<strong>3D</strong> CoSeedis goes 24!: New 24 well plate<br />

<strong>3D</strong> CoSeedis from our partner abc biopply is a novel scaffold-free <strong>3D</strong><br />

cell co-culture system. It c<strong>on</strong>sists of a unique agarose-based matrix c<strong>on</strong>taining<br />

an array of c<strong>on</strong>ical microwells. The particular and proprietary topography<br />

of this arrangement allows the formati<strong>on</strong> of spheroidal and n<strong>on</strong>spheroidal<br />

cell aggregates in a highly-reproducible and c<strong>on</strong>sistent manner.<br />

The c<strong>on</strong>ically shaped microwells <str<strong>on</strong>g>with</str<strong>on</strong>g>in the array allow precise determinati<strong>on</strong><br />

of aggregate volume, hence cell growth. The modular compositi<strong>on</strong> allows<br />

co-culturing of feeder cells that are physically separated from the actual<br />

test cells (distance co-culture). C<strong>on</strong>sequently, this set-up counts<br />

resp<strong>on</strong>sible for the standardisati<strong>on</strong> of protocols under defined c<strong>on</strong>diti<strong>on</strong>s.<br />

The system is now as a 24-well plate<br />

available. The advantage: You <strong>on</strong>ly<br />

need 1–2 ml of media per well (as<br />

compared to 4 ml for the 6-well plate).<br />

And even for seeding it requires<br />

1.9 ml of media as compared to 9.5<br />

ml for the 6-well plate). It has slightly<br />

smaller dimensi<strong>on</strong>s of micro-wells in<br />

a more densely packed grid (0.5 mm<br />

distance from microwell to microwell) and is so ideally suited to induce cell<br />

HemEx-TypeA is a serum-free media, which c<strong>on</strong>tains Polyvinyl Alcohol (PVA)<br />

and can be used for mouse Hematopoietic Stem Cells (HSCs).<br />

In general, culture media for HSCs c<strong>on</strong>tains serum albumin. Such albumin is<br />

required to induce cell divisi<strong>on</strong> of HSCs. On the other hand, albumin inhibits<br />

stable undifferentiated state.<br />

Widely used purified or recombinant albumin c<strong>on</strong>tains small amount of c<strong>on</strong>taminants,<br />

but are tough to remove.<br />

It is reported that such c<strong>on</strong>taminants induce differentiati<strong>on</strong> of HSCs. Moreover,<br />

protein's oxidati<strong>on</strong> reacti<strong>on</strong> induces cellular senescence. Thefore, albumin<br />

needs to be replaced by other chemicals.<br />

Dr. Satoshi Yamazaki at The Institute of Medical Science, The University of Tokyo<br />

reported that PVA can replace albumin (1). Moreover, PVA is a stable chemical<br />

<str<strong>on</strong>g>with</str<strong>on</strong>g> no oxidati<strong>on</strong> reacti<strong>on</strong> in medium so aging of HSCs are also suppressed.<br />

These advantages help to do expansi<strong>on</strong> culture of HSCs while maintaining<br />

undifferentiated state.<br />

CFC assays are well characterized and validated in vitro functi<strong>on</strong>al assays that<br />

can detect an increase or decrease in the frequency of hematopoietic progenitor<br />

LipiDye: Lipid Droplet Green<br />

Lipid droplets(LDs) are organelles which store neutral lipids such astriglycerides<br />

and steryl esters. LDs are frequently observed in adipose tissue and<br />

are c<strong>on</strong>sidered as sites for energy storage or lipid turnover. Recent studies<br />

discovered that LDs are not <strong>on</strong>ly in adipocytes, but also in ubiquitous cells<br />

such as skeletal muscle cells, macrophages, and glia cells.<br />

LDs in adipocytes are about >10 µm and can be detected by using commercial<br />

probes including Nile red. On the other hand, LDs in those n<strong>on</strong>adipocytes<br />

are too small (~0.1 µm) to detect by these probes because of<br />

high background and low specificity.<br />

Sensitive and specific probes are desired to detect small LDs. LipiDye is a<br />

novel green fluorescent probe for specific detecti<strong>on</strong> of LDs (ref.1). LipiDye<br />

shows high sensitivity, low<br />

cell toxicity and high photostability<br />

compared <str<strong>on</strong>g>with</str<strong>on</strong>g><br />
