Preface
Chemical testing
Todays requirements as applied to manufacturers of
pharmaceuticals are higher than ever before. Safety of
course has absolute priority; however, the shortening
of development time required for the research and development
process, the consequent reduction of costs
and the lengthening of the time during which the
developed drug can be marketed are also important.
Merck is highly involved as a partner in the pharmaceutical
industry and accompanies and supports the
efforts of our customers during the entire drug development
process. The materials available from our
LIFE SCIENCE PRODUCTS Division represent a comprehensive
range of applications extending from the
research laboratory to manufacture.
From the beginning of the development process
through scale-up and on to production, we can provide
our customers with the necessary help and support for
chemical synthesis and biotechnological processes in
the form of meaningful cooperation.
We fully comply with all relevant cGMP requirements
for production, analysis and packaging; this is constantly
demonstrated within the scope of customer audits and
FDA inspections.
Our customer-oriented product offerings of advanced
separation media for many types of process recovery
operations is complemented by a comprehensive range
of production-focused biochemicals. A selection of
such products and supportive information is reproduced
in this brochure. However, should the product
you require not be included, please do not hesitate to
contact us; we will be happy to help.
The name Merck stands for product diversity, competence,
flexibility, quality, safety and information.
Why not utilize this potential to the full?
Amino Acids for Processing
Amino acids are the building blocks of life, the substances
that proteins are made of. L-Amino acids and their derivatives
are key compounds in the life sciences. They are used
in large quantities as raw materials, excipients and active
ingredients in the pharmaceutical and chemical industries.
In the area of biotechnology, they are essential components
in practically all fermentation and cell culture processes.
The quality demanded of such amino acids for use in
pharmaceutical applications is prescribed by procedures
contained in the most common used pharmacopoeias (DAB,
Ph Eur, USP, etc.). In addition, when used in biotechnological
applications, particularly for the manufacture of pharmaceutical
active ingredients, further requirements have to
be fulfilled with respect to product properties. Our stringent
analytical control procedures ensure high and uniform
quality and excellent batch-to-batch consistency.
Amino acids for pharmaceutical applications
These are tested according to the relevant current
pharmacopoeias Ph Eur, BP and USP as mentioned in the
product name.
Amino acids for biochemistry
These are mainly used in research and development. The
quality and specifications of these products is therefore
performed according to this application. Essential contents
of the specifications are purity, optical purity, the absence
of biological inhibitors (heavy metals, salts, etc.) and
theabsence of foreign amino acids.
Biological Buffers for Processing
Working with biologically active
molecules invariably involves the use
of buffer solutions. The task of the
buffer is to create a defined environment
for the biomolecules so that they
can be kept in a stable and native
form and stays fully active. The buffer
itself, however, should not exert any
influence on the compound. Equally
important, the buffer should not
contain any impurities that may have
a negative effect on the activity or
stability of the molecule. Hence, only
highly-purified substances of defined
specifications should be used to ensure
reproducible processes on a lab or
production scale.
Merck supplies buffers in various
grades. The differences between them
rarely involve purity itself but more
the additional parameters that are
checked prior to delivery. For
example, whilst the nucleases may be
an important quality criterion for
biotechnological applications, the
fulfilling of the requirements of a
particular pharmacopoeia may well be
critical when it comes to manufacturing
a pharmaceutical active ingredient.
However, for certain applications,
it may be desirable to specify
additional quality parameters not
included in our “Certificate of
Analysis”. Should you require any
special testing, we will be able to offer
you a customized product in most
cases. The same applies to individual
package sizes. We will be glad to
prepare such a special service for your
convenience. Please contact us.

Benzonase ® endonuclease –
the Smart Solution for DNA Removal
Benzonase® endonuclease is a
genetically engineered endonuclease.
With its ability to rapidly hydrolyze
all types of nucleic acids and its extremely
high purity, it is the ideal
choice for a wide range of applications
in biochemical research and biotechnology.
Benzonase® endonuclease brings
benefits to the industrial-scale
manufacturing of recombinant
proteins and other biologicals,
irrespective of the type of production
organism or cell-line.
Benzonase® endonuclease...
... attacks and degrades all forms of
DNA and RNA.
... is free of detectable proteolytic
activity.
... is effective over a wide range of
operating conditions.
... possesses an exceptionally high
specific activity.
... is manufactured under strictly
regulated conditions in order to meet
industrial requirements for a reliable
supply and constant high quality.
... is ideal for a variety of applications
including:
- purification of proteins and other
biologicals.
- reduction of viscosity caused by
nucleic acids.
- sample preparations in electrophoresis
and chromatography.
FDA guidelines for the manufacture of
recombinant biologicals for therapeutic
use demand that nucleic acid contamination
should be limited to 10 pg
per dose in the end product.
Benzonase® endonuclease, when used
under appropriate reaction conditions,
will degrade all nucleic acid sequences
down to oligonucleotides of approximately
3 to 5 base pairs in length –
which is significantly below the hybridisation
limit – enabling recombinant
proteins to meet the FDA
guidelines for nucleic acid contamination.
The ability of Benzonase® endonuclease
to rapidly hydrolyze nucleic acids
makes the enzyme an ideal choice for
reducing cell lysate viscosity – both in
the research laboratory and the manufacturing
plant.
Using Benzonase® endonuclease to
reduce viscosity results in a number
of benefits including:
· Reduced processing time
· Increased yield of protein products
· Improved separation of pellet and
supernatant in centrifugations
· Easier filtration of solutions,
especially ultrafiltration
· Increased efficiency of chromatographic
purification steps
(e.g. in expanded bed adsorption).
