physical parameters of the<br />
stationary phase and the column size. Using a<br />
funnel, the suspended material is filled into the<br />
packing tube attached to the column. Vacuum is<br />
applied. After the suspension has been sucked<br />
into the column tube, the packing tube is<br />
removed and the upper column cap lid (flange) is<br />
sealed. The column bed is then washed with<br />
3-5 column volumes of a suitable<br />
solvent and simultaneously<br />
compressed over several<br />
steps to a bed compression that<br />
depends on the sorbent type<br />
being used (see operating<br />
instructions). In this way, rapid<br />

· You can choose between an economic upper screw cap and the convenient<br />
chain-closing system for the 50 and 100mm ID packing stands.<br />
· Customized column lengths can be provided on request.<br />

Chromolith® Prep monolithic stationary phases are<br />
new ultra-pure silica phases that have special<br />
properties resulting from their bimodal pore<br />
structure. This structure is based on new “Sol-<br />
Gel” technology and consists of macropores and<br />
mesopores.<br />
The mesopores, with an average diameter of<br />
12 nm, form the fine porous structure of the<br />
column interior and create a very large surface<br />
area on which adsorption of the target compounds<br />
occurs.<br />
The large macropores, with a pore diameter<br />
ranging from 3 µm, form a dense network of<br />
pores and allow their high flow rate due to their<br />
low resistance factor. The resulting excellent<br />

The product description of each enzyme<br />
gives information on the origin<br />
of the product, enzyme activity and the<br />
numerical classification (EC number).<br />
Definitions of enzyme activity are<br />
often historical in nature and relatively<br />
arbitrarily selected. As long as there<br />
are no standard procedures and very<br />
few internationally recognized ones,<br />
one has to be most careful when comparing<br />
enzyme qualities. Initially, it<br />
β-Glucoronidase<br />
This preparation is used for the gentle hydrolysis of<br />
steroid conjugates, e.g. in the assay of urinary steroids<br />
and mucopolysaccharides.<br />
β-Glucoronidase/Arylsulfatase (from Helix pomatia)<br />
stabilized aqueous solution for biochemistry,<br />

Lysozyme<br />
Lysozyme is a mucolytic enzyme. It is used for<br />
bacteriolysis.<br />
Lysozyme catalyzes the hydrolysis of β-(1-4) bonds<br />
between N-acetylmuramic acid and 2-acetamido-2deoxy-D-glucose<br />
residues in mucopolysaccharides,<br />
mucopeptides and chitin.<br />
Lysozyme also gently breaks up bacterial cell walls, e.g.<br />
of micrococcus luteus or micrococcus lysodeicticus; in<br />
the presence of EDTA e.g. also of bacillus subtilis and<br />
escherichia coli.<br />

Pepsin<br />
Pepsin is any one of the small family of closely related<br />
aspartic proteinase enzymes.<br />
Pepsin is used for the hydrolysis of peptides and<br />
proteins, e.g. insulin.<br />
Preferential cleavage sites are Phe- and Leu-.<br />
Pepsin only attacks the peptide bonds; esters and<br />
amides are not hydrolyzed.<br />
Furthermore, the enzyme can be used for the pepsincatalyzed<br />
synthesis of oligopeptides.<br />

should be checked whether enzymes<br />
of identical origin are involved (e.g.<br />
plant cells, animal origin). Preparations<br />
from different manufacturers<br />
can be only compared if their activities<br />
have been determined under<br />
identical conditions. A few degrees<br />
of reaction temperature, different<br />
buffer systems, pH, different enzyme<br />
substrates or different reaction times<br />
make exact comparison practically<br />
impossible. Differently determined<br />
units can rarely be converted to each<br />
other. If a comparison is necessary, the<br />
only way to do so is by experiment.<br />
Pronase E<br />
Pronase E may be used for the gentle hydrolysis of<br />
proteins or polypetides.<br />
Compared with acid total hydrolysis, the enzymatic<br />
splitting amounts to approximately 70 -90 %, due to<br />
the very low substrate specificity of pronase E.<br />
The enzyme is also used for the isolation of amino-acidglycoside<br />
components from mucoproteins.<br />

