With this a single spot isapplied near a corner of the plates. After chromatographyin the first direction, the plate is dried, rotated 90° anddeveloped in the second dimension.with another mobilephase.Evaluation of the ChromatogramEvaluation can be as simple as a visual inspectionunder ultra violet light to determine the existence of acomponent. Or it could be as complicated as exposingthe plate to a chemical spray, followed by heat charringand densitometric scanning to compare the sampleto standards on the plate and achieve an accuratemeasurement of how much of a component is on theplate.Anyway you look at evaluation, the one constant is thecalculation of the retention factor (Rf value). This valueis measured as the relative distance traveled from thespot origin to the mobile phase solvent front. As longas all other variables are kept constant, the Rf value ofa sample component should remain constant and bereproducible.Inherent Advantages of TLCTLC permits the simultaneous analysis of many samplesin the same time period required for one HPLC analysis.Samples and standards are analyzed under exactly thesame conditions rather than serially as in HPLC.TLC uses a fresh, new adsorbent for each analysis. Thisinsures reproducible results and eliminates the HPLCproblems of adsorbent contamination from previousanalyses and lost efficiency from worn columns. Also,with the use of smaller developing tanks, the amount ofdeveloping solvent required is much less than with HPLC.TLC simplifies methods development. Unlike chromatographyperformed on columns, the sample components remain in theadsorbent and can always be located and retrieved for furtherexperimentation.Applications of TLCTLC is generally used for one of three purposes.1) QUALITATIVE AnalysisTo determine the presence or absence of a particularsubstance in a mixture. A rough estimation of the level of thesubstance may also be performed.2) QUANTITATIVE AnalysisTo determine, precisely and accurately, the amount of aparticular substance in a sample mix.3) PREPARATIVE AnalysisTo purify and isolate a particular substance by separating itfrom any contaminants.All three cases share the common procedures of sampleapplication, chromatographic separation, and samplecomponent visualization. The conditions under which theseprocedures are performed vary according to the required endresult. Layer Thickness<strong>Analtech</strong> UNIPLATES are 250 microns thick for regular analytical TLC and between 500 and 2000microns for preparative TLC. Tapered preparative UNIPLATES have a wedge-shaped layer 300µm to 1700µm with a700µm preadsorbent area (see notes on page 9). High Performance UNIPLATES have a thinner layer of adsorbentbetween 150-200 microns. All thickness designations are nominal. Actual thicknesses may vary but are consistent frombatch to batch.Plate SizeUNIPLATES are available in the following standard sizes: 20 x 20 cm,10 x 20 cm, 5 x 20 cm as well as 2.5 x 10 cm and 2.5x 7.5 cm. This sample applicationregion serves as a “holding zone” for sample spots untildevelopment of the plate is initiated. There is no significantresolution of sample components by the stationary phase inthis region. All soluble components migrate with the solventfront, concentrating diffuse sample spots into vertically narrowbands.Preadsorbent plate (in stages)The adsorbent zone extends from the upper edge of the preadsorbent zone and covers the remaining area of theTLC plate. This zone exhibits the selective retention of sample components characteristic of the adsorbent employed.Separation of sample components occurs in the adsorbent zone in the same way as on a standard TLC plate.Preconcentration of the sample results in compact zones at the beginning of the separation. RPS Reversed Phase PlatesThe basis of retention in Reversed Phase Chromatography is just theopposite of that seen in normal phase (e.g., silica gel) chromatography.In normal phase chromatography the adsorbent is a polar material.Adsorption occurs as a result of polar interactions. Thus, in normal phasechromatography, more polar sample components will be more tightlyadsorbed and will exhibit lower Rf values than less polar sample components.Rf values can be increased by making the mobile phase more polar.In Reversed Phase Chromatography the adsorbent is a relatively nonpolarmaterial. Adsorption occurs as a result of nonpolar interactions. Thus, inReversed Phase Chromatography less polar sample components will bemore tightly adsorbed and will exhibit lower Rf values than more polar samplecomponents. In Reversed Phase Chromatography Rf values can generally beincreased by making the mobile phase less polar.