Benzonase® endonuclease may be
used with all methods of cell lysis, including
lysozyme treatment freezethawing
procedures and high-pressure
homogenization.

Biochromatography
Downstream processing of proteins
and other biomolecules represents a
key role in the production of therapeutic
substances. The major requirement
is to maintain the biological activity of
these biomolecules during purification,
i.e. maintaining the intact native
structure through-out the separation
process. Therefore, to avoid protein
denaturation, most biochromatography
unit operations employ aqueous
buffer systems. After the complete
purification process has been developed
on a small scale in the lab, the
protocols, transferred to large-scale
production, must give the same results.
Normally, the large-scale production
of bio-molecules is a multi-step process
where column chromatography
plays a crucial role. The overall separation
and purification sequence for a
protein typically involves 5 -10 unit
operations. Each step must be operated
with the highest possible efficiency in
order to optimize product yield, ensure
feed stream
centrifugation, filtration
capture
high throughput; reduction of volume
intermediate purification
higher resolution; removal of host cell proteins
final polishing
high resolution; removal of aggregates and DNA
process reproducibility and minimize
process economics. The purification
strategy is to determine experimentally
the shortest route from the starting
material to the final product that meets
the required specifications for contaminants
and impurities. A schematic
overview is given in Figure 1.
Fractogel® EMD media enable the user
to meet all relevant requirements.
Capture steps require high flow rates
with no loss of capacity. All Fractogel®
EMD media provide excellent results
for ion exchange, hydrophobic interaction,
affinity and size exclusion
chromatography. High capacities, high
recoveries and high chemical and
mechanical stability of Fractogel® EMD
provide the downstream specialist a
powerful tool for protein separations.
At least five times the number of cycles
can be run on Fractogel® EMD resins
using the same column packing as
with soft gels based on carbohydrate
matrices.
Figure 1:
Flow chart of
downstream
processing

New Robust Tentacle Media
Tentacle surface technology, based on the grafting
of functionally substituted acrylamides to
the surface of porous particles, has now been
transferred to a novel hydrophilic vinyl polymer
resin. The resulting Fractoprep® ion exchange
media show excellent protein binding capacities
at high flow rates, with much improved mechanical
and chemical stability. The resins are extremely
resistant to sodium hydroxide treatment,
the most important clean-in-place step in almost
every production process and, in addition, pro-
Ion Exchange Chromatography on Fractogel® EMD Tentacle Resins
In ion exchange chromatography, charged molecules
are separated on column materials that
carry an opposite charge. The ionic groups of ion
exchange resins are covalently bound to the gel
matrix and are partially neutralized by counter
ions present in the buffer. Because proteins are
multivalent anions or cations, at low salt concentrations
they will bind to stationary phases
that carry the opposite charge. The binding is
strongly influenced by the pH of the buffer. For
efficient use of ion exchange chromatography, it
is important to operate at pH values where the
exchangers are mostly ionised and the biopolymers
have a net charge (Table 1). Excessively
high salt concentrations, however, cause
shielding of the charges on the protein surface
and effective binding of protein to an exchanger
can no longer take place. Once bound to the
matrix, biomolecules are in turn displaced by an
increasing salt gradient or pH changes. Proteins
with different net charges can be separated.
Desorption of proteins from the column begins
only with increasing salt concentrations or pH
changes, when the protein no longer binds. The
compounds that have a higher charge density are
bound more firmly to the column while the
others elute more rapidly. In contrast to strong
ion exchangers, which can be used over a wide
pH range, weak ion exchangers have changing
degrees of ionization depending on the pH. Thus,
the pH range over which you can use the ion
exchangers is limited by these properties., e.g.
Fractogel® EMD COO - can be used above pH 5,
Fractogel® EMD DEAE below pH 8.5. The weak
vide other features essential for application in
bio-pharmaceutical production. Due to their
excellent mechanical rigidity, Fractoprep® media
are easy to handle and can be packed in large
production columns without any problems. For a
complete list of references citing tentacle media
products, please visit our web-page. This list of
references includes papers using all the different
types of Fractogel® EMD and Fractoprep® media.
ion exchangers are used, in general, in situations
where the pH of the load is consistent and doesn't
change from run to run. Strong ion exchangers
that are charged regardless of pH are generally
used in production were the feed streams
sometimes change.
Recently, a new generation of Fractoprep®
tentacle media was launched. Fractoprep® beads
have a particle size range between 30 and 150 µm
and were designed for capture steps. The new
Fractoprep® ion exchangers provide high productivity
especially in the capture step. Large
pores enable a free diffusion of proteins into the
beads and allow high throughput at very low
back pressure. Three different types of ion
exchangers, Fractoprep® DEAE, Fractoprep®
TMAE and Fractoprep® SO 3 - , extend the product
range of the well-known Fractogel® tentacle
resins.
matrix
protein
pore structure
Advantages of the Tentacle Ion Exchangers
With most ion exchange gels the functional
groups are fixed directly, or with
short linkers, onto the surface of the
matrix. On any given gel matrix, only
a certain number of ionic groups can
be attached because of the chemistries
used and the limited inner surface of
the matrix. Once attached, the functional
groups must be accessible in
order for biopolymers to bind. The
spatial requirements for this binding
dictates the limit of the protein binding
capacity for these kinds of ion exchangers.