Trypsin<br />
Trypsin is a serine endopeptidase which catalyzes the<br />
hydrolytic cleavage of proteins and peptides specifically<br />
at the carboxyl groups of basic amino acids L-Arginine<br />
and L-Lysine.<br />
Furthermore, esters and amide bonds of synthetic<br />
derivatives of these amino acids, e.g. N α-benzoyl-Larginine<br />
ethyl ester, are also hydrolyzed.<br />

Are you in charge of a laboratory?<br />
If so, you are also responsible for the safety and<br />
well-being of all persons working there as well as<br />
for the environment.<br />
Your duties include being aware of what might<br />
happen, establishing what must not happen, and<br />
implementing measures aimed at avoiding<br />
accidents.<br />
· When planning an experiment, decide whether<br />
hazardous substances are in fact required or<br />
whether a less hazardous substance might not be<br />
used instead. (To help you, we have published<br />
information in a Chemical Data Base under<br />
www.chemdat.de).<br />
· In addition to the risks that the substances<br />
themselves pose, you must establish if there are<br />
any risks specific to your factory or plant. Find out<br />
whether any hazardous chemical reactions could<br />
conceivably occur and decide how these can be<br />
avoided.<br />
· Specify all measures needed in order to assure<br />
personal health and safety, according to the<br />
ranking technical – organizational – personal, and<br />
check these are in place before any hazardous<br />
substances are handled.<br />
· Ensure that hazardous substances are shelved and<br />
stored in such a way that they can pose no threat<br />
to human health and the environment. Restrict the<br />
amounts of combustible substances used in the<br />
laboratory to the absolute minimum needed.<br />
· Issue workers with suitable, effective personal<br />
protective clothing and equipment. Ensure that it<br />
is kept clean and in good condition.<br />
· Compose a set of laboratory rules that cover all of<br />
the potential health and environmental risks that<br />
may be encountered in the laboratory, and also<br />
specify general preventive measures and rules of<br />
conduct.<br />
· Compile specific operating procedures for activities<br />
that involve the use of hazardous substances (see<br />
SafeDAT software (item no. 403032)). Display these<br />
operating procedures prominently within the laboratory,<br />
and use them to instruct workers at least<br />
once a year in the handling of hazardous substances<br />
and in the potential dangers of working with<br />
them.<br />
· Always strive to improve industrial safety and<br />
personal well-being in the area for which you are<br />
responsible.<br />

Do you work in a laboratory?<br />
When handling chemicals, always follow safety<br />
procedures, even if the chemical in question is not<br />
labeled as hazardous.<br />
· Make sure the laboratory is kept clean and tidy.<br />
· Use only such hazardous substances and<br />
equipment as are absolutely necessary for you to<br />
carry out your duties.<br />
· Always ensure that the sash front is down on any<br />
fume hood.<br />
· If possible, keep hazardous substances in their<br />
original Merck containers and packaging. These are<br />
able to withstand normal conditions of use and are<br />
suitably labeled.<br />
· If you need to use a different container for day-today<br />
use, ensure it is at least labeled with the name<br />
of the chemical, its hazard symbol(s) and clear text<br />
warnings. Keep containers with hazardous substances<br />
no higher than the height at which they<br />
can still be safely dispensed and shelved.<br />

· Where possible, keep very toxic and toxic<br />
substances under lock and key.<br />
·Keep any substance that emits harmful vapors or<br />
gases in a place that allows these to be permanently<br />
extracted.<br />
· Check the laboratory inventory at least annually<br />
and dispose of any chemicals that have become<br />
unusable or no longer required.<br />
· Never pipet by mouth. Always use a mechanically<br />
