Die Realisierung des Zentrums <strong>für</strong> Molekulare Biowissenschaften<br />

DEVELOPMENT OF MOLECULAR BIOSCIENCES<br />
Die Entwicklung der <strong>molekulare</strong>n Biowissenschaften an der<br />
naturwissenschaftlichen Fakultät der Universität Graz begann<br />
1963 mit der Errichtung einer Lehrkanzel <strong>für</strong> Biochemie am<br />
Institut <strong>für</strong> Physikalische Chemie. 1969 wurde ein eigenes<br />
Institut <strong>für</strong> Biochemie an der Naturwissenschaftlichen Fakultät<br />
unter dem Vorstand E. Schauenstein gegründet. Die Biochemie<br />
galt lange Zeit als Hilfs- und Ergänzungsfach der Chemie, ein<br />
Faktum, das sowohl der räumlichen als auch der personellen<br />
Ausstattung im Vergleich zu anderen Instituten der Fakultät<br />
hinderlich war. Ähnlich bescheiden startete die Molekular -<br />
biologie/Mikrobiologie in Bezug auf ihre Organisationsgröße.<br />
Im Jahr 1983 errichtete die Universität das Institut <strong>für</strong> Mikro -<br />
biologie innerhalb des Fachbereichs Biologie. Vorstand des<br />
Instituts wurde G.Högenauer mit ursprünglich zwei Universi -<br />
tätsassistenten als akademische Mitarbeiter. Dieser im inter -<br />
nationalen Vergleich geradezu erschreckend minimalistischen<br />
Ausstattung der <strong>molekulare</strong>n Biowissenschaften an der<br />
Universität Graz standen schon damals sichtbare Forschungsund<br />
Lehrerfolge entgegen. Die große Anzahl der Studierenden<br />
an der Mikrobiologie machten einen eigenen Studienzweig<br />
<strong>für</strong> dieses Fach sinnvoll und H. Esterbauer (Vorstand des<br />
Instituts <strong>für</strong> Biochemie seit 1990) wurde zu einer international<br />
anerkannten Koryphäe der sich rasant entwickelnden Gebiete<br />
Lipidoxidation und Sauerstoffradikale. Seine drei am häufigsten<br />
zitierten wissenschaftlichen Arbeiten wurden laut ISI Citation<br />
Index 2007 insgesamt 5211 mal zitiert, welches ihren bahn -<br />
brechenden und nach wie vor hochaktuellen Charakter<br />
eindrucksvoll dokumentiert. H. Esterbauer initiierte das<br />
erste Groß<strong>for</strong>schungsprojekt im Bereich der Lipid <strong>for</strong>schung<br />
(SFB Biomembranen) in Graz, und sein Konzept des<br />
„Offenen Labors“ lebt über den Bereich der <strong>molekulare</strong>n<br />
Biowissenschaften hinaus weiter.<br />
The development of <strong>molecular</strong> biosciences at the Faculty of<br />
Natural Sciences at the University of Graz started in 1963,<br />
with the implementation of a chair <strong>for</strong> Biochemistry at the<br />
Department of Chemistry. In 1969, the Institute of Biochemistry<br />
was founded under E. Schauenstein. At the time, “Biochemistry”<br />
was still considered a satellite topic within the Chemistry<br />
c urricula. This lack of independent standing severely hampered<br />
both spatial and personnel developments in the beginning.<br />
Microbiology/Molecular Biology started in a similarly modest<br />
manner in terms of organizational size. In 1983, the Institute of<br />
Microbiology, headed by G.Högenauer, was constituted within<br />
the Biology division with a starting size of only two faculty<br />
members! This minimalistic configuration – in the international<br />
context – of the <strong>molecular</strong> life sciences at the University of Graz<br />
soon stood in sharp contrast to highly visible achievements<br />
both in research and teaching. Successful and popular teaching<br />
led to new curricula in microbiology. The research achievements<br />
of H. Esterbauer (head of the Department of Biochemistry from<br />
1990) made him an internationally well-known and respected<br />
scientist in the rapidly developing fields of lipid peroxidation and<br />
reactive oxygen species. His three most important articles<br />
(according to ISI Citation Index 2007) were cited more than<br />
5200 times, demonstrating the ground-breaking and highly<br />
relevant character of his research. H. Esterbauer initiated the<br />
first local network research program in the lipid field in Graz<br />
(SFB Biomembranes), and his concept of “laboratories without<br />
walls” still lives on in the <strong>molecular</strong> biosciences, and beyond.<br />

