Amoeba saline: Five stock solutions are prepared as<br />

Make final medium by adding 10 ml of each stock<br />

solution to 950 ml of.glass distilled water.<br />

methane (NH2.C.(CH2OH)3) 0.5 g<br />

- Supplied by Water Life <strong>Research</strong> Industries.<br />

Biotin 0.2 mg<br />

Calcium pantothenate 20.0 mg<br />

Cyanocobalamin 4.0 mg<br />

Folic acid 0.4 mg<br />

Nicotinic acid 20.0 mg<br />

Thiamine 100.0 mg<br />

Thymine 600.0 mg<br />

(Vitamin solution may be stored frozen at -20°C)<br />

Preparation: 50 ml of soil extract is added to<br />

950 ml of stock solution (i) and 3.75 ml of stock<br />

solution (ii), then the pH is adjusted to 7.6 - 7.8<br />

using 1N HC1. It is then filtered, dispensed into<br />

appropriate containers and autoclaved. Vitamin<br />

solution (iii) is added after sterilization through<br />

a 0.22 micrometre filter, 0.25% by volume.<br />

Many marine algae grow in Erdschreiber medium:<br />

Medium M12<br />

<strong>Natural</strong> seawater (or artificial<br />

sea salts, see Medium M11) 1 litre<br />
