Food <strong>Microbiology</strong><br />
Dynabeads ® for food microbiology remove the need for traditional media-<br />
based selective enrichment. They offer increased sensitivity, down to 1 viable<br />
organism per 25 g of sample or 100 organisms per ml of pre-enrichment<br />
broth, and a significant reduction in background flora. The Dynabeads ® are<br />
2.8 µm in diameter and covalently coated with affinity-purified antibodies<br />
against specific surface markers on the microorganisms. The method<br />
is rapid & simple to perform. It can be performed manually using Dynal<br />
Magnets & Mixers (see chapter 8) or the process can be automated using<br />
the BeadRetriever TM bench top instrument.<br />

Water Testing<br />
Dynal Biotech is the world leader in the water industry in sample preparation<br />
for Cryptosporidium and Giardia and Cryptosporidium (GC) combination<br />
testing. Dynabeads ® anti-Cryptosporidium and GC Combo are the gold<br />
standard method validated by the US Environmental Protection Agency for<br />
selective enrichment of these protozoa (US EPA Method 1622 &1623; UK DETR<br />
Water Supply Regulation 2000 SI No. 3184 England & SI No. 3911 Wales). The<br />
Dynabeads ® are covalently coated with affinity-purified antibodies against<br />
specific surface markers on the oocyst/cyst. This protocol processes a 10 ml<br />
contaminated water sample concentrate (obtained from any untreated (raw)<br />
or treated water) to produce one clean microscope slide with significantly<br />
reduced background debris. Recoveries of oocyst/cysts are regularly greater<br />
than 90%.<br />
The most recent addition to Dynal Biotech’s water testing product portfolio is<br />
Dynabeads ® anti-Legionella. This product is for rapid, selective separation of<br />
Legionella species from water samples and water concentrates. The method<br />
can be performed manually using Dynal Magnets & Mixers (see chapter<br />
8) or the process can be automated using the BeadRetriever TM bench top<br />
instrument.<br />

Dynal MIC RELI is a new product for Dynal <strong>Microbiology</strong>. It is a molecular test<br />
that utilises a generic PCR amplification with biotinylated primers that flank<br />
the specific DNA target region of interest. This is followed by hybridisation<br />
of the resulting amplicon to an array of Sequence Specific Oligonucleotide<br />
probes immobilised on a membrane. The immobilisation of these probes<br />
is referred to as a Reverse Line blot and thus gives rise to the acronym<br />
“RELI”.<br />
Under appropriate wash conditions, the probes remain bound only to their<br />
complementary sequences in the amplified DNA. The sensitivity of the system<br />
enables single nucleotide differences to be distinguished. The presence of<br />
the amplicon/probe complex is detected using a simple colorimetric reaction<br />
and appears as a blue precipitate (fig. 5, section 2).<br />
The first product in this range is MIC RELI 4 Meningococcus (fig. 5, section<br />
3). This kit provides a presence/absence test for Neisseria meningitidis and<br />
identification of the common serogroups (fig. 5, section 4). The assay can<br />
be performed manually or can be automated using the Dynal AutoRELI 48<br />
instrument (fig. 5, section 1).<br />