Angesichts der dynamischen Entwicklung der Molekularen<br />
Biowissenschaften in Graz und eines „role models“ in Wien,<br />
nämlich des Vienna Bio<strong>center</strong>, beschlossen die Professoren<br />
Esterbauer, Högenauer und Kostner (nunmehr Medizinische<br />
Universität) ab 1990 das Projekt eines gemeinsamen „Zentrums<br />
<strong>für</strong> Biochemie und Molekularbiologie“ nach internationalem<br />
Vorbild zu entwickeln. Damit sollte der extremen räumlichen<br />
Zersplitterung der <strong>molekulare</strong>n Biowissenschaften in Graz<br />
e ntgegengewirkt werden. Es wurde klar erkannt, dass nur ein<br />
umfassendes Kooperationsprojekt jene kritische Masse an<br />
wissenschaftlicher Kompetenz erzeugt, die <strong>für</strong> international<br />
sichtbare Forschungserfolge benötigt wird. Schon 1994 be <strong>für</strong> -<br />
wortet das Fakultätskollegium der Naturwissenschaftlichen<br />
Fakultät den Bau eines „Zentrums <strong>für</strong> Biochemie und<br />
Mole kularbiologie“ und der Akademische Senat der KFUG<br />
fasst einen Beschluss zur Errichtung dieses Zentrums.<br />
Nach einer Vielzahl von Evaluierungen durch internationale<br />
und nationale Gremien sowie die zuständigen Ministerien,<br />
einem fehlgeschlagenen Architektenwettbewerb und mehreren<br />
Umplanungen, gelingt es schließlich unter tatkräftiger Mithilfe<br />
des Rektorenteams, eine Übernahme der Mietkosten und<br />
damit eine Umsetzungszusage durch Frau Bundesministerin<br />
Gehrer zu erwirken.<br />
Damit waren aber noch nicht alle Schwierigkeiten beseitigt und<br />
finanziellen Hürden überwunden. Im Rahmen der Detailplanung<br />
tat sich noch ein Finanzierungsloch von ca. 30% der geplanten<br />
Gesamtkosten auf und es ist nur dem hochengagierten Einsatz<br />
von Rektor Gutschelhofer und Vizerektor Zettl zu verdanken,<br />
dass es gelungen ist, diese Lücke zu schließen. Zusammen<br />
mit den Sektionschefs des BMWF, Dr. S. Höllinger und<br />
Dr. F. Faulhammer, sowie der Führung der Bundesimmobilien -<br />
ge sellschaft konnte das Projekt ZMB schließlich durchfinanziert<br />
und umgesetzt werden.<br />

In view of the dynamic developments of <strong>molecular</strong> biosciences<br />

the Faculty of Natural Sciences and the Senate approved the<br />
erection of a Center <strong>for</strong> Biochemistry and Molecular Biology.<br />
A number of evaluations by national and international agencies<br />
and the Federal Ministry, a first architectural competition and<br />
numerous plan modifications followed. Finally, the tremendous<br />
support of the rector’s team and agreement <strong>for</strong> covering<br />
the rental costs resulted in the confirmation by minister<br />
Gehrer.<br />
Yet during the detailed planning stage, when a projected cost<br />
increase of 30% almost killed the project, the heroic ef<strong>for</strong>ts<br />
of Rector Gutschelhofer and Vice rector Zettl filled the gap<br />
in resources, ultimately saving the project. Together with<br />
Sektionschefs of the Federal Ministry <strong>for</strong> Science and Research,<br />
Dr. S. Höllinger and Dr. F. Faulhammer, as well as the director<br />
of the Bundesimmobiliengesellschaft, the project “ZMB”<br />
was finally financed and built.<br />

STÄDTEBAULICHE ASPEKTE<br />
Drei biomorphe, kompakte, unterschiedlich große Volumen<br />
lassen differenzierte Außenräume zwischen Neubau und<br />
Bestand entstehen. Entspannt fügt sich das Ensemble in die<br />
umgebende städtebauliche Situation ein. Es zitiert den Campusgedanken<br />
als Ausdruck der Eigenständigkeit der Fakultät, bzw.<br />
des Zentrums in der Universitätsstruktur. Die Gruppierung<br />
der Kubaturen ist in der Lage, neue Bewegungsrichtungen in<br />
bzw. durch das Ensemble und damit auch der Umgebung zu<br />
<strong>for</strong>mulieren ohne vorhandene räumliche Qualitäten zu verlieren.<br />
Durchwegungsströme umspülen die Baukörper.<br />
Das Ensemble schafft Orte der Ruhe und der Blicke. Das<br />
Reihen der Neu- und Bestandsbauten um den zentralen Körper<br />
EX_USU (aus der Erfahrung, durch Übung) verstärkt dabei die<br />
Imagination einer Ideenzelle als Knotenpunkt interner wie<br />
externer wissenschaftlicher Arbeit. Er beinhaltet den Hörsaal,<br />
die Seminarräume, die gemeinschaftlich genutzte Bibliothek<br />
sowie das öffentlich zugängliche Café. Der Körper schafft eine<br />
Schnittstelle zwischen Öffentlichkeit, Lehre und Forschung.<br />
Er wird von Architekturstudenten aus aller Welt zu Lehrzwecken<br />
besucht.<br />

