<strong>葉</strong> <strong>綠</strong> <strong>素</strong> <strong>含</strong> <strong>量</strong> <strong>測</strong> <strong>定</strong> <strong>之</strong> <strong>應</strong> <strong>用</strong> 43討論Fanizza 等 人 以 <strong>葉</strong> <strong>綠</strong> <strong>素</strong> 計 (SPAD-502) <strong>測</strong> <strong>量</strong> 葡 萄 <strong>葉</strong> <strong>片</strong> <strong>在</strong> 乾 旱 逆 境 處 理 後 <strong>之</strong> 讀 值 , 與 溶 劑 萃取 所 得 <strong>之</strong> <strong>葉</strong> <strong>綠</strong> <strong>素</strong> <strong>含</strong> <strong>量</strong> 呈 線 性 相 關 (3) , 因 而 認 為 <strong>葉</strong> <strong>綠</strong> <strong>素</strong> 計 可 作 為 葡 萄 品 <strong>種</strong> 耐 旱 性 選 拔 <strong>之</strong> 工 具 。顯 示 非 破 壞 性 的 <strong>葉</strong> <strong>綠</strong> <strong>素</strong> <strong>速</strong> <strong>測</strong> <strong>法</strong> 適 <strong>用</strong> 於 估 算 植 物 <strong>葉</strong> <strong>片</strong> <strong>之</strong> <strong>葉</strong> <strong>綠</strong> <strong>素</strong> <strong>含</strong> <strong>量</strong> 。 而 Marquard 及 Tipton 以 二部 同 型 <strong>之</strong> 可 攜 帶 式 <strong>葉</strong> <strong>綠</strong> <strong>素</strong> 計 (SPAD-501) <strong>測</strong> 相 同 <strong>之</strong> 材 料 , 所 得 讀 值 並 不 相 同 。 <strong>在</strong> 12 <strong>種</strong> 材 料 與<strong>用</strong> DMF 萃 取 30 小 時 後 與 二 部 <strong>葉</strong> <strong>綠</strong> <strong>素</strong> 計 讀 值 <strong>之</strong> 比 較 , 其 r 2 (linear relationship) 介 於 0.827~0.969<strong>之</strong> 間 (7) , 由 此 可 知 , 進 行 非 破 壞 性 <strong>葉</strong> <strong>綠</strong> <strong>素</strong> <strong>速</strong> <strong>測</strong> <strong>法</strong> <strong>應</strong> 考 <strong>量</strong> 作 物 及 儀 器 所 產 生 <strong>之</strong> 誤 差 。本 試 驗 三 <strong>種</strong> 材 料 不 同 生 育 階 段 各 20 <strong>片</strong> <strong>葉</strong> <strong>片</strong> , 以 二 <strong>種</strong> 方 <strong>法</strong> 所 得 <strong>之</strong> 值 與 丙 酮 萃 取 <strong>量</strong> 比 較 分析 , 除 荔 枝 成 熟 <strong>葉</strong> 外 , 其 他 <strong>之</strong> 比 較 決 <strong>定</strong> 係 數 都 相 當 低 ( 表 一 ), 顯 然 利 <strong>用</strong> <strong>速</strong> <strong>測</strong> <strong>法</strong> <strong>應</strong> 以 不 同 生 長階 段 <strong>之</strong> <strong>葉</strong> <strong>片</strong> 以 得 到 較 正 確 的 檢 <strong>量</strong> 線 (calibration curve), 否 則 可 能 影 響 所 換 算 <strong>之</strong> <strong>葉</strong> <strong>綠</strong> <strong>素</strong> <strong>含</strong> <strong>量</strong> <strong>之</strong>值 。Monje 及 Bugbee (6) (3)以 及 Fanizza 等 人 認 為 <strong>葉</strong> 脈 或 <strong>葉</strong> <strong>綠</strong> <strong>素</strong> 分 佈 不 平 均 , 都 會 影 響 以 光 學 儀器 作 非 破 壞 性 <strong>葉</strong> <strong>綠</strong> <strong>素</strong> <strong>速</strong> <strong>測</strong> <strong>法</strong> <strong>之</strong> 準 確 性 , 雖 然 本 試 驗 <strong>測</strong> <strong>量</strong> 時 已 避 開 <strong>葉</strong> <strong>片</strong> 主 要 <strong>葉</strong> 脈 , 但 可 能 嫩<strong>葉</strong> 因 <strong>葉</strong> <strong>綠</strong> <strong>素</strong> 分 佈 較 不 均 勻 , 因 而 受 到 的 影 響 較 大 , 以 致 <strong>在</strong> 嫩 <strong>葉</strong> <strong>速</strong> <strong>測</strong> <strong>法</strong> 與 丙 酮 萃 取 <strong>量</strong> 間 決 <strong>定</strong>係 數 較 低 ( 表 一 )。比 較 色 差 儀 與 <strong>葉</strong> <strong>綠</strong> <strong>素</strong> 計 <strong>之</strong> 讀 值 , <strong>葉</strong> <strong>綠</strong> <strong>素</strong> 計 <strong>在</strong> 三 <strong>種</strong> <strong>果</strong> <strong>樹</strong> 所 得 <strong>之</strong> 讀 值 與 丙 酮 萃 取 <strong>量</strong> 分 析 結<strong>果</strong> 決 <strong>定</strong> 係 數 均 較 色 差 儀 高 , 而 色 差 儀 <strong>在</strong> 芒 <strong>果</strong> 及 荔 枝 與 丙 酮 研 磨 萃 取 <strong>之</strong> 比 較 分 別 達 0.88 與0.93, 但 梨 僅 有 0.75。 