#65 Obesity-Induced Leptin Inhibits Streptococcus Mutans Growth But NotImmunostimul<strong>at</strong>ory CytokinesR.J. TSENG, J.T. WRIGHT, and M.A. BECKDepartments <strong>of</strong> Pedi<strong>at</strong>ric <strong>Dentistry</strong> and NutritionOBJECTIVE: Research indic<strong>at</strong>es obese children have decreased caries. Our objective was to determine if thisoccurred indirectly via leptin-induced modul<strong>at</strong>ion <strong>of</strong> cytokines important in host immune response, or directly, viainhibition <strong>of</strong> Streptococcus mutans (SM) growth. METHODS: Body mass index (BMI) was calcul<strong>at</strong>ed for 80children aged 2-5 years to determine normal (CON) or overweight/obese (OW) weight st<strong>at</strong>us. Blood samples werecollected and assayed for serum leptin, tumor necrosis factor-alpha (TNF-alpha) and interleukin-6 (IL-6)concentr<strong>at</strong>ions. Decayed or filled surfaces (dfs) score and number <strong>of</strong> carious teeth were calcul<strong>at</strong>ed. SM wascultured on agar pl<strong>at</strong>es with recombinant human leptin. Growth alter<strong>at</strong>ion was determined by calcul<strong>at</strong>ing area <strong>of</strong>zone <strong>of</strong> inhibition, and verified by spectrophotometrically measuring turbidity <strong>at</strong> 600nm. RESULTS: OW childrenhad significantly elev<strong>at</strong>ed serum leptin, fewer decayed teeth, and decreased dfs score compared to CON. TNFalphaand IL-6 expression were not significantly different in OW versus CON. Leptin inhibited growth <strong>of</strong> SM onagar pl<strong>at</strong>es in a dose-dependent fashion (0.1-10ng/ml).