With tentacle ion exchangers,
however, the functional groups are
covalently anchored to the matrix via
flexible linear polymer chains. Therefore,
the binding capacity for proteins
functional groups
Figure 2:
left – conventional
ion exchanger
right – tentacle ion
exchanger
can be increased significantly. The
tentacles allow biopolymers greater
accessibility to the functional groups
due to less steric hindrance (Figure 2).
Especially for large proteins, tentacle
media provide higher binding capacities
compared to conventional
supports. Due to the good selectivity
and high efficiency, columns packed
with tentacle media provide good
chromatographic results and highly
concentrated fractions can be obtained.
The mechanical stability of all tentacle
media also allows the use of high flow
rates without a significant loss of
capacity.
Fractogel® EMD media as well as
Fractoprep® media are pressure-stable
and can be easily regenerated with
sodium hydroxide.
The main characteristics are:
· High dynamic protein binding
capacity
· Chemical stability (acids, alkali)
· Mechanical stability (pressure stable)
· Large pore diameter ( > 800 Å )
·Resistance to micro-organisms
· Reduced non-specific interactions
· High selectivity
· Availability at constant quality

Fractogel® EMD Tentacle Phases for Hydrophobic Interaction Chromatography
High performance hydrophobic interaction chromatography
is a powerful technique for the isolation
of proteins on an analytical and preparative
scale. The method of hydrophobic interaction
chromatography (HIC) is based on the interaction
between hydrophobic ligands on the chromatographic
support and hydrophobic areas located on
the surface of proteins. The hydrophobic feature
of a given protein is caused by certain amino acid
residues like isoleucine, valine, leucine, phenylalanine
and others.
Although most of the hydrophobic amino acid
residues are located in the center of the molecule,
recent structure analyses have shown
hydrophobic patches on the protein’s surface.
These areas are often correlated with the biological
function of the protein. The hydrophobic
character of a protein is promoted by high salt
and increased temperatures.
Advantages of HIC tentacle materials
The high selectivity of Fractogel® EMD Propyl (S)
Metal Chelate Affinity Chromatography on Fractogel® EMD Chelate
Advantages of tentacle media for metal chelate
affinity chromatography
The high efficiency of the method is based on the
interaction of a covalently bound chelating ligand
immobilized on a chromatographic support with
proteins containing a large number of histidine
residues. Some recombinant proteins have His
tags engineered at the end of the protein sequence
to facilitate purification on IMAC resins. Iminodiacetic
acid has been chosen as the metal-chelating
ligand for Fractogel® EMD Chelate. The
iminodiacetic acid residue is suitable as a chelating
compound because a bidentate chelating
moiety remains free after immobilization, onto
which a metal ion can be co-ordinated. Various
metal ions can be immobilized on the stationary
phase (e.g. Cu 2+ ; Ni 2+ ; Co 2+ ; Zn 2+ ). Free coordination
sites of the metal ions are used to bind
different proteins and peptides.
Compared to conventional affinity gels synthesised
with spacer molecules, Fractogel® EMD
and Fractogel® EMD Phenyl (S) allow efficient
purification steps. Both phases, with a particle
size of 20-40 µm, are mechanically stable and
resistant to biological and chemical degradation.
The functional groups are on linear polymer
chains. The ligands differ with respect to their
hydrophobicity. Ligand density of the functional
groups bound to the matrix is optimised by the
tentacle technology so that excessive hydrophobicity
is avoided. Strong hydrophobic resins
inhibit recovery and resolution.
Application areas
Both hydrophobic phases are suitable for a wide
range of applications. The main application areas
for HIC is the purification of cytosolic proteins,
antibodies, and recombinant proteins. Membrane
proteins can also be isolated on Fractogel® EMD
Propyl (due to its weak hydrophobicity this resin
is more suitable). For the elution of strong
hydrophobic proteins, a detergent should be
used. In addition HIC is suitable for the removal
or exchange of non-ionic detergents.
Chelate allows the binding of larger amounts of
proteins. Because the immobilized metal ion can
be arranged in the best position relative to the
binding sites on the protein’s surface, proteins
can be bound more tightly to Fractogel® EMD
Chelate than comparable conventional gels.
Application areas
The existence of surface-accessible histidine
residues is important for the binding of a native
protein. As recombinant proteins become more
important, expression systems using a polyhistidine
tag for the protein enable rapid isolation
of those proteins by metal chelating affinity
chromatography. With peptides, the alpha amino
group also plays a role, so that peptides can be
retained even if no histidine residues are present.
In the case of peptides, other metal binding amino
acids like cysteine and tryptophan contribute to
the binding.

Thiophilic Adsorption Chromatography on Fractogel® EMD TA
Advantages of tentacle-mediated
thiophilic adsorption
The binding mechanism of thiophilic adsorption
chromatography is based on the specific interaction
of a covalently bound sulphur-containing
ligand immobilized on a chromatographic support
with proteins or peptides. An electron
donor-acceptor mechanism between the affinity
ligand and tryptophan residues on the surface of
the target molecule is responsible for the selective
binding. Fractogel® EMD TA is designed specifically
for the efficient isolation of antibodies. The
synthesis is performed according to the tentacle
technology where the group-specific ligands are
present in high density. Thus, the thiophilic tentacle
material has a high protein-binding capacity
and is suitable for antibody purification on
an analytical as well as a preparative scale. Due to
the steric accessibility of the ligand, the resin provides
minimized non-specific interactions with
proteins combined with high protein binding capacities.
