PELONews <strong>Spring</strong> <strong>2024</strong> Overview MSC-EVs Reprinted from “Extracellular Vesicle Separation by Density Gradient Ultracentrifugation”, by BioRender.com The culture and expansion of Mesenchymal Stem/ Stromal Cells (MSCs) play a crucial role in exosome production, with the choice of expansion method significantly affecting the yield and quality of exosomes. Researchers have been investing immortalized MSC (iMSC) lines, created by inducing the expression of human telomerase reverse transcriptase (hTERT), as a potential solution to the senescence issue encountered during primary MSC culture expansion. While iMSC lines offer a reliable and scalable source for exosome production, concerns exist regarding their functional alterations compared to primary MSCs. Also, the selection of culture media and serum for MSCs culture and expansion can also affect the exosome quality and yield. The choice of isolation technique is crucial as it impacts exosome purity and yield for therapeutic applications. Techniques like ultrafiltration, density gradient purification, size-based isolation, polymer precipitation, and immuno-affinity purification have been developed for exosome purification, with tangential flow filtration being recommended for industrial-scale manufacturing due to its efficiency in concentrating extracellular vesicles from large fluid volumes, providing with diverse options for efficient exosome extraction in various biomedical applications.
