B E F O R E & A F T E R S A M P L E SABSTRACTSBiotechnology & ForensicsRNA Extraction from Knee Cartilage TissueA study to determine the connection between arthritic diseasesand certain genes was conducted. Knee cartilage tissue wastaken from patients and snap frozen in liquid nitrogen for storage.Samples were ground in the large Freezer/Mill using small vialsand a multi-vial adapter. Grinding was completed in three cyclesof two minutes each. The total processing time of 12 minutesincluded precool, grinding, and cooling between grinding cycles.RNA was subsequently pur ed using a phenol-chloroformextraction method with yields of RNA between 2 & 20 µg.Polymers & Materials ResearchA P P L I C A T I O N N O T E SGrinding Polymers for Qualitative and Quantitative AnalysisCompact synthetics (pellets, webbing fabric, textiles) andpolymer yarn ce material and laments were successfullyground in the small Freezer/Milll. Cryogenic grinding wasrequired for accurate qualitative and quantitative analysis ofadditives such as antioxidants using HPLC and INOC-Test. TheFreezer/Mill iently reduced these samples to particle sizes ofless than 500 µm in less than 15 minutes without thermal degradation.Examples: : SP005 – RNA Extraction from CartilageTissue using Cryogenic Grinding: : SP006 - Preparation of Rice Husks forContent Analysis: : SP007 - Grinding Polymers for Qualitativeand Quantitative Analyses: : SP015Cetacean Samples (RNA Extraction fromWhale Skin and Blubber)Crop ResearchPreparation of Rice Husks for Content AnalysisA study to analyze the quantity of vitamins and importantminerals in rice husks was performed. 8g of sample was groundin the large Freezer/Mill. Rice husks are extremely tough and in3 grinding cycles of 3 minutes each, very good homogenizationof the rice husks was achieved. The analytical neness of thehomogenized sample enabled cient extraction of the desiredcomponents for analysis. Please visitwww.spexsampleprep.com/appnotesfor more FREEZER/MILL application notes.Customer ServiceSampleprep@spex.com15 Liberty Street , Metuchen, NJ 08840732-623-0465 • Fax 732-906-2492www.spexsampleprep.com