The chemistry of the 3-S type ligand that
is used for Fractogel® EMD TA allows significantly
better thiophilic binding compared to the 2-S
types which have more hydrophobic interaction.
Application areas
Thiophilic adsorption chromatography is specifically
designed for the purification of immunoglobulins
(monoclonal and polyclonal antibodies)
including single-chain antibodies and chicken
antibodies. Albumins are not adsorbed on thiophilic
media, which often simplifies the effective
separation of antibodies. The binding of the protein
takes place mainly via accessible tryptophan
and/or phenylalanine residues. Corresponding
motifs can be observed within conserved regions
of various antibodies. Antibodies of the IgM subclass
can also bind to a Fractogel® EMD TA
column. The binding of antibodies from different
species to thiophilic adsorption supports offers
advantages over protein A. Elution conditions at
physiological pH values enable high recoveries of
biologically active antibodies.
Molecular Size Exclusion Chromatography on Fractogel® EMD BioSEC (S)
This method, also known as gel filtration
(GPC) or molecular sieve chromatography
is based on the different size
and shape of proteins. Proteins of
different sizes penetrate into the internal
pores of the beads to different
degrees. Small protein molecules are
retarded by the column while large
molecules pass through more rapidly.
A column packed with Fractogel® EMD
BioSEC can be used for the separation
of proteins in the range between 5 kDa
and 1,000 kDa (Figure 3). The mechanical
stability of the resin allows higher
flow rates compared to conventional
soft gels. This is very helpful during
regeneration and equilibration of the
column. The pressure stability also enables
the packing of larger columns
that are needed for polishing steps in
the production of bio-molecules.
Figure 3: Separation of a complex mixture on
Fractogel® EMD BioSEC (S) with a column
dimension of 1000 x 50 mm. The sample
contained ferritin (1), immunoglobulin G (2),
transferrin (3), ovalbumin (4), myoglobin (5),
aprotinin (6) and vitamin B12 (7). 5 ml of the
mixture were injected onto the column at a
flow rate of 3 ml/min (eluent: 20 mM sodium
phosphate buffer, 0.1 M NaCl pH 7.2).
Application areas
SEC is very easy to perform up through
production scale and the results are
very reproducible. SEC is generally
performed late in the process when the
number of protein contaminants are
small, as in a “polishing” step. The
goal is the final removal of possible
impurities such as structurally similar
or closely related forms of the product
(aggregates, deamidated or oxidised
product, etc.), host proteins, reagents
and buffer substances, leachables from
chromatographic supports, endotoxins,
nucleic acids (DNA, RNA) or
viruses. However, if a target protein is
extremely large or small, SEC can be
utilized earlier in the purification process.
Principal applications of SEC on Fractogel®
EMD BioSEC are purification of
cytosolic proteins as well as recombinant
proteins and, simultaneously, the
determination of their molecular mass
after the column has been calibrated.
Membrane proteins can also be isolated
by this technique and SEC has been
used in blood plasma fractionation.
With the introduction of recombinant
proteins there has been an emphasis
on the determination of aggregates
and other by-products that co-purify
with the target protein. Size exclusion
chromatography is also very useful for
the purification of monoclonal and
polyclonal antibodies. Albumins with
a molecular weight of about 60 kDa
elute earlier than IgG or IgM molecules.
Since no gradient elution has to be
applied, a programmable gradient
mixing system is not necessary and
comparatively simple equipment is
needed for operation. In addition, the
method can be easily integrated into
purification schemes and most of the
operational steps can be automated.

Process and Production Chromatography
Standardized Silica Gels
The basis for reproducible chromatography
in laboratory and production
Preparative column chromatography
is a separation and purification method
that plays an important role in purifying
valuable compounds in research,
pilot plant operation and production.
This method can be used not only to
purify substances to a very high degree,
but to do this in a rapid and
economical way.
In practice, standardized sorbents are
required. These provide a high degree
of method reliability, direct transfer
from analytical scale and optimised
throughput per unit of time. Only silica
gels with a defined pore structure fulfill
the requirements of process chromatography.
Merck has specialized over decades in
the manufacture of standardised silica
gel packing materials for preparative
chromatography and offers a wide
range of customized sorbents. The product
range is completed with several
special types, e.g. silica gels of the
type LiChroprep® or highly purified
acid-washed silica gels.
Such gels provide the user with unique
method reliability. Regardless of which
chromatography process is used
(HPTLC, HPLC, preparative chromatography),
direct and rapid transfer to a
production scale is guaranteed and
without risk. This is ensured by the
fact that identical raw materials are
used for all stationary phases, thus
providing identical chromatographic
selectivity. In order to be able to
achieve this goal, Merck produces all
silica gel 60 packing materials for
analysis, pilot plant processes and
production using the same silica gel
raw material of specified surface area
and a defined specific pore volume.
Once a particular silica gel raw material
has been tested with respect to its suitability
for pore structure, specific pore
volume, specific surface, chemical
purity and other requirements for chromatography,
it is classified into various
categories and given defined particle
size ranges.
These particle size ranges are selected
in such a way that an optimum in separating
performance and permeability
results for the chromatographic
process used (DC, HPTLC, HPLC, preparative
chromatography).
Thus, once a successful configuration
has been found on an analytical scale,
it can be transferred rapidly to processes
on a pilot plant or production
scale. Merck guarantees direct scaleup
without compromise.
