AGAR (CCA) (01-695)<br />

Chromogenic medium CCA (Chromogenic Coli<strong>for</strong>m<br />

Agar) is a selective culture medium <strong>for</strong> the detection<br />

and enumeration of coli<strong>for</strong>ms and E. coli with an adjusted<br />

and precise selective capability. Ideal <strong>for</strong> water<br />

analysis. With antibiotics cefsulodin and vancomycin<br />

included in its <strong>for</strong>mulation.<br />

Compatible with a variety of different membrane brands<br />

using our combination of CCA and Coli<strong>for</strong>m CV selective<br />

supplement;obtaining improved results and recoveries.<br />

Formulated according to an alternative method proposed<br />

by the Spanish Authorities.<br />

Principle. Based on the detection of two enzymatic activities:<br />

ß-D-Glucuronidase (ß-gluc)<br />

ß-D-Galactosidase (ß-gal)<br />

The selectivity is attained, partially, by the Tergitol ® 7,<br />

which inhibits the growth of gram positive bacteria and<br />

some gram negative ones without negative effect on the<br />

coli<strong>for</strong>m bacteria. But the genuine selectivity is due to the<br />

presence of cefsulodin that acts against pseudomonas and<br />

other gram negative oxidase positive bacteria and also to the<br />

vancomycin, which inhibit the growth of enterococci and<br />

other gram positive bacteria. After an incubation period<br />

of 18-24 hours at 36 ± 2ºC, E. coli appears deep blue<br />

to violet, while coli<strong>for</strong>ms are salmon-rose to red. Other<br />

gram negatives show no colour.<br />

Chromogenic culture medium, selective and differential<br />

that allows <strong>for</strong> a direct identification of Escherichia coli<br />

and coli<strong>for</strong>ms in water and food samples. Can be used<br />

in the membrane filtration method.<br />

Principle. Based on the detection of two enzymatic<br />

activities:<br />

ß-D-Glucuronidase (ß-gluc)<br />

ß-D-Galactosidase (ß-gal)<br />

This <strong>for</strong>mula has a better inhibition of accompanying<br />

gram-positive flora as it uses bile salts instead of<br />

”tergitol”.<br />

After an incubation period of 24 hours at 37ºC, E. coli<br />

appears blue, while coli<strong>for</strong>ms are salmon coloured.<br />

The other Enterobacteria show no colour.<br />

COLIFORMS<br />

PRESENCE / ABSENCE BROTH<br />

Allows simple and direct detection of the presence or absence<br />

of total coli<strong>for</strong>ms and E. coli in water samples.<br />

Principle. Chromogenic and fluorogenic broth combining<br />

the ß-D-glucuronidase enzyme and ß-D-galactosidase test<br />

with the indol test in a single ready to use medium.<br />

Instructions. Add 100 ml of the sample in question to the<br />

bottle with this medium.<br />

Incubation. Incubate the sample <strong>for</strong> 18-24 hours at<br />

35-37 °C.<br />

Interpretation. Total coli<strong>for</strong>ms turn the medium a<br />

green colour. Escherichia coli emit fluorescence under UV<br />

light and test positive <strong>for</strong> indol *.<br />

➜ *For the test of indol add 2-3mL of Kovacs Reagent (6-018-100).<br />

to ISO 16649-1 and 16649-2 standards. Can be used <strong>for</strong><br />

mass seeding or in the membrane filtration method .<br />

Principle. Based on the detection of the enzyme activity:<br />

ß-D-Glucuronidase (ß-gluc).<br />

Gram-positive accompanying flora are inhibited by the use of<br />

bile salts and a high temperature of incubation.<br />

After an incubation between 18-24 hours at 44°C, the colonies<br />

of E. coli ß-glucuronidase positive appear blue-green.<br />
