równie¿ miesiêczn¹ emisjê limonenu<br />
przez hodowane w kolbach transgeniczne<br />
roœl<strong>in</strong>y <strong>tytoniu</strong> wysokoœci 12 cm. Ostatecznie<br />
autorzy tych eksperymentów stwierdzili,<br />
¿e produktywnoœæ limonenu przez tytoñ jest<br />
niska (reprezentacyjna l<strong>in</strong>ia produkowa³a<br />
143 nanog/g œwie¿ej masy liœcia). Wykazano<br />
jednak mo¿liwoœæ <strong>in</strong>¿ynierii metabolicznej<br />
monoterpenów w <strong>tytoniu</strong> i nakreœlono<br />
kierunki dalszych badañ w celu poprawienia<br />
wydajnoœci.<br />
Biotransformacje w kulturze <strong>in</strong> <strong>vitro</strong><br />
<strong>tytoniu</strong><br />
Biotransformacja w <strong>biotechnologii</strong> polega<br />
na chemicznej modyfikacji zwi¹zku obcego<br />
(egzogennnego) dla fizjologii danej<br />
komórki przez enzymy tej komórki, co jest<br />
mo¿liwe dziêki niezbyt œcis³ej specyficznoœci<br />
substratowej tych enzymów. Przyk³adem<br />
tego zjawiska z wykorzystaniem kultur komórkowych<br />
<strong>in</strong> <strong>vitro</strong> <strong>tytoniu</strong> jest biotransformacja<br />
hydroksykumaryn do <strong>ich</strong> glukozydów<br />
[6]. Stwierdzono, ¿e hydrokumaryny wywo-<br />
³ywa³y fragmentacjê genomowego DNA i<br />
wydzielanie do pod³o¿a kultury wewn¹trzkomórkowej<br />
fitoaleksyny (zwi¹zek o funkcji<br />
obronnej wydzielany na skutek czynników<br />
stresowych odzia³ywuj¹cych na roœl<strong>in</strong>ê) -<br />
skopoletyny (zwi¹zek kumarynowy). Równoczeœnie<br />
egzogenne hydroksykumaryny<br />
ulega³y transformacji do <strong>ich</strong> beta-D-glukozydów,<br />
które nie powoduj¹ fragmentacji<br />
DNA. Badania te wykaza³y, z jednej strony<br />
zdolnoœæ komórek do glukozydacji egzogennych<br />
zwi¹zków fenolowych, stwierdzonej<br />
tak¿e u <strong>in</strong>nych roœl<strong>in</strong>, jak i mechanizm reakcji<br />
obronnej <strong>tytoniu</strong> na te zwi¹zki.<br />
Podsumowanie<br />
Tytoñ w kulturach <strong>in</strong> <strong>vitro</strong> wystêpuje w<br />
postaci kalusa, organów – pêdów, korzeni,<br />
ca³ych roœl<strong>in</strong>, komórek w zawies<strong>in</strong>ie i protoplastów.<br />
Otrzymuje siê z niego l<strong>in</strong>ie haploidalne<br />
i transformowane genetycznie, mutanty<br />
i hybrydy. Przebadano na nim mo¿liwoœci<br />
biosyntezy i biotransformacji <strong>in</strong> <strong>vitro</strong>.<br />
By³ i jest jedn¹ z najwa¿niejszych roœl<strong>in</strong> eksperymentalnych<br />
w <strong>biotechnologii</strong> roœl<strong>in</strong>. Tym<br />
samym przyczyni³ siê do odkrycia zjawisk o<br />
potencjalnym zastosowaniu dla dobra ludzkoœci.<br />
Tytoñ wiêc to nie tylko u¿ywka, niew¹tpliwie<br />
dla pal¹cych przyjemna, lecz nios¹ca<br />
zagro¿enie zdrowia i ¿ycia, ale bardzo<br />
u¿yteczna, cenna roœl<strong>in</strong>a badawcza.<br />
