Preparation of 20-L carboys, bags and tanks<br />

h Wash with diluted chlorine (10%), rinse and leave<br />

to dry.<br />

h Fill with 1-µm filtered and UV-sterilised<br />

seawater.<br />

h Add chlorine (12.5% active) at a concentration of<br />

0.2 ml/L for a minimum of 4 hours, or overnight<br />

for up to 24 hours without aeration.<br />

h Turn on the aeration for 10–15 minutes to remove<br />

residual chlorine. Add sodium thiosulphate (stock<br />

solution at 250 g/L) at a rate of 0.2 ml/L for<br />

1 hour to neutralise residual chlorine. Conduct a<br />

chlorine test.<br />

After several minutes, add agricultural fertiliser<br />

such as Aquasol (2.5 g/100 L). For diatom cultures,<br />

also add sodium metasilicate (3.75 g/100 L).<br />

Inoculate with algal cultures.<br />

Sodium metasilicate is added for diatom cultures such as Chaetoceros sp., Navicula sp. and Nitzschia sp.;<br />

diatoms use the silicate for production of an external shell.<br />