„Durchwegungsströme“<br />
Flow of air and motion<br />
Das äußere Erscheinungsbild des Gebäudekomplexes wird<br />
durch Sonnenschutzlamellen aus Glas geprägt. Sie sind<br />
geschosshoch auf der kompletten Fassade montiert. Die<br />
Sonnenschutzlamellen ändern ihren Öffnungswinkel im Tagesverlauf<br />
je nach Sonnenstand. Zusätzlich kann der Nutzer individuellen<br />
Einfluss auf „seine“ Sonnenschutzlamellen nehmen, je<br />
nach persönlichen An<strong>for</strong>derungen diese öffnen oder schließen.<br />
So ergibt sich ein variantenreiches Spiel von geöffneten, halbgeöffneten<br />
und geschlossenen Lamellen. Der Blick wird auf die<br />
darunterliegende, farbintensive Fassade freigegeben.<br />
URBAN PLANNING<br />
Three compact buildings with varied sizes create sophisticated<br />
spaces between new and existing structures. The group of<br />
b uildings blends well into the surroundings. The proximity of the<br />
buildings creates a feeling of a campus and demonstrates the<br />
self-reliance of the faculty. With its specific shape the complex<br />
creates new directions <strong>for</strong> movement along and through the<br />
arrangement of buildings without losing the existing spatial<br />
qualities. Flowing motions wash around the buildings.<br />
The buildings create space <strong>for</strong> silence. Visitors are encouraged<br />
to seek different perspectives from different angles and to<br />
search <strong>for</strong> different viewpoints. The arrangement of new and<br />
old buildings around the central building EX_USU (from Latin:<br />
through experience, through practice) strengthens the idea<br />
of it being the junction between internal and external scientific<br />
activity. It contains an auditorium, lecture rooms, the library<br />
and a public café – the hub where the public, teaching and<br />
research converge.<br />
FACADE<br />
The external views of the building are dominated by vertical<br />
glass-louvres, which function as a sun-shading device. The<br />
louvers extend to the same height as the buildings’ floors and<br />
are mounted all around the exterior. To protect well, the louvers<br />
pivot and follow the course of the sun. As an option, people<br />
working in the building can change the setting of their<br />
“personal” louvers by opening and closing them individually.<br />
The appearance of the facade changes and the richly coloured<br />
cladding appears behind the louvers.<br />

Die Mitglieder des ZMB streben Exzellenz in der Forschung an, was sich u.a.<br />
in zahlreichen nationalen und internationalen Forschungsprojekten manifestiert.<br />
Das derzeitige Finanzvolumen der laufenden eingeworbenen Forschungsprojekte<br />
beträgt mehr als 9,6 Mio Euro. 39 Diplomarbeiten werden derzeit von den<br />
23 wissenschaftlichen Universitätsangehörigen des ZMB betreut, und über<br />
60 DoktorandInnen, PostDocs und nichtwissenschaftliches Personal werden<br />
durch laufende Forschungsprojekte finanziert.<br />

Ziel ist es, bislang<br />
unbekannte Gene und Gen -<br />
produkte, die <strong>für</strong> die zelluläre<br />
Aufnahme, Speicherung und<br />
Mobilisierung von Fetten<br />
verantwortlich sind, zu<br />
identifizieren und zu<br />
charakterisie ren. In diesem<br />
multidiszipli nären Projekt<br />
werden neu entdeckte Gene<br />
und Proteine von Hefe, Maus<br />
und Mensch bezüglich ihrer<br />
biochemischen Eigenschaften,<br />
ihrer physiologischen Funktion<br />

The GEN-AU project<br />
“GOLD – Genomics of<br />
Lipid-associated disorders”<br />
comprises ten research<br />
teams in Graz, Vienna, and<br />
Innsbruck and investigates<br />
the <strong>molecular</strong> basis of<br />
obesity and lipid-associated<br />
disorders. The consortium<br />
aims to identify and<br />
character ize yet unknown<br />
genes and gene products<br />
that are involved in the<br />
cellular uptake, storage,<br />
and mobilization of lipids.<br />
In a multidisciplinary<br />
approach newly identified<br />
genes and proteins from<br />
yeast, mouse, and human will<br />
be analyzed <strong>for</strong> biochemical<br />
properties, physiological<br />
function, and protein<br />
structure. Moreover, genetic<br />
variations in candi date<br />
genes and the association<br />
of these variations with<br />
lipid-associated disorders<br />
are being investigated. The<br />
identification of new targets<br />

bereich (SFB) LIPOTOX ist<br />
ein Konsortium hochkarätiger<br />
Grazer Forschungsgruppen,<br />
die gemeinsam ein zentrales<br />
Thema untersuchen: Lipotoxizität.<br />
Unter Lipotoxizität<br />
versteht man die fehlgesteuerte<br />
Aufnahme, Produktion<br />
oder Aktivität von Fettsäuren<br />
und Lipiden, die zur Bildung<br />
(lipo)toxischer Substanzen<br />
führen, die Fehlfunktionen<br />
von Zellen und Geweben<br />
bewirken und im Zelltod<br />
enden können. Es werden<br />
jene metabolischen Vorgänge<br />
und <strong>molekulare</strong>n Mechanis -<br />
men er<strong>for</strong>scht, die durch lipotoxische<br />
Effektoren ausgelöst<br />
werden und die patholo -<br />
gische Basis verbreiteter<br />
Erkrankungen, wie zB. dem<br />
Metabolischen Syndrom,<br />
Typ-2 Diabetes und Athero -<br />
sklerose, darstellen. Durch<br />
den Einsatz moderner genomischer,<br />

