<strong>Protea</strong>’s <strong>Gel</strong> <strong>Protein</strong> <strong>Recovery</strong> System (GPR) is an advanced,<br />
microfluidics-based system for the rapid and efficient extraction<br />
of proteins from polyacrylamide gels.<br />
The GPR-800 system uses a proprietary plastic microfluidic<br />
chip in a high voltage environment, to allow fast electroelutions<br />
simultaneously in eight parallel microchannels. The GPR<br />
Electroelution Buffers contains a proprietary mass spec friendly<br />
surfactant that allows for easy degradation and clean-up prior to<br />
mass spectrometry. <strong>Protea</strong>’s GPR-800 is a closed system with<br />
minimal dead volume, assuring no introduction of contaminants.<br />

RESEARCH APPLICATIONS:<br />
Top-Down Proteomics – Provides the ability to combine the<br />
resolution and quantitation of 2D electrophoresis with top-down<br />
proteomics using state of the art mass spectrometry techniques.<br />
Intact Mass Measurement – Quickly confirm protein mass<br />
from 1D and 2D gels using mass spectrometry.<br />
Determine Modification State of <strong>Protein</strong>s – Use the<br />
resolving power of 2D electrophoresis to resolve isoforms,<br />
degradation products, and modifications states. Quickly recover<br />
separated species for mass spectrometry analysis.<br />
Recover and Store – <strong>Protein</strong>s can rapidly be recovered from<br />
1D or 2D gels and stored for later experimentation.<br />
Effective Range of the GPR<br />
The plot shows the amount of protein needed (in gel) as<br />
a function of molecular weight for successful usage of<br />
the GPR-800. In general, higher protein loads and lower<br />
molecular weight proteins yield the best results. The blue<br />
shaded area represents the range of protein gel loads where<br />
it is routine to obtain an intact mass measurement after<br />
protein recovery. The area shaded in green represents the<br />
range where it is routine to successfully identify a recovered<br />
protein after digestion using the GPR-800. It should be<br />
noted these areas represent areas of routine success. In<br />
many cases the ranges can be extended. For example we<br />
can, in many cases, show intact mass measurements for<br />
small proteins with loads down to 100 ng. In addition, we<br />
have successfully performed post electroelution digestion on<br />
many larger proteins (75 kDa) down to 250 ng.<br />

The GPR-800 was used to recover<br />
protein isoforms of myoglobin that<br />
were separated by 2-dimensional<br />
gel electrophoresis. Intact mass<br />
measurements showed that each<br />
resolved spot had statistically<br />
different masses. This approach<br />
allows researchers to easily<br />
quantitate and separate protein<br />
isoforms using gel electrophoresis<br />
followed by the rapid and efficient<br />
recovery of the intact molecules<br />
from the gel. Top down mass<br />
spectrometric techniques can then<br />
be used to determine intact mass<br />
and sequence information for each<br />
protein isoform.<br />

The <strong>Protea</strong><strong>Gel</strong> Mini Precast <strong>Gel</strong>s have been designed to<br />
improve polyacrylamide gel electrophoresis (PAGE) for both<br />
1D and 2D protein separations. <strong>Protea</strong><strong>Gel</strong> polyacrylamide gel<br />
formulations are featured in the mini precast gels, permitting<br />
high resolution protein gel separation across a broad molecular<br />
weight range. The BR series of <strong>Protea</strong><strong>Gel</strong> Mini Precast <strong>Gel</strong>s are<br />
compatible with the Bio-Rad Mini-PROTEAN ® , Mini-PROTEAN<br />
Tetra cell, and the Mini-PROTEAN 3 Dodeca cells, as well as,<br />
other standard mini gel systems. The IV series of <strong>Protea</strong><strong>Gel</strong><br />
Mini Precast gels are compatible with the Invitrogen XCell6<br />
Multi<strong>Gel</strong> and XCell SureLock Mini electrophoresis systems.<br />
The <strong>Protea</strong><strong>Gel</strong> Mini product line offers a variety of precast gels to<br />

<strong>Protea</strong><strong>Gel</strong>s “ready-to-run” SDS polyacrylamide solutions<br />
polymerize into an advanced molecular sieve for the<br />
electrophoretic separation of proteins. Because of the advanced<br />
buffer chemistry used in the gel matrix solution, <strong>Protea</strong><strong>Gel</strong>s allow<br />
a single separating gel. A stacking gel is not required, as the<br />
<strong>Protea</strong><strong>Gel</strong> proprietary formulation inherently stacks the protein<br />
samples during the normal electrophoresis run. However, for the<br />
highest resolution a stacking gel usng 5% <strong>Protea</strong><strong>Gel</strong> solution can<br />
be used. Band resolution is unparalleled over a molecular range<br />
of 2.5 to 250 kDa. The new hybrid formulation of <strong>Protea</strong><strong>Gel</strong>s<br />
gives these gels an increased gel strength, which allows for<br />

SuperBlue G250 and R250 Coomassie Stains<br />
<strong>Protea</strong>’s SuperBlue Coomassie stains are an excellent choice for<br />
visualizing proteins separated by SDS-PAGE gel electrophoresis.<br />
Our SuperBlue G250 stain is a colloidal Coomassie formulation<br />
with a sensitivity of ~20-40 ng protein. As a colloidal stain, the<br />
SuperBlue G250 reacts only with proteins and not the gel itself,<br />
resulting in a reduced background staining that permits rapid<br />
visualization of the gel bands. <strong>Protea</strong>’s SuperBlue R250 stain<br />
is a traditional Coomassie stain, with a sensitivity of ~20-40 ng.<br />

Silver Stain Kit<br />
Silver staining of proteins is a useful technique for the detection<br />
of minor protein components that are difficult to visualize by<br />
standard Coomassie blue stains. Silver stains are 50 to 100-fold<br />
more sensitive than Coomassie and thus capable of detecting<br />
nanogram quantities (~2 ng) of protein on gels in comparison to<br />
Coomassie stains (20 - 40 ng).<br />

Silver Staining is a very powerful technique to detect<br />
small amounts protein after one- or two-dimensional gel<br />

Zinc Stain<br />
ZincPlus Stain Kits reagents are used to stain one- or twodimensional<br />
gels following SDS-PAGE. The ZincPlus Stain is a<br />
negative stain, which produces a whitish opaque background in<br />
the gel, while the protein bands themselves remain unstained.<br />
The inherent benefit of a negative stain is that the protein band<br />
does not contain any dye or stain to present as a contaminant for<br />
downstream sample processing and analysis. ZincPlus Stain Kit is<br />
a fast and reversible method for visualizing proteins separated on<br />
polyacrylamide gels. The ZincPlus Stain has a detection limit of ~1<br />
ng of protein, and the entire staining process is completed within<br />
