Hybridisation<br />
• I due strand di <strong>DNA</strong> sono complementari l’uno all’altro.<br />
• The mRNA synthesized from a gene is identical to one strand of the<br />
<strong>DNA</strong> and complementary to the other.<br />
• Complementary strands could be separated (denaturation) and when<br />
allowed to rejoin (renaturation) do so with high degree of specificity<br />
forming <strong>DNA</strong>-<strong>DNA</strong>, <strong>DNA</strong>-RNA, or RNA-RNA hybrids.<br />
Methods developed on these properties:<br />
• Southern blot: Hybridization of <strong>DNA</strong> to <strong>DNA</strong> (solid/solution).<br />
• Northern blot: Hybridization of <strong>DNA</strong> to RNA (solid/solution).<br />
• RNAse protection assay: Hybridization of RNA to RNA (solution).<br />
• In situ hybridisation: <strong>DNA</strong>-<strong>DNA</strong> or <strong>DNA</strong>-RNA (tissue section, cells)