The Joint Research Project<br />
SFB LIPOTOX is a highprofile<br />
research consortium<br />
in Graz that focuses on one<br />
common theme defined as<br />
the anomalous uptake,<br />
generation, and activity of<br />
lipid derivatives mediating<br />
adverse, “lipotoxic” effects<br />
including dysregulation of<br />
metabolic pathways, cell- and<br />
organelle dysfunction, and<br />
cell death. Molecular and<br />
cellular mechanisms activated<br />
by lipotoxic substances are<br />
investigated to identify the<br />
pathological basis of human<br />
disease, such as metabolic<br />
syndrome, type-2 diabetes,<br />
und atherosclerosis. Genomic,<br />
proteomic, and lipidomic<br />
technology will be utilized<br />
to discover novel lipotoxic<br />
pathways. Mutant mouse and<br />
yeast models will be analyzed<br />
to elucidate the mechanisms<br />
that cause the production of<br />
toxic lipid compounds, lead<br />
to cellular dysfunction, and<br />
induce apoptosis or other<br />
<strong>for</strong>ms of cell death.<br />

Der SFB F018 „Molekulare<br />
und Immunologische<br />
Strategien zur Prävention,<br />
Diagnose und Behandlung<br />
von Typ-I-Allergien“ ist ein<br />
Sonder<strong>for</strong>schungsbereich mit<br />
dem Ziel, medizinisch relevante<br />
Allergene zu charakterisieren<br />
und deren Verwendung<br />
in Diagnose und Therapie zu<br />
er<strong>for</strong>schen. Die Kenntnis der<br />
3D-Struktur von Allergenen<br />
ist unentbehrlich <strong>für</strong> die<br />
genaue Analyse von IgE-<br />
Epitopen. Daher ist das Ziel<br />
unseres Projektes die<br />
Strukturaufklärung von<br />
Haupt allergenen unserer<br />
Umwelt und das Design von<br />
hypo-allergenen Proteinen,<br />
die <strong>für</strong> die Entwicklung von<br />
Vakzinen verwendet werden<br />
können. Darüber hinaus<br />
entwickeln wir Methoden zur<br />
Vorhersage von kreuz-reaktiven<br />
Epitopen, basierend auf<br />
den Strukturen und immu -<br />
nologischen Daten von<br />

The SFB F018 “Molecular<br />
and Immunological<br />
Strategies <strong>for</strong> Prevention,<br />
Diagnosis and Treatment<br />
of Type I Allergies” is a<br />
Collaborative Research Center<br />
focused on the <strong>molecular</strong> and<br />
immunological characterization<br />
of medically important<br />
allergens and their use as<br />
diagnostic and therapeutic<br />
tools. The 3D structures of<br />
allergens are essential <strong>for</strong><br />
the precise analysis of<br />
IgE-epitopes. The aim of<br />
our project is the structure<br />
elucidation of major environmental<br />
allergens as a basis<br />
<strong>for</strong> rational design of hypo -<br />
allergenic derivatives <strong>for</strong><br />
vaccine development. In<br />
addition we are developing<br />
methods to predict crossreactive<br />
epitopes of environmental<br />
allergens and food<br />
allergens based on structural<br />
and immunological data.<br />

Forschungsergebnisse zeigen,<br />
kommt Apoptose (programmierter<br />

bei Tieren vor. Außerdem<br />
existieren offenbar mehrere<br />
programmierte und daher<br />
regulierte Wege zum Zelltod.<br />

The TransDeath project<br />
focuses on the fundamental,<br />

IN DEN MOLEKULAREN BIOWISSENSCHAFTEN<br />
Der Lehre kommt am ZMB neben der Forschung höchste<br />
Priorität zu. Neue Bachelor- und Master-Studien in den<br />
Molekularen und Technischen Biowissenschaften werden<br />
im Rahmen von NAWI Graz gemeinsam mit der Technischen<br />
Universität Graz angeboten. NAWI Graz zielt auf eine gemeinsame<br />
Nutzung der Ressourcen beider Universitäten ab, ein<br />
Projekt, das österreichweit durch seinen innovativen Charakter<br />
große Beachtung gefunden hat. Zu den neuen Curricula<br />
zählen das Bachelor-Studium Molekularbiologie sowie die<br />
Master-Studien Biochemie und Molekulare Biomedizin,<br />
Molekulare Mikrobiologie und Biotechnologie. Diese gemein -<br />
sam durchgeführten Curricula im Rahmen von NAWI Graz<br />
bieten ein attraktives Angebot moderner Studien und bieten<br />
die Grundlage <strong>für</strong> ein breites Spektrum an Berufsmöglichkeiten<br />

