Gel Protein Recovery ® (GPR)
Protea’s Gel Protein Recovery System (GPR) is an advanced,
microfluidics-based system for the rapid and efficient extraction
of proteins from polyacrylamide gels.
The GPR-800 system uses a proprietary plastic microfluidic
chip in a high voltage environment, to allow fast electroelutions
simultaneously in eight parallel microchannels. The GPR
Electroelution Buffers contains a proprietary mass spec friendly
surfactant that allows for easy degradation and clean-up prior to
mass spectrometry. Protea’s GPR-800 is a closed system with
minimal dead volume, assuring no introduction of contaminants.
FEATURES:
Advanced Microfluidic Chip Design – Allows for
efficient extraction of proteins down to 100 ng. Intact mass
measurements are routinely achieved from coomassie stained
gels. Limited surface area greatly improves recovery rate.
Eight Channel Parallel Processing – 8 gel bands can be
processed simultaneously in less than 20 minutes. More than 100
proteins can be recovered in a standard work day.
Proprietary Decoupled Protein Recovery – Allows
for easy and efficient collection of protein samples for post
electroelution clean-up.
System and Reagents Designed for Mass
Spectrometry – Proteins can be mass spectrometry ready
without extensive clean-up.
Closed System and Disposable Chips – No worries about
contamination or carry over. Use a new, clean chip for each
recovery.
RESEARCH APPLICATIONS:
Top-Down Proteomics – Provides the ability to combine the
resolution and quantitation of 2D electrophoresis with top-down
proteomics using state of the art mass spectrometry techniques.
Intact Mass Measurement – Quickly confirm protein mass
from 1D and 2D gels using mass spectrometry.
Determine Modification State of Proteins – Use the
resolving power of 2D electrophoresis to resolve isoforms,
degradation products, and modifications states. Quickly recover
separated species for mass spectrometry analysis.
Recover and Store – Proteins can rapidly be recovered from
1D or 2D gels and stored for later experimentation.
Effective Range of the GPR
The plot shows the amount of protein needed (in gel) as
a function of molecular weight for successful usage of
the GPR-800. In general, higher protein loads and lower
molecular weight proteins yield the best results. The blue
shaded area represents the range of protein gel loads where
it is routine to obtain an intact mass measurement after
protein recovery. The area shaded in green represents the
range where it is routine to successfully identify a recovered
protein after digestion using the GPR-800. It should be
noted these areas represent areas of routine success. In
many cases the ranges can be extended. For example we
can, in many cases, show intact mass measurements for
small proteins with loads down to 100 ng. In addition, we
have successfully performed post electroelution digestion on
many larger proteins (75 kDa) down to 250 ng.
The GPR-800 was used to recover
protein isoforms of myoglobin that
were separated by 2-dimensional
gel electrophoresis. Intact mass
measurements showed that each
resolved spot had statistically
different masses. This approach
allows researchers to easily
quantitate and separate protein
isoforms using gel electrophoresis
followed by the rapid and efficient
recovery of the intact molecules
from the gel. Top down mass
spectrometric techniques can then
be used to determine intact mass
and sequence information for each
protein isoform.
ProteaGels 5%, 10%, 12% and 15% Solutions
ProteaGels “ready-to-run” SDS polyacrylamide solutions
polymerize into an advanced molecular sieve for the
electrophoretic separation of proteins. Because of the advanced
buffer chemistry used in the gel matrix solution, ProteaGels allow
a single separating gel. A stacking gel is not required, as the
ProteaGel proprietary formulation inherently stacks the protein
samples during the normal electrophoresis run. However, for the
highest resolution a stacking gel usng 5% ProteaGel solution can
be used. Band resolution is unparalleled over a molecular range
of 2.5 to 250 kDa. The new hybrid formulation of ProteaGels
gives these gels an increased gel strength, which allows for
easier handling. ProteaGels will work with all types of universal
electrophoresis apparatus. Our gel mixtures are formulated for
optimal performance in mass spectrometry-based proteomics
experiments.
FEATURES:
• High gel strength – allows easier handling
• Ready to use in less than 10 minutes – just add TEMED and
APS to polymerize the gel
• No stacking gel required – permits longer gel separations
• High resolution gels for protein separation across a broad
molecular weight range
• Stable at room temperature for 1 year
• Each batch tested for MS compatibility – Certified MS Grade
SuperBlue G250 and R250 Coomassie Stains
Protea’s SuperBlue Coomassie stains are an excellent choice for
visualizing proteins separated by SDS-PAGE gel electrophoresis.
