Oncolyn Neutralizes the Oxidative Damage by Asbestos<br />

<strong>in</strong> vitro, <strong>in</strong> vivo, <strong>and</strong> <strong>in</strong> Patients<br />

Asbestos can cause pulmonary <strong>in</strong>jury, as well as fibrosis <strong>and</strong><br />

mesothelioma; <strong>and</strong> that alveolar macrophages play an important<br />

role <strong>in</strong> these processes. The disorder <strong>of</strong> oxidation <strong>and</strong> antioxidation<br />

may contribute to the process <strong>of</strong> fibrosis <strong>and</strong> mesothelioma caused<br />

by asbestos. Oncolyn® is a formulated extract from three edible<br />

plants conta<strong>in</strong><strong>in</strong>g polyphenols, proanthocyanid<strong>in</strong>s, plant sapon<strong>in</strong>s<br />

<strong>and</strong> other natural plant <strong>in</strong>gredients <strong>in</strong> synergistic comb<strong>in</strong>ation with<br />

effective anti<strong>oxidants</strong> <strong>and</strong> cytoprotective function. The current<br />

study <strong>in</strong>vestigated the effect <strong>of</strong> Oncolyn on the neutralization <strong>of</strong><br />

oxidative damage <strong>in</strong> alveolar macrophages, both <strong>in</strong> vitro <strong>and</strong> <strong>in</strong> rats<br />

<strong>and</strong> used <strong>in</strong> patients with confirmed Mesothelioma. The DNA<br />

str<strong>and</strong> breakage, nitric oxide (NO) generation <strong>and</strong>, superoxide dismutase<br />

(SOD) activity, glutathione peroxidase (GSH-Px) activity,<br />

<strong>and</strong> NO synthesize enzyme were measured <strong>in</strong> alveolar macrophages<br />

treated with asbestos <strong>and</strong> Oncolyn.<br />

In blood <strong>and</strong> lung tissue <strong>of</strong> Wistar rats exposed to asbestos, the<br />

<strong>in</strong>teraction between <strong>in</strong>haled asbestos <strong>and</strong> membranes <strong>of</strong> alveolar<br />

macrophage (AM) is an important event <strong>in</strong> the development <strong>of</strong> fibrosis.<br />

Asbestos can <strong>in</strong>jure alveolar macrophages directly, <strong>and</strong> can<br />

also <strong>in</strong>duce them to release a variety <strong>of</strong> mediators which play a key<br />

role <strong>in</strong> the <strong>in</strong>itial lung <strong>in</strong>jury. Alveolar macrophages obta<strong>in</strong>ed from<br />

Wistar rats cultured with asbestos <strong>and</strong> Oncolyn for one or two<br />

hours. Results from analysis by Comet assay demonstrated that the<br />

percent <strong>of</strong> Comet cells decreased, <strong>and</strong> the length <strong>of</strong> DNA migration<br />

became shorter, <strong>in</strong> alveolar macrophages treated with Oncolyn as<br />

compared to alveolar macrophages receiv<strong>in</strong>g no Oncolyn. Oncolyn<br />

also <strong>in</strong>hibited nitric oxide generation, as measured by the amount <strong>of</strong><br />

nitrite released <strong>in</strong>to the culture medium. NO synthesize enzyme <strong>in</strong><br />

soluble extracts <strong>of</strong> alveolar macrophages treated with Oncolyn for 2<br />