Teaching, in a highly active research environment, is a top<br />
priority at the ZMB. New bachelor and master studies in<br />
Molecular and Technical Biosciences have recently been<br />
implemented, as part of the NAWI Graz initiative, together<br />
with institutes at the Graz University of Technology. A primary<br />
aim of NAWI Graz has been to conduct joint studies in natural<br />
sciences between the two universities. The project earned<br />
much recognition in Austria due to its highly innovative<br />
character. A new curriculum at the bachelor’s level is offered<br />
in Molecular Biology. Students may further specialize at the<br />
master’s level in Biochemistry and Molecular Biomedicine,<br />
Molecular Microbiology, or Biotechnology. These curricula offer<br />
an attractive spectrum of up-to-date studies, providing a solid<br />
basis <strong>for</strong> job opportunities in academic or industrial research,<br />
production and quality control, chemistry, pharmaceutical<br />
development and biotechnology, environmental and medical<br />

DK MOLEKULARE ENZYMOLOGIE<br />
Der Österreichische Fonds zur Förderung der Wissenschaft -<br />
lichen Forschung (FWF) hat das Förderinstrument Doktoratskolleg<br />
(DK) eingeführt, um die Verbesserung der Doktorats -<br />
ausbildung an Österreichischen Universitäten zu beschleunigen.<br />
DKs stellen eine Projektförderung dar, die in einem hoch<br />
kompetitiven Verfahren an Konsortien von herausragenden<br />
Forscherinnen und Forschern vergeben wird. Das DK Molekulare<br />
Enzymologie war das erste Projekt dieser Art in Graz und<br />
wird bis 2016 laufen. Sechs Mitglieder des ZMB sind Projekt -<br />
leiterinnen bzw. Projektleiter, inkl. der Projektkoordinatorin<br />
Dr. Ellen Zechner. Das Konsortium umfasst Forschergruppen<br />
mehrerer Institute der KFUG und der TUG und wird sehr stark<br />
durch beide Universitäten sowie NAWI Graz unterstützt.<br />
Fast 600 Studierende aus 32 Ländern haben sich <strong>für</strong> Stellen<br />
im Rahmen des DK Programms bisher beworben; davon<br />
wurden 14 hervorragende österreichische und 17 internationale<br />
Studierende <strong>für</strong> das Programm ausgewählt. Neun dieser<br />
DoktoratskandidatInnen werden am ZMB ausgebildet.<br />
Das DK zielt darauf ab, den Studierenden in einem hervorragenden<br />
Forschungsumfeld Fähigkeiten als WissenschafterInnen,<br />
sowie fundierte Karriereentscheidungen zu vermitteln und sie<br />
auf die Verantwortung als ForschungsleiterInnen vorzubereiten.<br />
Unsere Trainingsinitiativen beschränken sich jedoch nicht<br />
nur auf DK-Studierende sondern auf alle Studierende der<br />
Molekularen Biowissenschaften.<br />

DK MOLECULAR ENZYMOLOGY<br />
The Austrian Science Fund (FWF) has adopted the funding<br />
instrument Doktoratskolleg (DK) as a means to accelerate<br />
improvements in doctoral training offered at Austrian<br />
universities. DKs are competitive grants awarded to a<br />
consortium of faculty with strong research records. The DK<br />
Molecular Enzymology was the first grant of this kind to be<br />
awarded in Graz and will continue until 2016. Six ZMB faculty<br />
are members of this DK including the program’s coordinator,<br />
Dr. Ellen Zechner. The consortium of faculty integrates research<br />
groups from several institutes of the KFUG and the TUG.<br />
The project is strongly supported by both universities and<br />
NAWI Graz. Nearly 600 young people from 32 countries have<br />
applied <strong>for</strong> positions linked to the Molecular Enzymology<br />
t raining program. Thus far 14 excellent Austrian and 17 inter -<br />
national students have been recruited. Nine are trained at<br />
the ZMB. The DK aims to ensure an excellent research<br />

Die Organisation von Doktoratsstudien steht europaweit in<br />
einem dynamischen Entwicklungsprozess. Die Re<strong>for</strong>men<br />
betreffen sowohl eine Vereinheitlichung des Doktoratstrainings<br />
auf zumindest drei Jahre sowie eine Stärkung der wissenschaftlichen<br />
Komponente der Doktorarbeit. An der KFU Graz nimmt<br />
das ZMB eine Führungsposition in diesem Entwicklungsprozess<br />
ein: dies betrifft zum einen das DK Molekulare Enzymologie<br />
als auch die Leitung der Doktoratsschule <strong>für</strong> Molekulare<br />
Biowissenschaften und Biotechnologie. Diese Doktoratsschule<br />
umfasst mehr als 200 UniversitätslehrerInnen und Studierende<br />
an verschiedenen Instituten der KFUG und der TUG. Unter<br />
dem Motto „exzellente Doktoratsausbildung durch exzellente<br />
Forschung“ wird dabei besonders auf die Qualität des<br />
Forschungsumfelds geachtet. Das Programm hat sich zum Ziel<br />
gesetzt, die Interdisziplinarität des Trainings sowie die inter -<br />
nationale Mobilität sowohl durch Rekrutierung aus ländischer<br />
Studierender als auch durch die Förderung von Auslands -<br />
aufenthalten zu <strong>for</strong>cieren. Die laufende Überprüfung des<br />
wissenschaftlichen Fortschritts erfolgt durch inter-institutionelle<br />
und inter-universitäre Komitees. Konkrete Maßnahmen tragen<br />
dazu bei, die Tiefe und Vielfalt von spezifischen Lehrinhalten zu<br />
verbessern. Durch ein breites Angebot an Intensiv-Workshops<br />
über ethische, persönliche und wissenschafts-relevante<br />
Fähigkeiten, die zur Entwicklung einer erfolgreichen Wissenschaftskarriere<br />
beitragen sollen, hat das ZMB in Graz einmal<br />
mehr bei der Ausbildung junger Studierender einen neuen<br />

