47.Adjust the FSC Area Scaling on Laser tab of Cytometer – <strong>FACSAria</strong>II,<br />
until the mean of FSC-A and FSC-H are APPROXIMATELY SAME.<br />
Return to Parameters tab and finely re-adjust voltage of FSC and SSC.<br />
Population of interest should be shown in the center of FSC v SSC dot plot.<br />
48.Adjust voltage of each of the channels; hence their signal peaks sit next<br />
to the Y-axis of the histograms, with majority of the peak from the<br />
population of interest greater than ZERO but less than 10 2 .<br />
49.Press ‘Unload’ on Acquisition Dashboard to unload<br />
the current sample tube.<br />
50.Repeat step 17 TO step 20 with the POSITIVE CONTROL sample tubes.<br />
Adjust the voltage of corresponding channels if their signal peaks are outside<br />
the limit of the histograms.<br />
51.Set the current tube pointer to Unstained / Negative Control tube<br />
