Euthanasia-Amphibians<br />
Euthanasia must only be undertaken by trained personnel and must be in<br />
consultation with the Animal Welfare Officer if being undertaken outside of approved<br />
application (eg emergency situations).<br />
60mg/ml Pentobarbitone∗ – 60mg-100mg/kg - IV or IC<br />
Buffered MS222 >200mg/kg intracoelomic or immersion 200mg/L**<br />
Aqui-S immersion- 40mg/ml<br />
Pithing, exsanguination, freezing and decapitation ONLY<br />
after anaesthesia∗∗*<br />
Destruction of CNS, heart removal, high dose of cardioplegic agent (lignocaine, K+<br />
or Mg + salts) may be necessary to prevent recovery. Destruction of CNS .<br />
Decapitation is ONLY acceptable after anaesthesia or destruction of the CNS∗∗<br />
* Pentobarbitone is irritant to tissues when injected into a body cavity or<br />
perivascularly. It also leaves crystalline deposits rendering the contaminated<br />
tissues useless for histopathological examination. Where possible<br />
barbiturates should always be injected intravenously<br />

**MS-222 can change the pH to more acidic when added to water. This complication<br />
is mitigated if the water is well buffered, eg. has adequate alkalinity to buffer the pH<br />
change. The alkalinity of water should be known if the water is to be used in an<br />
anaesthetic bath. Water with alkalinity ó50 mg/L should either not be used in MS-222<br />
anaesthesia or the water first treated with a buffer (e.g. sodium hydroxide) to counter<br />
the low pH effect of MS-222.<br />

Physical methods of euthanasia should only be performed by an<br />
appropriately trained operator.<br />
Disposal<br />
<br />
Euthanased animals must be placed in a plastic bag and put in the carcass bin in<br />
the level 1 freezer (biology room 130). Disposal will be treated as medical waste<br />
and will be incinerated at <strong>Flinders</strong> Medical Centre.<br />
SOP Review<br />
This SOP currently applies to the animals housed in the Biological Sciences Animal<br />
House facilities and field sites named in approved applications from the AWC. This<br />
<strong>Standard</strong> <strong>Operating</strong> <strong>Procedure</strong> applies to fish(excluding sharks) and Cephalopods<br />
only, with some guidance on current practices for Marine Mammals. This SOP will be<br />
reviewed 5 yearly, but also updated more frequently as policies, techniques and<br />
animal care requirements change<br />
Any questions regarding the above guidelines and any technical advice/ assistance<br />
required can be directed to Animal Facilities Officer. Enquiries regarding changes to<br />
space requirements, modifications, should be sent via email in the first instance<br />