The organization of doctoral education is in a dynamic state<br />
of change across Europe. Re<strong>for</strong>ms include standardizing the<br />
length of training to at least 3 years and strengthening the<br />
component of original research. At the University of Graz,<br />
the ZMB is taking a leadership role in doctoral studies not only<br />
by coordinating the DK Molecular Enzymology but also by<br />
directing the Doctoral School of Molecular Biosciences and<br />
Biotechnology. This doctoral school comprises over 200 faculty<br />
and students from several institutes at the KFUG and the TUG.<br />
Based on the premise that excellence in doctoral training<br />
depends on excellence in research we place a heavy emphasis<br />
on the quality of the research environment. The program strives<br />
to broaden the interdisciplinary nature of training, and to<br />
increase international mobility both in recruiting students and<br />
in facilitating research stays abroad. Supervision of thesis work<br />
is expanded to inter-institutional and inter-university faculty<br />
teams. Concrete steps have been taken to improve the depth<br />
and variety of instruction. The ZMB has set precedence in Graz<br />
<strong>for</strong> training young scientists by offering intensive workshop<br />
programs in ethical, personal, and professional research-related<br />
skills essential to their development as future research leaders.<br />

Durch vermehrte Reiseaktivitäten sind bakterielle Infektionskrankheiten<br />
auf dem Vormarsch und stellen ein besorgnis -<br />
erregendes Gefährdungspotenzial <strong>für</strong> den Menschen dar.<br />
Einer der Faktoren zur Beschleunigung dieser Entwicklung<br />
ist die einhergehende, sich rasch ausbreitende Antibiotika -<br />
resistenzbildung bei pathogenen Bakterien. Diese Resistenzen<br />
können leicht weitergegeben werden, da die Bakterien im<br />
Laufe der Evolution sehr effiziente Übertragungsmechanismen<br />
entwickelt haben. Zusätzlich passen sich pathogene Bakterien<br />
sehr schnell an wechselnde Wirtsbedingungen an und besitzen<br />
ein großes Repertoire an <strong>molekulare</strong>n Mechanismen und<br />
Virulenzfaktoren. Die Infektionsgruppen am ZMB beschäftigen<br />
sich mit den <strong>molekulare</strong>n Mechanismen der Adaptation,<br />
Kolonisierung und Persistenz einiger relevanter Erreger <strong>für</strong><br />
Mensch, Tier und Pflanzen. Um die Infektionsmechanismen<br />
besser verstehen zu können werden verschiedene Interaktionsmechanismen<br />
zwischen Wirt und Erreger, infektionsrelevante<br />
Stoffwechselwege und Strukturen der bakteriellen Oberfläche<br />
studiert. Ein Forschungsschwerpunkt dient beispielsweise der<br />
Charakterisierung von Transportmechanismen, welche die<br />
Bakterien zur Verbreitung von Antibiotikaresistenz- und<br />
Virulenzfaktoren benützen. Die <strong>molekulare</strong> Charakterisierung<br />
dieser Transportmechanismen und bakteriellen Oberflächenstrukturen<br />
sind von zentraler Bedeutung und werden <strong>für</strong> die<br />
Entwicklung neuer antibakterieller Therapien in der Zukunft<br />

Infectious disease due to bacterial pathogens is a growing<br />
c hallenge to human health. The severity of the problem is<br />
multiplied by a continuous increase in antibiotic resistance<br />
within the microbial world. Resistance spread among bacteria<br />
occurs frequently because of the many processes that bacteria<br />
have evolved to mobilize genes from one bacterium to another.<br />
Moreover, pathogenic bacteria adapt very effectively to<br />
c hallenging environmental changes such as a host immune<br />
response or antibiotic action. Research in the Bacterial<br />
Pathogenesis Groups of the ZMB analyzes the <strong>molecular</strong><br />
mechanisms underlying optimal growth, adaptation,<br />
colonization, and persistence of pathogens that infect<br />
humans, animals and plants. To understand the outcome of<br />
pathogen–host interactions our research focuses on metabolic<br />
pathways and structural features of the cell surface. In the latter<br />
category, we pay particular attention to the macro<strong>molecular</strong><br />
transport systems that enable bacteria to export virulence<br />
factors as well as antibiotic resistance genes. Molecular<br />
c haracterization of adaptive processes and surface exposed<br />
structures may in turn provide alternative targets <strong>for</strong> anti -<br />
bacterial therapies in the future.<br />

