SDS-, fatty acid-, pH-, Zn 2+ -concentration dependent proteolytic activity measurement of 20S<br />

proteasome from various sources.<br />

Subunit compositional analysis of 20S proteasome by 1- and 2-dimensional electrophoresis, Western<br />

blotting.<br />

Identification of all subunits of 20S proteasome by MALDI-TOF MS.<br />

Investigation of proteolytic activity of 20S proteasome by shift of cell culture from permissive<br />

temperature to restrictive temperature.<br />

Complementation study of yeast mutant.<br />

Proposition of proteasome channel in rpt6-1 yeast mutant.<br />

Isolation and Characterization of antagonistic Streptomyces species.<br />

Selection of the suitable medium for optimum growth of the organism.<br />

ACHIEVEMENT OF RESEARCH<br />

Ph. D:<br />

Very low level of proteasome activity was found in rpt6-1 yeast mutant.<br />

Subunit composition pattern of 20S proteasome in rpt6-1 mutant was different to that of wild-type<br />

counterpart.<br />

Two subunits of proteasome in rpt6-1, α1 & α7, showed slow mobility and different isoelectric<br />

focusing in 2-DE compared to that of wild type.<br />

The changes on α1, α7 were not a consequence of phosphorylation.<br />

The changes on α1, α7 were disappeared when culture temperature had been shifted to restrictive<br />

temperature.<br />

A precursor form of α1, α7 regulate the proteasome channel in absence of Rpt6 ATPase.<br />

M. Pharm.:<br />

Two antimicrobial compounds were isolated. One of which is di- (2-ethyl hexyl) phalate and another<br />

one had not been identified due to its poor amount.<br />

All the compounds showed moderate to strong antimicrobial activities.<br />

Biochemical parameters were normal on subacute toxicity studies on rats with the isolated<br />

compounds.<br />

TRAINING OBTAINED<br />

An in-plant training of the period of six weeks duration had been taken in “Beximco Pharmaceutical<br />

Company Ltd” an ISO certified leading pharmaceutical company in Bangladesh. During the training<br />

period dispensing, manufacturing, packaging, in process quality control operation of various<br />

pharmaceutical products of different dosage forms were observed and carried out in practice.<br />

A cGMP and Validation training had been taken in “Opsonin Chemical Industries Ltd<br />

(Pharmaceutical Branch)” another ISO certified leading pharmaceutical company in Bangladesh.<br />
