Unique Patented Image Positioning and Slant Correction<br />

Positioning tools on ULTima AQ software help position gels or other samples<br />

from the computer. The positioning is easy, accurate and fast with minimum<br />

handling of fragile gels or other samples. Every user of an imaging system has<br />

experienced the problem that the gel is never in the right place initially. With<br />

most commercially available systems the user has to open the drawer or door<br />

and manually handle the gel for the best visualization. The ULTima, however,<br />

is the only imaging system with patented optics for image positioning without<br />

loss of optical resolution. This also ensures that lens adjustments are not<br />

required. The image is positioned with the positioning keys without having to<br />

open the cabinet or adjust a fragile gel.<br />

Slant correction tools permit optical rotation<br />

of the sample for best ‘straight’ visualization<br />

without touching the gel. Slant correction<br />

is a unique feature that allows the user to<br />

turn the image optically ± 5°. In this way<br />

the sample can be positioned so the analysis<br />

software is better able to perform lane<br />

detection.<br />

Flatfielding<br />

ULTima imaging systems allow<br />

accurate quantitative measurements<br />

of samples with a unique feature<br />

called flatfielding. Flatfielding is done<br />

in two easy steps after selecting the<br />

flatfielding option on the ULTima AQ<br />

software. Flatfielding removes<br />

artifacts resulting from small<br />

variations in the CCD and uneven<br />

background presenting a true image<br />

of sample intensities. The Orange<br />

conversion plate is used to create a<br />

flatfield image and the ULTima AQ<br />

software uses this flatfield image to<br />

“clean up” any variations to present<br />

raw image data that is highly<br />

accurate.<br />

Image Addition<br />

ULTima imaging systems allow users<br />

to simultaneously capture multiple<br />

images of the same sample at a set<br />

interval with a unique feature called<br />

Image Addition. The user can set the<br />

number of images to be ‘added’ and<br />

the time interval between<br />

acquisitions. This feature allows the<br />

ULTima to be used in kinetic studies.<br />

If the image addition is used in<br />

combination with the Auto-Save<br />

function, all the individual images will<br />

be stored. The added result should be<br />

stored manually.<br />

Templates<br />

Templates allow the user to define the<br />

settings for image acquisition,<br />

including the excitation and emission<br />

filters. The parameters are stored per<br />

specific application.<br />

Orange Conversion Plate<br />

The optional orange conversion plate<br />

allows transillumation of visible dyes<br />

like Coomassie and silver stain on a<br />

UV transilluminator. The conversion<br />

plate is also required for flatfielding<br />

the image.


How to choose<br />

your ULTima?<br />

There are four systems plus the optional Optix illumination module. All models<br />

can be upgraded to higher specified versions giving you the flexibility to<br />

purchase the model that meets your needs and budget today and upgrade it<br />

as your needs increase. All models come with ULTima AQ control and image<br />

acquisition software and ethidium bromide emission filter.<br />

ULTima 10si & 16si<br />

Use ULTima 10si for basic documentation purposes. With<br />

ULTima 16si your measurements are more sensitive and<br />

allow more accurate quantification. Both models can be<br />

used for imaging ethidium bromide stained DNA agarose<br />

gels, Coomassie stained gels, SYBR Green stained gels,<br />

SYPRO Ruby stained gels, film archiving and petri dishes.<br />

ULTima Pro<br />

Designed with innovative 2-stage Peltier cooling of the image<br />

sensor. Rather than attempt to cool the entire CCD chip to<br />

a very low temperature, the ULTima Pro cools only the areas<br />

of the chip where cooling is essential to achieve low noise<br />

levels at long measurement times. This substantially<br />

lengthens the lifetime of the CCD chip and the camera<br />

system. As with ULTima 16si Plus, the ULTima Pro is equipped<br />

with an additional computer-controlled focus and iris control.<br />

The ULTima Pro comes with a small external power supply<br />

that drives the Peltier cooling system.<br />

Optix<br />

Illumination<br />

Module<br />

The Optix module is a patented epiillumination<br />

option for ULTima Pro; for<br />

the excitation of fluorophores within<br />

the VIS/NIR range. When Optix is used<br />

for ethidium bromide stained gels, it<br />

offers the following advantages:<br />

DNA remains intact for applications<br />

downstream.<br />

Lower background means higher<br />

detection limits.<br />

Selection of excitation wavelength is<br />

fully computer controlled and, in<br />

combination with the automatic<br />

emission filter changer, true multi<br />

fluorophore capability is achieved.<br />

Multiple images with different<br />

excitation/emission filter sets allow<br />

the detection of multiple<br />

fluorophores within the same<br />

sample. The Optix can<br />

accommodate a maximum of five<br />

different excitation filter sets. The<br />

Optix module can be added to all<br />

models of the ULTima line.<br />

Applications that are possible with<br />

the addition of the Optix module<br />

include: CyDye imaging, array<br />

detection, in vivo GFP detection and<br />

GFP tissue and organ screening.<br />

Molecular Weight Calibration<br />

The TL 100 and TL 120 versions both enable precise molecular weight determination of<br />

band material from a variety of 1D gels. TL 120 contains tools which allow increased<br />

control over calibration for particularly distorted 1D gel images.<br />

Quantitation and Normalization of Band Material<br />

The TL 100 and TL 120 software solutions both contain a complete range of tools to<br />

quantify band material and for the calculation of normalized band volumes. For in-depth<br />

quantitative studies, the advanced features in TL 120 offer a higher degree of flexibility<br />

and facilitate accurate quantitation of bands on more problematic 1D gel images.<br />

Lane Relationship Studies<br />

The TL 120 software has a powerful band matching facility which is flexible yet easy to<br />

use. There are tools available to visualize the results of matching and identify similarities<br />

within a single gel. When you have performed intra gel matching, lanes can be classified<br />

and identified.<br />