Die Bäckerhefe Saccharomyces cerevisiae ist ein einzelliger<br />
Eukaryot mit relativ kleinem Genom, das leicht untersuchbar<br />
und manipulierbar ist. Die meisten Bestandteile grundlegender<br />
zellulärer Funktionen sind zwischen Hefe und menschlichen<br />
Zellen konserviert, daher ist Hefe ein exzellenter Modellorganismus<br />
<strong>für</strong> die Untersuchung komplexer Regulationsvorgänge.<br />
Wir setzen Hefe zur Untersuchung der grundlegenden<br />
Prinzipien von Apoptose, Alternsvorgängen und der Biogenese<br />
der Ribosomen ein, sowie zur Identifizierung wichtiger an<br />
diesen Vorgängen beteiligter Gene. Apoptose ist das zelluläre<br />
Selbstmordprogramm, in höheren Organismen unverzichtbar<br />
<strong>für</strong> die normale Entwicklung und zum Schutz des Organismus<br />
vor Krebs und Virusinfektionen. Wegen der Konservierung des<br />
apoptotischen Apparates können wir in Hefe nach Substanzen<br />
suchen, die den Todesprozess stimulieren oder hemmen,<br />
und so auch die Lebensdauer eines Organismus beeinflussen.<br />
Wie menschliche Zellen altert auch Hefe im Laufe der Zeit<br />
(chronologisches Altern) und durch wiederholte Zellteilung<br />
(replikatives Altern). Wir untersuchen konservierte zentrale<br />
Alternsregulatoren und Exekutoren, wie Mitochondrien,<br />
oxidativen Stress und Apoptose. Bemerkenswerterweise wurde<br />
der einzige bekannte universell konservierte Altersregulator,<br />
SIR2, zuerst in Hefe entdeckt. Ein weiterer Schwerpunkt<br />
unserer Arbeit sind Substanzen, die die Ribosomen-Biogenese<br />
beeinflussen.<br />

The baker’s yeast Saccharomyces cerevisiae is a unicellular<br />
eukaryotic organism with a relatively small genome that is easily<br />
accessible to genetic manipulation. Most components of basic<br />
cellular functions are conserved between yeast and human<br />
cells. There<strong>for</strong>e, yeast are an excellent model organism to<br />
investigate complex regulatory processes. We use yeast cells<br />
to unravel the underlying principles of apoptosis, aging and<br />
ribosome biogenesis and to identify essential genes involved<br />
in these complex processes. Apoptosis is a cellular suicide<br />
program that has a vital role in normal development of multi -<br />
cellular organisms and acts protectively in cancer and viral<br />
infections. Since the basic apoptotic apparatus is conserved,<br />
we can screen yeast <strong>for</strong> drugs stimulating or inhibiting the<br />
death process and thus affect the life span of an organism.<br />
Like human cells, yeast age both over time (chronological<br />
aging) and by repeated cell division (replicative aging).<br />
We investigate the conserved central aging regulators and<br />
executors, as mitochondria, oxidative stress or apoptosis.<br />
Of note, the only established ubiquitously conserved aging<br />
regulator, SIR2, was originally discovered in yeast.<br />
Drugs affecting ribosome biogenesis, a process essential<br />
to organisms are another focus of our work.<br />

DES LIPID- UND ENERGIESTOFFWECHSELS<br />
Lipide sind <strong>für</strong> alle Lebewesen als Energie-Substrate und<br />
Bestandteil biologischer Membranen von größter physiologischer<br />
Bedeutung. Die zentralen Forschungsthemen der Lipid-<br />
Arbeitsgruppen umfassen die Entdeckung der enzymatischen<br />
Mechanismen der Synthese und des Abbaus von Lipiden.<br />
Ein tief greifendes Verständnis dieser Prozesse ist deshalb so<br />
wichtig, weil Defekte und Fehlregulationen des Lipid- und<br />
E nergiehaushaltes bei der Entstehung der weltweit häufigsten<br />
Stoffwechselerkrankungen beteiligt sind, nämlich Fettleibigkeit,<br />
Diabetes und Atherosklerose.<br />
In mehreren Groß<strong>for</strong>schungsprojekten, wie dem SFB Bio -<br />
membranen und dem Österreichischen Genomprojekt GEN-AU,<br />
haben wir uns speziell mit Lipid-abbauenden Enzymen, so<br />
genannten Lipasen beschäftigt. Dabei ist uns die Entdeckung<br />
einer bislang unbekannten Lipase, der ATGL gelungen. Dieses<br />
Enzym initiiert den Fettabbau in Adipozyten (Fettzellen) und<br />
anderen Körperzellen und benötigt <strong>für</strong> diese Funktion einen<br />
ebenfalls von unserer Gruppe entdeckten Coaktivator, CGI-58.<br />
Genetisch modifizierte Mäuse, denen das Enzym und/oder der<br />
Coaktivator fehlen, zeigen schwere Stoffwechselstörungen<br />
und werden derzeit intensiv in unserem Labor untersucht.<br />
Lipid- und Energiestoffwechsel sind auf zellulärer Ebene hoch<br />
konserviert. Wir machen uns daher auch das einzellige Modellsystem<br />
Hefe zunutze, um grundlegende zelluläre Prozesse der<br />
Lipid- und Energiehomöostase zu analysieren und im Kontext<br />
der Lipid-assoziierten Erkrankungen zu verstehen.<br />
Im Rahmen eines neu bewilligten Großprojektes (SFB-Lipotox),<br />
das im Frühjahr 2007 gestartet wurde, untersuchen die Lipidgruppen<br />
am ZMB Enzyme, die an der Konversion von an sich<br />
physiologisch wichtigen Lipiden zu gefährlichen, krankheits -<br />
verursachenden Substanzen beteiligt sind. Die Studien lassen<br />
einerseits Schlüsse auf <strong>molekulare</strong> Ursachen Lipid-assoziierter<br />
krankhafter Veränderungen zu, und bieten andererseits inter -<br />
essante neue Ansatzpunkte <strong>für</strong> pharmakologische Intervention.<br />