The selectivity of Merck’s silica gels
for analytical and preparative applications
is identical. Resolution, however,
decreases, according to theory,
with increasing particle size. Improved
separation can be achieved at the
expense of longer retention times and
if the selectivity of the entire system is
optimized using another mobile phase.
Practice has shown that for chromatographic
separations in production,
silica gel 60 types i.e. silica gels of a
mean pore size of 60 Å and in particle
size ranges 40–63 µm and 63–200 µm
are excellent for the separation of
complex mixtures.

LiChroprep®
The Guarantee for Selectivity, Reproducibility and Quality
Selectivity is one of the most important
factors in chromatography. It
is decisive in determining the quality
and the reproducibility of a chromatographic
separation and, in addition,
on a preparative scale, it also
determines, to a large extent, the
yield.
In order to be able to guarantee the
batch-to-batch reproducibility over a
period of years, it is not only sufficient
to carry out a simple performance test
using aromatic substances. We also
provide comprehensive selectivity
LiChrospher® for preparative highperformance
chromatography is a
spherical and porous silica gel
characterized by:
· Uniform and homogeneous silica gel
matrix from bach-to-batch.
· Very narrow particle size
distribution for high performance
and high packing stability.
· High permeability at low pressure
drop in the column.
· Reproducible specific surface area
and pore size distribution.
documentation and carry out an
additional selectivity test routinely for
each batch release.
This selectivity test, which is carried
out using special solutes under exactly
defined practice-oriented chromatographic
conditions, serves to determine
the smallest change in the selectivity
properties of the sorbent.
Merck can therefore guarantee that
LiChroprep® packing materials will
produce reproducible results within
very narrow band ranges. This provides
the highest reliability for chro-
LiChrospher® Packing Materials – Sorbents for High-Performance
Preparative Chromatography
· Different selectivities (Si 60, RP-18,
RP-select B and WP 300 RP-18)
ensure a simple and rapid optimisation
of the chromatography system
with respect to selectivity and sample
composition.
· Two different particle size ranges
(12µm and 15µm) ensure the
necessary chromatographic performance
to be selected for the desired
application and thus optimised
throughput for the chosen selectivity.
Non-derivatized silica gels still play
an important role in preparative
chroma-tography. For routine
applications, it is important that
spherical silica gels are produced
according to the same stringent guidelines
as irregular Merck silica gels.

matographic separations in
the laboratory, the pilot
plant and in full scale
production.
The advantages:
· Identical selectivity of
analytical and preparative
material means scale-up
without compromise.
· Identical batch-to-batch
selectivity is the guaranteed
assurance for 100%
reproducibility – over a
period of years.
This means that only those
spherical silica gels are released
for sale that meet
all required specifications
with respect to pore structure,
specific pore volume,
specific surface area and
chemical purity as well as
complying with additional
important chromatographic
parameters.
Independent of the chromatography
process used to
develop a particular method,
the direct and rapid transfer
from lab to production scale
is guaranteed. Identical raw
materials used in spherical
silica gel manufacturing
ensures direct scale-up.
Other Sorbents:
Cellulose · Magnesium Silicates · Silica
Standardized Silica Gels – Surface-Modified and Enantioselective Carriers
Microcrystalline Cellulose
This is a hydrophilic polysaccharide
packing material preferred for the raw
separation of amino acids and related
compounds. Cellulose is also used for
the gentle purification of biomolecules.
Magnesium Silicates – Florisil®
Florisil® is a polar highly selective
magnesium silicate of the approximate
composition MgO/SiO 2 (15/85) which
is particularly suitable for the separation
of steroids, alkaloids, antibiotics
etc. This stationary phase is also
used for the sample preparation of
environmental samples such as pesticide
residue analysis, chlorinated
Reversed phase material for production scale
Because of the organic nature of
cellulose, it can only be used in a preswollen
state and under gentle or
hydrostatic pressure.
hydrocarbons and pesticides. For
sample preparation in the case of
pesticides, a specially purified and
activated Florisil® is often used (Cat.-
No. 1.12994). The normal activation
temperature for Florisil® is 650°C,
whilst activation at 260°C produces a
less active material.
· An alternative material to non-modified silica gel because non-toxic solvents
in the RP-mode are used.
· For the economic purification of products where conventional RP-phases
are too expensive.
· High loading capacity due to the high specific surface area of the base silica gel
(60 Å pore diameter, approx. 500 m 2 /g); therefore column dimensions/amount of
sorbent can be reduced or the sample volume increased.
· High pressure stability under extreme production conditions
for long-term column usage.
PharmPrep®
PharmPrep® media are a new line
of spherical silica sorbents designed
for process chromatography in the
pharmaceutical market.
The first product to be made available,
PharmPrep® CC for preparative
chromatography, is a spherical, totally
porous Silica Gel manufactured by a
large-scale process from water glass
and sulphuric acid to produce a large
particle spherical product.
The spherical materials have the
follwing advantages for classical open
column chromatography:
Aluminium Oxide for Preparative Chromatography
Aluminium oxide is one of the most
versatile sorbents for preparative
chromatography. Due to its weaker
adsorption than silica gel, standard
aluminium oxide can be used in
specifically defined pH-ranges.