<strong>Analtech</strong> Reversed Phase Separation (RPS) UNIPLATES are a hydrocarbonimpregnated silica gel. The hydrocarbon is adsorbed onto the silica gelsurface and is not chemically anchored to the particle. The impregnated RPSUNIPLATE offers several advantages over the bonded RP18 type TLC platesalong with some limitations.The main advantages of the hydrocarbon impregnated Reversed Phase plateare compatibility with aqueous solutions and parallel behavior to most C18HPLC packings. The RPS UNIPLATE will accept 100% water either as themobile phase or spotting solvent. Therefore, RPS Reversed PhaseUNIPLATES are an excellent mediumfor rapid, inexpensive screening ofchromatographic conditions in HPLCmethod development. Mixtures of water orbuffers plus a polar organic modifier (eg.,Methanol, acetonitrile, THF, etc.) are easilyscouted with RPS UNIPLATES. However,since the hydrocarbon is adsorbed butnot chemically anchored to the silicagel, the RPS UNIPLATES cannot toleratenonpolar organic solvents. Modifiied Silica Plates<strong>Analtech</strong> manufactures a number of modified silica gelson a routine basis. Often the addition of some specificmaterial to the base adsorbent aids in discrimination ofparticular analytes. The most commonly used modifiedplates are listed here. Modification can be accomplishedin two ways. The additive material can be dissolved inthe solution used to prepare the spreading slurry, or afinished TLC plate can be impregnated with the desiredagent. If you have some particular requirement other thanthose shown, call <strong>Analtech</strong> for prices and delivery.Sodium Hydroxide (0.1N) ModifiedSilica Gel G & GF UNIPLATESInstead of water, 0.1 N sodium hydroxide is used toprepare the slurry for these plates. DEAE Avicel Cellulose UNIPLATESDiethylaminoethyl (DEAE) Cellulose carries positive charges at neutral and acidic pH. Commonly employed forseparation of negatively charged molecules by ion exchange chromatography. Hydrophilic nature of the cellulosesubstrate makes it well suited to ion exchange separations of delicate biomolecules. Plates are available withmixed layers containing both DEAE Cellulose and unmodified cellulose. High Performance PlatesNormal PhaseHigh Performance UNIPLATES provide high resolutionseparations in less than five minutes with migration distances aslittle as 5 to 7 cm. These plates are prepared with high efficiencysilica gel having an 8 micron average particle diameter andnarrow particle size distribution. The adsorbent layers are only150 microns thick, resulting in more rapid solvent migration. Theadsorbent capacity is more than adequate for small samples usedin high performance TLC techniques. Typically, sample volumesfor high performance TLC are less than one microliter. Samplespots are normally smaller than one millimeter in diameter.Samples are spotted 1.0 to 1.5 cm from the bottom edge of theplate and 0.3 to 0.5 cm apart. Most analytes are acidic,basic or neutral. The best initial approach is to use a mid-range pH, such as pH7. The standard SMT SAM-C18 and C8 columns are the best choice for use atthis pH because they provide superior column lifetime, extremely high selectivityand resolution. Acetonitrile or methanol and water are normally the first choice formobile phase.Another option is combination of organic solvent with phosphate buffer (withbuffer range pH 6.2-8.2) or acetate buffer (with buffer range pH 3.8-5.8). Methoddevelopment optimization can continue from here by changing several factors,including mobile phase, pH, column temperature (up to 95 degrees C). SMTSAM-C18 is uniquely stable to high temperatures, a characteristic that can beused as an additional tool to improve resolution.Using a low pH mobile phase results in the best peak shapes for basiccompounds because these analytes are fully protonated and exhibit low retentionand tailing. SMT SAM C18 or C8 columns are the best choice providing longestavailable lifetime and performance at low pH.Separations at high pH region may also be the most appropriate for certaincompounds. For example, it may be possible to separate bases in their freeform – where they are not charged. Here the retention increases as the chanceof obtaining the desired selectivity improves. At least oneof the ligands (e.g. C18) is functional for theseparation while the other (e.g. C1) is used asa spacer molecule, although, it too can impactcertain selectivity needed for some separation.A typical coverage achievable with SAM is 7-8ºmole/m 2 . This coverage value is equivalentto the maximum achievable coverage on anysubstrate and it is about 50% higher thanthat achievable using the most exhaustiveconventional bonding and end-cappingmethods available in the market today.49
