They offer increased sensitivity, down to 1 viable organism per 25 g of sample or 100 organisms per ml of pre-enrichment broth, and a significant reduction in background flora. The Dynabeads ® are 2.8 µm in diameter and covalently coated with affinity-purified antibodies against specific surface markers on the microorganisms. The method is rapid & simple to perform. It can be performed manually using Dynal Magnets & Mixers (see chapter 8) or the process can be automated using the BeadRetriever TM bench top instrument. Water Testing Dynal Biotech is the world leader in the water industry in sample preparation for Cryptosporidium and Giardia and Cryptosporidium (GC) combination testing. Dynabeads ® anti-Cryptosporidium and GC Combo are the gold standard method validated by the US Environmental Protection Agency for selective enrichment of these protozoa (US EPA Method 1622 &1623; UK DETR Water Supply Regulation 2000 SI No. 3184 England & SI No. 3911 Wales). The Dynabeads ® are covalently coated with affinity-purified antibodies against specific surface markers on the oocyst/cyst. This protocol processes a 10 ml contaminated water sample concentrate (obtained from any untreated (raw) or treated water) to produce one clean microscope slide with significantly reduced background debris. Recoveries of oocyst/cysts are regularly greater than 90%. The most recent addition to Dynal Biotech’s water testing product portfolio is Dynabeads ® anti-Legionella. This product is for rapid, selective separation of Legionella species from water samples and water concentrates.
