Microbiology
The Dynabeads ® Microbiology product range for rapid detection of bacteria
and protozoa using immunomagnetic separation has become widely accepted
and routinely used in the Food & Environmental Microbiology Laboratories
around the world. Validations have been awarded from AOAC, USEPA, AFNOR,
DETR and other government bodies worldwide.
Food Microbiology
Dynabeads ® for food microbiology remove the need for traditional media-
based selective enrichment. They offer increased sensitivity, down to 1 viable
organism per 25 g of sample or 100 organisms per ml of pre-enrichment
broth, and a significant reduction in background flora. The Dynabeads ® are
2.8 µm in diameter and covalently coated with affinity-purified antibodies
against specific surface markers on the microorganisms. The method
is rapid & simple to perform. It can be performed manually using Dynal
Magnets & Mixers (see chapter 8) or the process can be automated using
the BeadRetriever TM bench top instrument.
Water Testing
Dynal Biotech is the world leader in the water industry in sample preparation
for Cryptosporidium and Giardia and Cryptosporidium (GC) combination
testing. Dynabeads ® anti-Cryptosporidium and GC Combo are the gold
standard method validated by the US Environmental Protection Agency for
selective enrichment of these protozoa (US EPA Method 1622 &1623; UK DETR
Water Supply Regulation 2000 SI No. 3184 England & SI No. 3911 Wales). The
Dynabeads ® are covalently coated with affinity-purified antibodies against
specific surface markers on the oocyst/cyst. This protocol processes a 10 ml
contaminated water sample concentrate (obtained from any untreated (raw)
or treated water) to produce one clean microscope slide with significantly
reduced background debris. Recoveries of oocyst/cysts are regularly greater
than 90%.
The most recent addition to Dynal Biotech’s water testing product portfolio is
Dynabeads ® anti-Legionella. This product is for rapid, selective separation of
Legionella species from water samples and water concentrates. The method
can be performed manually using Dynal Magnets & Mixers (see chapter
8) or the process can be automated using the BeadRetriever TM bench top
instrument.
NEW
112
New product
Dynal MIC RELI is a new product for Dynal Microbiology. It is a molecular test
that utilises a generic PCR amplification with biotinylated primers that flank
the specific DNA target region of interest. This is followed by hybridisation
of the resulting amplicon to an array of Sequence Specific Oligonucleotide
probes immobilised on a membrane. The immobilisation of these probes
is referred to as a Reverse Line blot and thus gives rise to the acronym
“RELI”.
Under appropriate wash conditions, the probes remain bound only to their
complementary sequences in the amplified DNA. The sensitivity of the system
enables single nucleotide differences to be distinguished. The presence of
the amplicon/probe complex is detected using a simple colorimetric reaction
and appears as a blue precipitate (fig. 5, section 2).
The first product in this range is MIC RELI 4 Meningococcus (fig. 5, section
3). This kit provides a presence/absence test for Neisseria meningitidis and
identification of the common serogroups (fig. 5, section 4). The assay can
be performed manually or can be automated using the Dynal AutoRELI 48
instrument (fig. 5, section 1).
