Gerlach * * Institute for Microbiology, Department of Infectious Diseases, University of Veterinary Medicine Hannover, Germany ** Heinz Knöll Institute, Jena, Germany Actinobacillus (A.) pleuropneumoniae is the causative agent of porcine pleuropneumonia which leads to high economic losses in the swine industry worldwide. Eradication of the pathogen from a herd is exceedingly difficult because the most commonly used bacterin vaccines do not provide extensive cross-serotype protection, nor do they prevent colonization. In addition, the differentiation between infected and vaccinated animals ("DIVA" concept) is a basic prerequisite for generating and maintaining specified pathogen-free herds. Following on the observation that pigs surviving natural infection are at least partially protected from clinical symptoms upon reinfection with any serotype, we set out to construct an attenuated A. pleuropneumoniae live vaccine allowing the differentiation of vaccinated from infected animals by successively deleting virulence-associated genes. From an A. pleuropneumoniae serotype 2 prototype live negative marker vaccine strain that contains a deletion in the ApxII toxin gene (W. Tonpitak, N. Baltes, I. Hennig-Pauka, and G.-F. Gerlach, Infect. Immun. 70:7120-7125, 2002), three genes encoding enzymes involved in anaerobic respiration, as well as the ferric uptake regulator Fur were deleted, which resulted in a highly attenuated sixfold mutant. This mutant was still able to colonize the lower respiratory tract and induced an immune response which was detectable in an ELISA based on a detergent wash extract of membraneassociated proteins. Upon a single aerosol application, this mutant provided significant protection from clinical symptoms upon heterologous infection with an antigenically distinct A. pleuropneumoniae serotype 9 challenge strain, and allowed the serological discrimination between infected and vaccinated groups using an ELISA based on the ApxII toxin. Since a practical application system is necessary for a potential future use of the vaccine in the field, the protective efficacy of the 6-fold mutant upon single intramuscular or intranasal application were also tested. The vaccine was shown to elicit protective immunity against challenge with A. pleuropneumoniae serotype 7 through both application routes. While intranasal application caused no adverse effects and still resulted in a highly protective immune response upon challenge with the heterologous serotype strain, intramuscular application resulted in clinical symptoms and abscess formation. Intranasal application may prove to be a suitable route for mass vaccinations of pigs against A. pleuropneumoniae in the future.
