Sonderforschungsbereich (SFB) 544: Control of Tropical Infectious Diseases Chairman: Hans-Georg Kräußlich, Department of Virology • • This special research program of the German Research Council (DFG) has been active since 1999; a new evaluation will be due in 2008. 4 out of 20 projects are contributed by group leaders of the BZH (Davioud-Charvet, Krauth-Siegel, Nickel, Schirmer), the other project leaders are affiliated with five university institutions and the European Molecular Biology Laboratory (EMBL). The objec- tives of the interdisciplinary SFB 544 can be sum- marized as follows: 50 Funding To understand biological mechanisms of pathogenic microorganisms (trypanosoma- tids, HIV, Plasmodium falciparum and toxo- plasma) in order to discover new targets and targeting mechanisms for drugs and vaccines as well as new vector control strategies. To understand the functions of health systems that limit the effectiveness of such control strategies and explore new ways of im- proving quality and utilization of public health systems. Schirmer and Krauth-Siegel were among the initi- ators of the SFB 544 and of the close cooperation between the SFB and the Centre de Recherche en Santé de Nouna (CRSN) in Burkina Faso. This resulted in the establishment of a biochemical laboratory for molecular parasitology at the CRSN. As a synthetic chemist with therapy-focussed concepts, Elisabeth Davioud-Charvet is a driving force for a number of projects in the SFB. Luise Krauth-Siegel´s elucidation of the trypanothione metabolism has led to new drug targeting routes in parasitic protozoa; she has been a member of the SFB´s Steering Committee since 1999. Parasitespecific protein transport in trypano somatids is the contribution of Walter Nickel. Using cell- biochemical methods, Heiner Schirmer studies safety and efficacy of affordable methylene blue- containing combination therapies against malaria and other parasitic diseases. Apart from the SFB, these studies are generously supported by the Dream Action Award of the Dutch pharmaceuti- cal company DSM. Cluster of Excellence, Cellular Networks: From molecular mechanisms to quantitative understanding of complex functions Coordinator: Hans-Georg Kräußlich, Department of Virology All cellular processes are facilitated by dynamic macromolecular complexes that interact with each other. Individual components undergo regulated conformational alterations which allow them to perform specific tasks. A cell may thus be described as a factory consisting of a dynamic network of hierarchically structured, interconnected assemblies. Enormous advances in knowledge have been achieved in the last decades including information on structures of molecules and complexes at the atomic level, composition and architecture of molecular assemblies, and on entire genomes and proteomes. A major challenge for the future is to understand complex networks both at the cellular and supracellular level. The central aim of the research cluster CellNetworks is to define behavior and dynamic alterations of complex networks and to develop a systemic understanding of their regulation and interplay. This is dependent on a closely interactive collaboration between scientists from different disciplines and also requires methodological and technological advances. Ultimately, an integrated view should be generated which is detailed enough to allow mathematical modeling and simulation of biological processes. The CellNetworks program is structured into four distinct but closely interconnected project areas according to
ascending levels of complexity, and two essential central technology platforms. Research area A investigates intracellular building blocks and their dynamic integration into macromolecular assemblies as well as their interaction within a network. Area B extends this to the higher architecture of the cell with a focus on the cytoskeleton and the mitotic spindle, and the interaction of the cell with the extracellular environment. In area C this is expanded to the supracellular level, studying signal processing and development. Research area D adds an additional level of complexity by addressing the changes of cellular networks by viral and parasite pathogens. Research groups of the BZH are an essential part of research area A of the cluster. In addition, central technology platforms of CellNetworks are established at the BZH. In particular, a crystallization platform and a facility for mass spectrometry of lipids are housed at the BZH. Furthermore, the BZH and the ZMBH jointly operate the CellNetworks facility for mass spectrometry of proteins.