The aluminium oxide matrix
comprises octahedral and tetrahedral
aluminium atoms coupled with
oxygen, whereby the surface
aluminium atoms are saturated with
free hydroxyl groups. The different
acidic centres (Broenstedt acid, Lewis
acid and Lewis basic groups) result in
the fact that, in contact to ionic salt
solutions, aluminium oxides possess
both anion and cation exchange
properties. Its better pH-stability,
especially in the alkaline range, in
comparison with silica gels is worth
mentioning.
They are easily packed into columns;
they have high bulk densities, which
result in good loadability; and they
exhibit low back pressures, allowing
for fast elutions.
PharmPrep® CC can be used in conjunction
with spherical sorbents for
performing process chromatography
at any scale and at a reasonable cost.
PharmPrep® CC is the ideal media for
high-quality, high-performance separations
at a true process scale.
Aluminium oxide occurs in various
crystal forms. For chromatography, the
γ-structure in the form of aluminium
oxide 90 with a mean pore diameter of
90 Å is mostly used. Further structural
types are aluminium oxide 60 (ε-type)
of 60 Å pore diameter and aluminium
oxide 150 (α-type) of 150 Å pore
diameter, each of which possesses
specific adsorption properties.
Standardized aluminium oxide 90 for
chromatographic adsorption analysis
according to Brockmann is a sorbent
of medium polarity and it is frequently
used when the cationic exchange
properties of the basic aluminium
oxide are required. In addition, it is an
alternative material to activated charcoal
where this product cannot be
used because of its organic character.

Customized Preparative Columns (25 and 50 mm, i.d.) of Higher Quality
We offer a set of Hibar® pre-packed, customized
stainless steel columns (20 and 50 mm, i.d.) for
high resolution lab-scale purification.
For your laboratory work, it is not enough just to
have a top-quality preparative column; you also
need a method that is and will remain, reliable
for years.
Customer benefits at a glance:
Column Packing Stands NW25, NW50 and NW100: Innovative Column Packing Technology
Conventional packing methods for preparative
columns use rapid filtration under pressure and
at high linear flow rates. This technology often
leads to bridging effects between the particles in
the stationary phase that have a negative impact
on the column performance. Our studies have
shown that this undesirable side-effect can be
overcome by employing a pulling instead of a
pushing mechanism.
The new column packing technology is based on
a combination of vacuum suction with subsequent
column bed compression; this stabilizes
the packing. A common water-jet pump is used
to generate the vacuum. The gel slurry is sucked
through a packing reservoir into the column by
applying low pressure at the bottom end of the
column.
This method allows precise adjustment of the desired
packing volume or gel bed height. In the
production environment, the new technology
You also need to be able to transfer it to other
laboratories throughout your parent or subsidary
companies in the world.
Our own long-term experience has shown how
important chromatographic procedures are and
we would like to pass on the benefits of this experience
to you in the form of a column with
higher quality and reproducibility.
Quality: Hibar®-customized pre-packed columns are manufactured to strictly
controlled conditions to ensure both excellent packing quality and
reproducibility of a separation.
Economic: The fully controlled gel bed compression of the 50mm (i.d.) column leads to a
longer lifespan of the columns and improves process economy. In addition,
with the hydraulic stand accessory, it can be upgraded to Packing Stand NW50.
Versatile: You can choose your packing materials (Merck or those of other manufacturers)
for the Hibar®-column (25 or 50mm, id) from an extensive range of
sorbents.
Simple: Columns are ready-to-use and can be easily connected with any HPLC system.
Problem-free: Methods developed on a particular sorbent on an analytical scale can be
directly scaled-up to preparative scale.
facilitates the determination of the column geometry
and thereby makes direct scale-up from
analytical to preparative scale easy.
The new column packing devices are available
for columns of 25 mm, 50 mm and 100 mm inner
diameters, covering the full range from semi-preparative
to pilot-scale and production scale.
Packing and Unpacking the Packing Stands
The required volume of gel slurry is prepared
according to the physical parameters of the
stationary phase and the column size. Using a
funnel, the suspended material is filled into the
packing tube attached to the column. Vacuum is
applied. After the suspension has been sucked
into the column tube, the packing tube is
removed and the upper column cap lid (flange) is
sealed. The column bed is then washed with
3-5 column volumes of a suitable
solvent and simultaneously
compressed over several
steps to a bed compression that
depends on the sorbent type
being used (see operating
instructions). In this way, rapid
Customer benefits at a glance:
· The new vacuum packing technology permits simple and flexible handling
of preparative columns.
· The separation columns can be prepared rapidly on-site.
sedimentation and homogeneous packing of the
column is achieved. The column is ready for
operation.
To unpack the column, the upper flange lid is
removed and the material is forced out by
moving the piston upwards with the hydraulic
pump. The column is now ready for the next
filling procedure. The stationary phase may be
kept for later use or disposed of.
· The fully controlled gel bed compression leads to a longer lifespan of the columns
and improved process economy.
· Even soft gels such as polymer-based gels can be packed with the new packing devices.
· One single packing stand can be used to pack an unlimited number of columns
of the same inner diameter.
· You can choose between an economic upper screw cap and the convenient
chain-closing system for the 50 and 100mm ID packing stands.
· Customized column lengths can be provided on request.
· Columns with temperature control jackets provide optimum separation
stability and performance (optional).
Chromolith® Prep Si
Increase in speed, efficiency and productivity
Chromolith® Prep monolithic stationary phases are
new ultra-pure silica phases that have special
properties resulting from their bimodal pore
structure. This structure is based on new “Sol-
Gel” technology and consists of macropores and
mesopores.
