Allexperiments were conducted onduplicate <strong>column</strong>s. Solutes used forperformance measurement weredimethylphthalate and fluorene.Step One:Column stability and measurementprecision was established for each<strong>column</strong> with multiple injections over18hrs under neutral conditions (70%MeCN/water, 1mL/min). All <strong>column</strong>swere very stable. The chromatograms ofa <strong>column</strong> from each manufacturer at theend of the 18 hour period are shown onthe left hand side.Step Two:Each <strong>column</strong> was flushed with 70%MeCN/0.5N NaOH solution at 1mL/minfor one hour (23ºC). The <strong>column</strong>s werethen purged and equilibrated under thestandard neutral conditions thenretested. The center chromatograms arethe results of the first test after neutralequilibrationManufacturer P300 C18Step Three:After the second step, the neutral testconditions were maintained for one hourand a repeat injection was made. Theresults are illustrated in thechromatograms on the right hand side ofthe figure.While Higgins Analytical does notrecommend flushing any silica-basedHP<strong>LC</strong> <strong>column</strong> under such aggressiveconditions, these data show the superiorakaline stability of Proteo 300 C18.13