探 討 二 <strong>種</strong> 方 <strong>法</strong> 讀 值 與 丙 酮 萃 取 <strong>量</strong> 間 決 <strong>定</strong> 係 數 差 異 <strong>之</strong> 原 因 , <strong>葉</strong> <strong>綠</strong> <strong>素</strong> 計 係以 紅 光 (650nm) 穿 透 <strong>葉</strong> <strong>片</strong> (8) , 除 色 <strong>素</strong> 分 佈 不 平 均 或 光 線 穿 透 性 影 響 準 確 度 外 (3,6) , <strong>葉</strong> <strong>片</strong> 厚 度 <strong>應</strong>不 超 過 1.2mm (8) , 由 圖 二 、 圖 四 及 圖 六 上 拋 的 二 次 函 數 線 性 模 式 , 顯 然 隨 芒 <strong>果</strong> 、 荔 枝 及 梨 <strong>葉</strong><strong>片</strong> <strong>葉</strong> <strong>綠</strong> <strong>素</strong> <strong>含</strong> <strong>量</strong> 增 加 時 ( 可 能 <strong>葉</strong> <strong>片</strong> 變 厚 或 組 織 間 <strong>葉</strong> <strong>綠</strong> <strong>素</strong> 累 積 ), 影 響 紅 光 <strong>之</strong> 穿 透 性 , 因 而 影 響 讀值 。 <strong>在</strong> Fanizza 等 人 (3) 、Marquard 及 Tipton (7) (12)、Sibley 等 人 與 Yadava (15) 等 利 <strong>用</strong> <strong>葉</strong> <strong>綠</strong> <strong>素</strong> 計 所 進行 <strong>之</strong> 試 驗 , 都 以 一 次 函 數 關 係 說 明 , 僅 Monje 及 Bugbee (6) 提 出 以 二 次 函 數 關 係 可 能 較 一 次 函數 關 係 更 適 當 , 而 本 試 驗 結 <strong>果</strong> 說 明 <strong>葉</strong> <strong>綠</strong> <strong>素</strong> 計 讀 值 與 丙 酮 萃 取 <strong>葉</strong> <strong>綠</strong> <strong>素</strong> <strong>量</strong> 間 <strong>應</strong> 是 呈 二 次 效 <strong>應</strong> 的函 數 關 係 。色 差 儀 以 光 線 反 射 <strong>之</strong> 原 理 , 因 此 <strong>葉</strong> <strong>片</strong> 厚 度 、 <strong>含</strong> 水 <strong>量</strong> 及 表 面 附 著 灰 塵 , 都 會 影 響 其 <strong>測</strong> <strong>定</strong>值 (6,13) , 本 試 驗 <strong>測</strong> <strong>量</strong> <strong>葉</strong> <strong>片</strong> 顏 色 值 , 係 以 CIE (Commission International I’Eclairage) L*a*b* 公 式計 算(10) , 根 據 公 式 原 理 當 顏 色 偏 <strong>綠</strong> 時 b* 值 變 大 , 因 此 <strong>葉</strong> <strong>綠</strong> <strong>素</strong> <strong>含</strong> <strong>量</strong> 增 加 時 (a*/b*) 2 <strong>之</strong> 值 變 小 ,因 此 圖 一 、 圖 三 及 圖 五 皆 呈 二 次 函 數 關 係 , 但 <strong>在</strong> Singha 及 Townsend (13) <strong>之</strong> 試 驗 , 並 未 提 出 相同 的 看 <strong>法</strong> 。 另 <strong>在</strong> 本 試 驗 以 色 差 儀 色 彩 值 與 丙 酮 萃 取 <strong>葉</strong> <strong>綠</strong> <strong>素</strong> <strong>量</strong> 間 決 <strong>定</strong> 係 數 , 芒 <strong>果</strong> (R 2 =0.88) 與荔 枝 (R 2 =0.93) 較 梨 (R 2 =0.75) 高 , 可 能 是 芒 <strong>果</strong> 及 荔 枝 <strong>葉</strong> <strong>片</strong> 表 面 光 滑 , 而 梨 <strong>葉</strong> 脈 <strong>之</strong> 支 脈 較 為 明顯 , 表 面 較 不 規 則 等 表 面 結 構 造 成 <strong>之</strong> 影 響 。綜 合 本 試 驗 <strong>之</strong> 結 <strong>果</strong> , <strong>葉</strong> <strong>綠</strong> <strong>素</strong> 計 (SPAD-502) 讀 值 及 色 差 儀 <strong>之</strong> (a*/b*) 2 值 作 <strong>葉</strong> <strong>綠</strong> <strong>素</strong> <strong>含</strong> <strong>量</strong> <strong>測</strong><strong>定</strong> , <strong>應</strong> 利 <strong>用</strong> 不 同 生 育 階 段 <strong>葉</strong> <strong>片</strong> 作 檢 <strong>量</strong> 線 , 可 得 到 較 正 確 的 數 值 。 而 二 <strong>種</strong> 儀 器 <strong>之</strong> 方 <strong>法</strong> <strong>在</strong> <strong>測</strong> <strong>定</strong>芒 <strong>果</strong> 荔 枝 及 梨 <strong>之</strong> <strong>葉</strong> <strong>片</strong> 時 , 所 得 到 的 迴 歸 方 程 式 皆 呈 二 次 效 <strong>應</strong> , 但 <strong>在</strong> 某 些 狀 況 , 如 <strong>葉</strong> <strong>片</strong> 厚 度超 過 1.2mm、 <strong>葉</strong> <strong>綠</strong> <strong>素</strong> <strong>含</strong> <strong>量</strong> 較 高 、 分 佈 不 均 勻 或 <strong>葉</strong> 表 結 構 不 適 合 等 , 都 會 影 響 估 算 值 <strong>之</strong> 準 確 性 。
