<strong>RNA</strong> Transfection: The Broad Spectrum TransIT-m<strong>RNA</strong> Kit<br />

The transfection of <strong>RNA</strong> molecules into mammalian cells can be used for a variety of experimental<br />

purposes such as rapid, short-term protein expression after m<strong>RNA</strong> transfection or production<br />

of infectious <strong>RNA</strong> virus after transfection of the viral genomic <strong>RNA</strong>. Both applications require a<br />

transfection reagent that delivers <strong>RNA</strong> molecules with high efficiency and low cellular toxicity.<br />

Kit Efficiently Delivers m<strong>RNA</strong> to a<br />

Variety of Cell Lines. The indicated<br />

cell lines were transfected with<br />

0.5 µg of a capped, polyadenylated<br />

in vitro transcript encoding EGFP<br />

using the TransIT-m<strong>RNA</strong> Transfection<br />

Kit. Eighteen hours post-transfection,<br />

the cells were analyzed by<br />

flow cytometry to identify the EGFP<br />

expressing cells.<br />

Nontransfected lacZ m<strong>RNA</strong> Transfected<br />

The TransIT-m<strong>RNA</strong> Transfection Kit Efficiently Delivers lacZ m<strong>RNA</strong> to<br />

CHO-K1 Cells. Using the TransIT-m<strong>RNA</strong> Transfection Kit, CHO-K1 cells<br />

were mock transfected (A) or transfected with a capped and polyadenylated<br />

lacZ encoding m<strong>RNA</strong> (B). Approximately 18 hrs post-transfection the<br />

cells were stained using Mirus Bio’s Beta-gal Staining Kit to identify the<br />

lacZ expressing cells.<br />