The mesopores, with an average diameter of
12 nm, form the fine porous structure of the
column interior and create a very large surface
area on which adsorption of the target compounds
occurs.
The large macropores, with a pore diameter
ranging from 3 µm, form a dense network of
pores and allow their high flow rate due to their
low resistance factor. The resulting excellent
accessibility of the mesopores (total porosity
kinetics because of the short diffusion length
inside the pores. This results in dramatically
reduced separation times and thus in a huge
increase in productivity.
To operate a monolithic column it is necessary
to ensure that no flow-through occurs in the
wall region. Therefore, the monolith is cladded
with a polymeric material (special PEEK) and can
be connected directly to any HPLC system and
used as a ready-to-use-column.
Contract Separation at Merck
Until recently in the pharmaceutical industry, the
splitting up of project work was only possible if
kept within the company. However, increasing
costs and the fact that development times for
new products are extending over 10 to 15 years
has forced companies into new, innovative
thinking. The principle of “do everything yourself”
is no longer meaningful in many areas, e.g.
in the case of chromatographic separations: the
considerable investment necessary for HPLC
equipment and the need for dedicated buildings
is a substantial risk bearing in mind that the
products in question still have to undergo phase
I and III clinical trials - and may not even be
approved for marketing at the end of the day.
Hence, “out-sourcing” – the sharing of work
with an external partner – is a meaningful
strategy pursued successfully by many companies
today in meeting this challenge. Chromatographic
separation problems in particular
can be ideally solved in this way if a suitable
out-sourcing partner can be found. The risks can
be substantially reduced and the costs kept to an
acceptable and controllable level.
Merck can provide a complete solution from one
source: apart from our comprehensive range of
chromatography products, we also have a contract
separation service available for our customers.
We provide know-how, experience, competence
and “capacity“ in two ways:
Merck is one of the largest production and sales
organizations for chromatography equipment
and sorbents and has accumulated extensive
know-how over many years in all areas of chromatography
– from analytical to preparative and
to production scale. Our highly qualified
specialists on the one hand and the stringent
maintenance of relevant standards (DIN ISO
9001, FDA, GMP, GLP etc.) on the other enable
us to tackle and solve any chromatographic
problem and to develop economic processes for
the production of amounts ranging from
milligrams to tons.
Each and every employee involved with your
problem is a “capacity” in his specialized area.
And, due to our sheer size and infrastructure, we
Enzymes for Processing
Below you will find a selection of
Merck enzymes for biochemistry
mainly used in downstream processes,
biochemical research and food
analysis.
The product description of each enzyme
gives information on the origin
of the product, enzyme activity and the
numerical classification (EC number).
Definitions of enzyme activity are
often historical in nature and relatively
arbitrarily selected. As long as there
are no standard procedures and very
few internationally recognized ones,
one has to be most careful when comparing
enzyme qualities. Initially, it
β-Glucoronidase
This preparation is used for the gentle hydrolysis of
steroid conjugates, e.g. in the assay of urinary steroids
and mucopolysaccharides.
β-Glucoronidase/Arylsulfatase (from Helix pomatia)
stabilized aqueous solution for biochemistry,
EC 3.2.1.31 + EC 3.1.6.1. (Merck Cat.-No. 1.04114)
Lysozyme
Lysozyme is a mucolytic enzyme. It is used for
bacteriolysis.
Lysozyme catalyzes the hydrolysis of β-(1-4) bonds
between N-acetylmuramic acid and 2-acetamido-2deoxy-D-glucose
residues in mucopolysaccharides,
mucopeptides and chitin.
Lysozyme also gently breaks up bacterial cell walls, e.g.
of micrococcus luteus or micrococcus lysodeicticus; in
the presence of EDTA e.g. also of bacillus subtilis and
escherichia coli.
Lysozyme (from egg white) 100,000 U/mg cryst.
(hydrochloride) for biochemistry, EC 3.2.1.17
(Merck Cat.-No. 1.05281)
Pepsin
Pepsin is any one of the small family of closely related
aspartic proteinase enzymes.
Pepsin is used for the hydrolysis of peptides and
proteins, e.g. insulin.
Preferential cleavage sites are Phe- and Leu-.
Pepsin only attacks the peptide bonds; esters and
amides are not hydrolyzed.
Furthermore, the enzyme can be used for the pepsincatalyzed
synthesis of oligopeptides.
Pepsin (from porcine gastric mucosa) 0.7 FIP-U/mg for
biochemistry, EC 3.4.23.1 (Merck Cat.-No. 1.07185)
Pepsin (from porcine gastric mucosa) cryst. lyophilized
10 FIP-U/mg for biochemistry, EC 3.4.23.1
(Merck Cat.-No. 1.07192)
should be checked whether enzymes
of identical origin are involved (e.g.
plant cells, animal origin). Preparations
from different manufacturers
can be only compared if their activities
have been determined under
identical conditions. A few degrees
of reaction temperature, different
buffer systems, pH, different enzyme
substrates or different reaction times
make exact comparison practically
impossible. Differently determined
units can rarely be converted to each
other. If a comparison is necessary, the
only way to do so is by experiment.
Pronase E
Pronase E may be used for the gentle hydrolysis of
proteins or polypetides.
Compared with acid total hydrolysis, the enzymatic
splitting amounts to approximately 70 -90 %, due to
the very low substrate specificity of pronase E.
The enzyme is also used for the isolation of amino-acidglycoside
components from mucoproteins.