Our SuperBlue G250 stain is a colloidal Coomassie formulation
with a sensitivity of ~20-40 ng protein. As a colloidal stain, the
SuperBlue G250 reacts only with proteins and not the gel itself,
resulting in a reduced background staining that permits rapid
visualization of the gel bands. Protea’s SuperBlue R250 stain
is a traditional Coomassie stain, with a sensitivity of ~20-40 ng.
Because Coomassie stains interact stoichiometrically with protein
in gels, they are useful for quantitation by densitometry. The
SuperBlue G250 stain is available as a ready-to-use formulation,
while the SuperBlue R250 stain is provided as a 10X concentrate
solution.
FEATURES:
• Sensitive – as low as 20 - 40 ng/band
• Non-fixative and fixative protocols available
• Useful for quantitation by densitometry
RESEARCH APPLICATIONS:
• Staining 1D and 2D SDS-PAGE gels
• Protein studies requiring non-fixative gel staining
• Densitometry studies for protein quantitation in SDS-PAGE
Silver Stain Kit
Silver staining of proteins is a useful technique for the detection
of minor protein components that are difficult to visualize by
standard Coomassie blue stains. Silver stains are 50 to 100-fold
more sensitive than Coomassie and thus capable of detecting
nanogram quantities (~2 ng) of protein on gels in comparison to
Coomassie stains (20 - 40 ng).
The Protea Silver Stain formulation is unique in that the staining
process can be completed in about 35 minutes (including the 15
min. fixing step) without sacrificing the sensitivity of detection. In
addition, the silver stain kit is compatible with MALDI analysis,
especially when used in conjunction with our silver destaining
reagent.
FEATURES:
• 35 minutes from fixation to visualization of protein bands
• Stable at room temperature for up to 6 months
• Sensitivity (1-2 ng) is 50 to 100x greater than that of
Silver Destaining Solution
Silver Staining is a very powerful technique to detect
small amounts protein after one- or two-dimensional gel
electrophoresis. Unfortunately, the silver deposits on the protein
tend to interfere with mass spectrometry analysis, and thus its
precise identification. Protea’s Silver Destaining Solution is readyto-use
and removes most if not all of the silver from proteins and
permits mass spectrometry analysis.
Zinc Stain
ZincPlus Stain Kits reagents are used to stain one- or twodimensional
gels following SDS-PAGE. The ZincPlus Stain is a
negative stain, which produces a whitish opaque background in
the gel, while the protein bands themselves remain unstained.
The inherent benefit of a negative stain is that the protein band
does not contain any dye or stain to present as a contaminant for
downstream sample processing and analysis. ZincPlus Stain Kit is
a fast and reversible method for visualizing proteins separated on
polyacrylamide gels. The ZincPlus Stain has a detection limit of ~1
ng of protein, and the entire staining process is completed within
15 minutes. Unlike most stains, ZincPlus Stain does not require a
fixation step, which leaves the proteins unaffected for downstream
processing.
FEATURES:
• Negative stain: no dye or stain contaminating the protein bands
• Fast staining technique:

TriColor Pre-stained Protein Molecular Weight Markers
Protea’s Blue and TriColor Pre-stained Protein Molecular Weight
Markers are designed for use with SDS-PAGE gels. They cover
a wide range of molecular weights from 7 to 175 kDa. Blue
Pre-stained Protein Molecular Weight Markers are comprised
of 8 proteins covalently coupled to a blue dye, allowing easy
visualization during electrophoresis. TriColor Pre-stained Protein
Molecular Weight Markers are comprised of 8 proteins covalently
coupled to a colored dye, including one orange and one green
band, allowing easy visualization during electrophoresis with
two main reference points. By covalently coupling the dye to the
protein, the electrophoretic behavior of proteins is influenced.
Due to this behavior change of pre-stained proteins, Protea’s
Protein Molecular Weight Marker (MM-101-1) should be used
when specific molecular weight determination is desired.
FEATURES:
• Reproducible markers
• Wide MW range from 7 to 175 kDa
• Real-time visualization of separation during electrophoresis
RESEARCH APPLICATIONS:
• SDS-PAGE separation of protein samples
• 1D and 2D gel electrophoresis
