3.5.4 Root<strong>in</strong>g<br />
The micro shoots were taken out <strong>and</strong> placed back <strong>in</strong> the tubes conta<strong>in</strong><strong>in</strong>g media with<br />
different concentrations of IBA <strong>and</strong> NAA for root<strong>in</strong>g.<br />
3.5.5 Ex vitro root<strong>in</strong>g <strong>and</strong> Harden<strong>in</strong>g<br />
Microshoots were taken out from the testtube, base of the shoot was moistened,<br />
treated with powder form IBA of different concentrations <strong>and</strong> planted <strong>in</strong> small pots conta<strong>in</strong><strong>in</strong>g<br />
different harden<strong>in</strong>g media. These plants were ma<strong>in</strong>ta<strong>in</strong>ed <strong>in</strong> polyhouse. They were watered<br />
once <strong>in</strong> two days <strong>in</strong>itially, then once <strong>in</strong> a week. Later they were transferred to green house<br />
after 15 days for further acclimatization.<br />
3.6 Experimental details<br />
3.6.1 Experiment 1 - St<strong>and</strong>ardization of surface sterilization<br />
In this experiment, the efficacy of HgCl 2 with a treatment of a fungicide <strong>and</strong> a<br />
bactericide for dis<strong>in</strong>fect<strong>in</strong>g the explants was studied. Fungicide (carbendazim) <strong>and</strong> bactericide<br />
(streptomyc<strong>in</strong>) were used for pretreatment <strong>in</strong> four treatments. Experiment was undertaken for<br />
<strong>dracaena</strong> <strong>and</strong> cordyl<strong>in</strong>e.<br />

T1 - 0.05% HgCl2 for 5 m<strong>in</strong>utes<br />
T2 - 0.1 % HgCl2 for 5 m<strong>in</strong>utes<br />

a. Percent contam<strong>in</strong>ation: Out of total tubes <strong>in</strong>oculated, the Number of tubes contam<strong>in</strong>ated<br />
were counted <strong>and</strong> expressed <strong>in</strong> percentage.<br />
b. Per cent survival of explants: The number of plants survived out of total <strong>in</strong>oculated was<br />
counted <strong>and</strong> converted <strong>in</strong> to percentage.
