Cleavage <strong>of</strong> 6.36-6.45 <strong>of</strong>f the resin was accomplished with TFA <strong>and</strong> subsequent<br />

hydrolysis <strong>of</strong> the rnhpd ester 2.62 with CszCOs gave the FrPe p-hydroxy-a-amino acids<br />

6.46-6.55 (Scheme 6.13). The free amino acids cleaved <strong>of</strong>f Wang resin generaily showed<br />

punties <strong>of</strong> ~90% as indicated by 'H NMR <strong>and</strong> HPLC analysis <strong>and</strong> were observable by<br />

ESI-MS with the exception <strong>of</strong> 655 which was not isolated. Unfortunately, the amino<br />

acids cleavage <strong>of</strong>f TentaGel PHBa resin were contarninated with up to 30% PEG from<br />

the spacer on the resin. Lower concentrations <strong>of</strong> TFA decreased the amount <strong>of</strong> PEG<br />

contamination but prolonged reaction times were required for complete cleavage <strong>of</strong>f the<br />

resin. Cation exchange chromaiography was necessary for the removal <strong>of</strong> contaminating<br />

PEG <strong>and</strong> cleavage saits <strong>and</strong> is describe in section 2.4.15. No significant difference in<br />

yield or purity exists between Wang <strong>and</strong> TentaGel PHBa resin as indicated in table 6.1.
