These marks are awarded for two strands of skills:<br />
• Making decisions about measurements or observations<br />
• Successfully collecting data and observations.<br />
Making decisions about measurements or observations<br />
Within an investigation, candidates should use the skills, knowledge and understanding of the AS Level<br />
Biology <strong>sy</strong>llabus to:<br />
• Identify the independent and dependent variable<br />
• Decide how the independent variable should be changed within a suitable range to provide accurate<br />
results. Candidates should:<br />
– decide the range to use for an independent variable and decide how to change its values, including:<br />
○ concentration (using simple or serial dilution)<br />
○ temperature (selected from above freezing to 100 °C)<br />
○ pH (using buffers)<br />
○ moving air (using a fan)<br />
○ humidity (using a plastic bag or calcium hydroxide)<br />
– decide the number of values at which measurements are recorded (a minimum of 5 measurements,<br />
replicates or more measurements around a specific value)<br />
– decide on the spacing of the values, for example, if more measurements are needed around a<br />
specific variable<br />
– decide intervals including concentration, temperature or pH<br />
– decide the number of replicates at each value<br />
– decide how to identify the presence of a biological molecule and estimate its quantity by<br />
standardising the appropriate test<br />
• Describe an appropriate control which removes the effect of the independent variable (including<br />
replacing a solution with the same volume of water or denaturing an enzyme by boiling)<br />
• Decide which variables to standardise because they may change the results if the variable changes<br />
during the investigation and decide how to standardise each variable to provide accurate results. These<br />
include:<br />
– volume (must be suitable for apparatus)<br />
– concentration<br />
– temperature<br />
– pH<br />
– biological material (for example same species; age; storage conditions; time of year; mass)<br />
– humidity (using a plastic bag or calcium hydroxide)<br />
– apparatus<br />