Pronase E (from streptomyces griseus) 4,000,000 PU/g
for biochemistry, EC 3.4.24.4 (Merck Cat.-No. 1.07433)
Trypsin
Trypsin is a serine endopeptidase which catalyzes the
hydrolytic cleavage of proteins and peptides specifically
at the carboxyl groups of basic amino acids L-Arginine
and L-Lysine.
Furthermore, esters and amide bonds of synthetic
derivatives of these amino acids, e.g. N α-benzoyl-Larginine
ethyl ester, are also hydrolyzed.
Trypsin (from porcine pancreas), cryst. lyophilized
40 U/mg for biochemistry, EC 3.4.21.4
(Merck Cat.-No. 1.08350)
Trypsin (from porcine pancreas), TPCK-treated,
lyophilized 40 U/mg for biochemistry, EC 3.4.21.4
(Merck Cat.-No. 1.24590)
Further enzymes are included in the
Safety in the Laboratory
Are you in charge of a laboratory?
If so, you are also responsible for the safety and
well-being of all persons working there as well as
for the environment.
Your duties include being aware of what might
happen, establishing what must not happen, and
implementing measures aimed at avoiding
accidents.
· When planning an experiment, decide whether
hazardous substances are in fact required or
whether a less hazardous substance might not be
used instead. (To help you, we have published
information in a Chemical Data Base under
www.chemdat.de).
· In addition to the risks that the substances
themselves pose, you must establish if there are
any risks specific to your factory or plant. Find out
whether any hazardous chemical reactions could
conceivably occur and decide how these can be
avoided.
· Specify all measures needed in order to assure
personal health and safety, according to the
ranking technical – organizational – personal, and
check these are in place before any hazardous
substances are handled.
· Ensure that hazardous substances are shelved and
stored in such a way that they can pose no threat
to human health and the environment. Restrict the
amounts of combustible substances used in the
laboratory to the absolute minimum needed.
· Issue workers with suitable, effective personal
protective clothing and equipment. Ensure that it
is kept clean and in good condition.
· Compose a set of laboratory rules that cover all of
the potential health and environmental risks that
may be encountered in the laboratory, and also
specify general preventive measures and rules of
conduct.
· Compile specific operating procedures for activities
that involve the use of hazardous substances (see
SafeDAT software (item no. 403032)). Display these
operating procedures prominently within the laboratory,
and use them to instruct workers at least
once a year in the handling of hazardous substances
and in the potential dangers of working with
them.
· Always strive to improve industrial safety and
personal well-being in the area for which you are
responsible.
To find out more on how you should handle our
reagents, please speak to your contact person
within your local Merck dealership. This person
can then, if necessary, contact our Industrial
Safety Department for further information.
Do you work in a laboratory?
When handling chemicals, always follow safety
procedures, even if the chemical in question is not
labeled as hazardous.
· Make sure the laboratory is kept clean and tidy.
· Use only such hazardous substances and
equipment as are absolutely necessary for you to
carry out your duties.
· Always ensure that the sash front is down on any
fume hood.
· If possible, keep hazardous substances in their
original Merck containers and packaging. These are
able to withstand normal conditions of use and are
suitably labeled.
· If you need to use a different container for day-today
use, ensure it is at least labeled with the name
of the chemical, its hazard symbol(s) and clear text
warnings. Keep containers with hazardous substances
no higher than the height at which they
can still be safely dispensed and shelved.
63

64 Appendix
· Where possible, keep very toxic and toxic
substances under lock and key.
·Keep any substance that emits harmful vapors or
gases in a place that allows these to be permanently
extracted.
· Check the laboratory inventory at least annually
and dispose of any chemicals that have become
unusable or no longer required.
· Never pipet by mouth. Always use a mechanically
operated pipet.
· Do not eat, drink or smoke in the labo-ratory.
· Never keep food or drink in the vicinity of
chemicals. Never place chemicals in containers
that are normally used for food or drink.
· In the event of a potentially harmful leakage of
gases, vapors or particles leave the laboratory
immediately and warn persons nearby. Before any
attempt is made to eliminate the danger, take
appropriate measures.
· Always work under a hood if you are using
spontaneously combustible substances, and keep a
suitable fire extinguisher close to hand.
· Use Perex-Test® (item no. 116206) to test for
peroxides in liquids that show a tendency to form
them, especially before distilling or evaporating
the liquid. Dispose of them properly.
·Keep liquids that show a tendency to form organic
peroxides away from light. This is no guarantee,
however, that peroxides will not form.
· Always use the personal protective clothing and
equipment that was issued to you. However, the
use of respirators and full-body protection should
be the exception rather than the rule.
·Wear suitable clothing and sturdy, closed, non-slip
footware when working in the laboratory.
· Always wear safety spectacles affording adequate
protection at the top and sides when working in
the laboratory.
· Always wear protective gloves when the hands in
particular may be at risk. Always check gloves
before using them, and replace any that are
damaged or otherwise unsuitable for use.

Risk and Safety Information (R and S phrases)
The properties of all Merck products are closely scrutinized. Items classed as
hazardous are marked as such in accordance with EU regulations.
These prescribe labelling with hazard symbols and plain-text warnings, and the
inclusion of risk and safety (R and S) information.
It must, however, be noted that, while the data reflect the state at their
generation, labels are continually updated to reflect the latest state of the art
and detail changes in regulations. Should data differ those given on the label
shall apply.
Attention: In the case of R phrases with the addition E,
the text starts with the word also.
