RIVM Report 7037190641 Introduction<strong>Drugs</strong> <strong>of</strong> <strong>abuse</strong> (DOA) <strong>and</strong> their degradation products have recently beenrecognised as environmental emerg<strong>in</strong>g contam<strong>in</strong>ants. They are among thegrow<strong>in</strong>g number <strong>of</strong> compounds that is detected <strong>in</strong> the water environment, whichis related to the <strong>in</strong>creas<strong>in</strong>g sensitivity <strong>of</strong> analytical methods. DOA refers to bothillegal drugs <strong>and</strong> the misuse <strong>of</strong> prescription drugs such as <strong>tranquilizers</strong>. DOAhave received <strong>in</strong>creased <strong>in</strong>terest s<strong>in</strong>ce Jones-Lepp et al. (2004) first reportedtheir occurrence <strong>in</strong> treated sewage effluents <strong>in</strong> the US. Follow<strong>in</strong>g consumption,DOA <strong>and</strong> their metabolites are cont<strong>in</strong>uously released <strong>in</strong>to the aquaticenvironment due to their partial elim<strong>in</strong>ation <strong>in</strong> sewage treatment plants (STPs).Recent studies have shown the occurrence <strong>of</strong> DOA <strong>and</strong> their metabolites <strong>in</strong> STPs<strong>and</strong> river water <strong>in</strong> the US (V<strong>and</strong>erford <strong>and</strong> Snyder, 2006; Bartelt-Hunt et al.,2009) <strong>and</strong> <strong>in</strong> European countries like Italy <strong>and</strong> Switzerl<strong>and</strong> (Zuccato et al.,2005; Zuccato et al., 2008; Castiglioni et al., 2006), Spa<strong>in</strong> (Boleda et al., 2007;Huerta-Fontela et al., 2007; Postigo et al., 2008; Bijlsma et al., 2009; Huerta-Fontela et al., 2008), United K<strong>in</strong>gdom (Kasprzyk-Hordern et al., 2007), Irel<strong>and</strong>(Bones et al., 2007), Germany (Hummel et al., 2006) <strong>and</strong> Belgium (Nuijs et al.,2009). Possible ecotoxicological <strong>and</strong> human toxicological effects <strong>of</strong> theirpresence <strong>in</strong> the aquatic environment have not been <strong>in</strong>vestigated so far.Besides the objective <strong>of</strong> monitor<strong>in</strong>g their environmental occurrence, severalauthors have developed analytical methodologies to determ<strong>in</strong>e DOA <strong>and</strong> theirmetabolites <strong>in</strong> water matrices with the objective to estimate collective drugconsumption at the community level (Daughton <strong>and</strong> Jones-Lepp, 2001; Zuccatoet al., 2005; Zuccato et al., 2008). Accord<strong>in</strong>g to Nuijs et al. (2009; 2009b)wastewater analysis is a promis<strong>in</strong>g tool to evaluate coca<strong>in</strong>e consumption at bothlocal <strong>and</strong> national scales.In the Netherl<strong>and</strong>s, little is known about the occurrence <strong>of</strong> DOA <strong>and</strong> theirdegradation products <strong>in</strong> the water environment. An exploratory study on theoccurrence <strong>of</strong> DOA <strong>in</strong> <strong>Dutch</strong> <strong>surface</strong> <strong>waters</strong> <strong>and</strong> STP effluents was conducted <strong>in</strong>2006–2007 by the KWR Watercycle Research Institute (Kiwa Water Research atthat time). At one STP effluent <strong>and</strong> four <strong>surface</strong> water sampl<strong>in</strong>g locations, atleast 4 out <strong>of</strong> the 14 DOA <strong>in</strong>vestigated were detected (Deltalab 2007;Hogenboom et al., 2009; De Voogt et al., <strong>in</strong> press). These <strong>in</strong>cluded opioids,benzoylecgon<strong>in</strong>e (human metabolite <strong>of</strong> coca<strong>in</strong>e), methadone <strong>and</strong>two <strong>tranquilizers</strong>, nordazepam <strong>and</strong> oxazepam. However, concentration levelscould not be calculated s<strong>in</strong>ce at that time no license to order, store <strong>and</strong> analysethese types <strong>of</strong> drugs was available.The <strong>Dutch</strong> M<strong>in</strong>istry for Hous<strong>in</strong>g, Spatial Plann<strong>in</strong>g <strong>and</strong> the Environment (VROM)asked the National Institute <strong>of</strong> Public Health <strong>and</strong> the Environment (RIVM), toperform a screen<strong>in</strong>g monitor<strong>in</strong>g <strong>in</strong> the Netherl<strong>and</strong>s. This screen<strong>in</strong>g was carriedout <strong>in</strong> close cooperation with the jo<strong>in</strong>t research programme (BTO) <strong>of</strong> the <strong>Dutch</strong>water companies, executed by KWR Watercycle Research Institute. The focus <strong>of</strong><strong>in</strong>terest <strong>in</strong> this first screen<strong>in</strong>g monitor<strong>in</strong>g campaign is the question <strong>of</strong> whetherDOA are present <strong>in</strong> <strong>Dutch</strong> <strong>surface</strong> <strong>waters</strong> <strong>and</strong> the dr<strong>in</strong>k<strong>in</strong>g water that isproduced from itThe ma<strong>in</strong> objectives pursued with<strong>in</strong> this study were:1. to evaluate the occurrence <strong>of</strong> DOA <strong>and</strong> metabolites residue <strong>in</strong> <strong>Dutch</strong><strong>surface</strong> <strong>waters</strong> that are important resources for dr<strong>in</strong>k<strong>in</strong>g waterproductionPage 15 <strong>of</strong> 90