Lipids play an instrumental physiological role as energy<br />
sub strates, mediators in cell signaling pathways, and<br />
constituents of cell- and organelle biomembranes.<br />
The research activities of our laboratories focus on the<br />
enzymology of lipid synthesis and degradation. A detailed<br />
understanding of these processes is important because<br />
dysregulation of lipid and energy homeostasis is directly<br />
associated with many prevalent metabolic diseases such as<br />
obesity, diabetes, and atherosclerosis.<br />
Within the SFB Biomembranes and the Austrian genome<br />
project GEN-AU, we have specifically worked with lipases<br />
(lipid hydrolases) that affect the metabolism of plasma<br />
lipoproteins and the lipid metabolism in fat cells, muscle, and<br />
macrophages. A key finding of this work was the discovery<br />
of a previously unknown fat degrading enzyme called adipose<br />
triglyceride lipase (ATGL). This enzyme is responsible <strong>for</strong> the<br />
initial step of triacylglycerol hydrolysis in adipose and nonadipose<br />
tissues. ATGL requires an activator called CGI-58 <strong>for</strong><br />
full enzyme function. Transgenic and knock-out mouse models<br />
that overexpress or lack the enzyme and/or its activator are<br />
currently analyzed to elucidate the physiological role of the<br />
ATGL/CGI-58 complex in mammalian physiology. Lipid and<br />
energy homeostasis is remarkably conserved in the single cell<br />
model organism, yeast, which we exploit to understand the<br />
<strong>molecular</strong> and cellular regulation of lipid homeostasis, in<br />
comparison to mammalian systems.<br />
In addition, the lipid groups are involved in identifying and<br />
characterizing novel enzymes that modulate the generation or<br />
catabolism of lipid mediators, both in yeast and mouse model<br />
systems (funded by the SFB-Lipotox). The goal of these<br />
studies is to elucidate the enzymatic pathways behind the<br />
conversion of physiological lipid substrates into noxious<br />
(lipotoxic) compounds, which can cause cell dysfunction as<br />
well as cell death. These studies are expected to yield important<br />
insights into causes of lipid-associated disorders at the<br />
<strong>molecular</strong> level, and to provide novel avenues <strong>for</strong> developing<br />
drugs <strong>for</strong> pharmacological intervention.<br />

Die strukturbiologischen Arbeitsgruppen beschäftigen sich<br />
mit der Bestimmung der dreidimensionalen Struktur von<br />
Proteinmolekülen. Die modernen Methoden der Röntgen -<br />
strukturanalyse sowie der magnetischen Kernresonanz-<br />
S pektroskopie erlauben es, die relative räumliche Anordnung<br />
aller Atome in einem Molekülverband sichtbar zu machen.<br />
Diese ungemein leistungsfähigen Methoden haben in den<br />
letzten zehn Jahren zu völlig neuen Einsichten über die<br />
<strong>molekulare</strong> Funktion sowie den <strong>molekulare</strong>n Mechanismus<br />
biologischer Makromoleküle geführt. Sie er<strong>for</strong>dern einen<br />
hohen instrumentellen Aufwand, der in Österreich nur an<br />
ganz wenigen Orten realisiert ist. Die Forschungsinteressen<br />
der strukturbiologischen Arbeitsgruppen beinhalten Enzyme<br />
mit einem B12-Cofaktor, biokatalytisch wichtige Enzyme,<br />
Enzyme des Lipidmetabolismus, Allergene sowie S-Layer<br />
Proteine.<br />

The structural biology research groups are dealing with the<br />
elucidation of the three-dimensional structure of proteins.<br />
The methods of X-ray Crystallography and Nuclear Magnetic<br />
Resonance Spectroscopy (NMR) enable the visualization of the<br />
spatial positions of all atoms in a macromolecule. In the last<br />
decade these highly powerful methods have allowed <strong>for</strong> novel<br />
insights into the <strong>molecular</strong> functions and mechanisms of<br />
bio logical macromolecules and their assemblies. However both<br />
methods require a high degree of know-how and complex<br />
instrumentation, available only in a few laboratories in Austria.<br />
Main research interests of the structural biology groups include<br />
B12-dependent enzymes, enzymes important to biocatalysis,<br />
enzymes in lipid metabolism, allergens and bacterial S-layer<br />
proteins.<br />
