III. Sample Extraction/DNA Isolation
Diagenode suggests some appropriate extraction methods:
Nuclisens easyMAG ® System (Biomerieux)
Clinical samples: bronchoalveolar lavage, sputum.
Follow manufacturer’s instructions with the following:
Human sample extracted volume: 200 µl (+ 10 μl of the IC DNA virus culture)
DNA isolation volume: 60 µl
MagNa pure LC system, Roche: MagNa pure LC Total Nucleic Acid Isolation Kit (Ref.:
03038505001)
Clinical samples: bronchoalveolar lavage, sputum.
Follow manufacturer’s instructions with the following:
Human sample extracted volume: 200 µl (+ 10 μl of the IC DNA virus culture)
DNA isolation volume: 60 µl
QIAamp DNA Mini Kit (50) (Blood and Body Fluid Spin Protocol) (Qiagen Ref. 51304)
Clinical samples: bronchoalveolar lavage, sputum.
Follow manufacturer’s instructions with the following:
Human sample extracted volume: 200 µl (+ 10 μl of the IC DNA virus culture)
DNA isolation volume: 60 µl
General extraction information:
If you perform the PCR on the same day, the DNA extraction can be conserved on ice. For a
longer conservation period, DNA must be stored at -20°C.
