3.6.4 Experiment 4: St<strong>and</strong>ardization ex vitro root<strong>in</strong>g <strong>and</strong> harden<strong>in</strong>g media<br />
Young shooted plantlets were taken out of the bottles, washed with distilled water,<br />
treated with PGR powder <strong>and</strong> planted <strong>in</strong> pots conta<strong>in</strong><strong>in</strong>g different harden<strong>in</strong>g media <strong>and</strong><br />
ma<strong>in</strong>ta<strong>in</strong>ed <strong>in</strong> a polyhouse. Required quantity of IBA was taken, dissolved <strong>in</strong> alcohol <strong>and</strong><br />
mixed with talc powder. The media used for harden<strong>in</strong>g were peat <strong>and</strong> pott<strong>in</strong>g mixture.<br />
Enriched coir pith peat was used after sterilization. The pott<strong>in</strong>g mixture media was prepared<br />
by mix<strong>in</strong>g the sieved soil, farmyard manure <strong>and</strong> s<strong>and</strong> <strong>in</strong> 1: 1: 1 ratio after sterilization.<br />
