We have previously shown that palm vitam<strong>in</strong> E accelerates excision<br />

<strong>and</strong> <strong>in</strong>cision wound closures <strong>in</strong> diabetic rats when supplemented<br />

at a high dose. In order to establish that palm vitam<strong>in</strong> E improves<br />

the rates <strong>of</strong> wound heal<strong>in</strong>g, we exam<strong>in</strong>ed the histological<br />

changes that occur dur<strong>in</strong>g the process <strong>of</strong> heal<strong>in</strong>g <strong>in</strong> supplemented<br />

<strong>and</strong> unsupplemented diabetic rats.<br />

Diabetes mellitus was <strong>in</strong>duced us<strong>in</strong>g streptozotoc<strong>in</strong> (50 mg/kg<br />

bw) <strong>in</strong>tramuscularly <strong>in</strong> 24 fast<strong>in</strong>g male Sprague Dawley rats. Experiment<br />

was started by creat<strong>in</strong>g four 6mm diameter wounds on the<br />

back <strong>of</strong> each animal under sterile conditions us<strong>in</strong>g a punch biopsy.<br />

The rats were then supplemented orally with either palm vitam<strong>in</strong>e<br />

E at doses <strong>of</strong> 5, 50 or 200 mg/kg bw or equivalent volume <strong>of</strong> olive<br />

oil daily. Wounds were collected ten days after the <strong>in</strong>itial <strong>in</strong>jury,<br />

cross sections were cut <strong>and</strong> sta<strong>in</strong>ed with Haematoxyl<strong>in</strong>’s & Eos<strong>in</strong><br />

sta<strong>in</strong> <strong>and</strong> Van Giesons sta<strong>in</strong>s.<br />

Results showed that rats supplemented with 200 mg/kg bw PV<br />

had the most proliferation <strong>of</strong> cells <strong>and</strong> growth <strong>of</strong> collagen fibres <strong>in</strong>dicat<strong>in</strong>g<br />

improved heal<strong>in</strong>g compared to the other groups.<br />
