Abstract:<br />

The goal of This study was the determination of genetic variation in COI-COII intergenic region of<br />

mtDNA in different Iranian honeybee populations by using RFLP-PCR. DNA extraction was carried<br />

out from 200 worker honey bees, collected from different locations of Iran, and COI-COII intergenic<br />

region of the samples were amplified using a universal primer pair. Based on the results of digestion of<br />

the amplicons by DraI, the samples were separated in two different digestion profiles. The first group<br />

with 4 restriction fragments 41, 47, 64 and 420 bp was distinguished from the second group with<br />

having 39, 47, 64 and 420 bp bands. According to DraI restriction analysis, Iranian honeybees were<br />

grouped as a member of C mtDNA lineage.<br />
