the residual breast tissue becoming a cation exchange material so that avidin is<br />

held in that tissue <strong>for</strong> several days. <strong>Yttrium</strong>-<strong>90</strong> r-BHD administered intravenously<br />

~1 d later homes in on the avidin held in the breast tissue to deliver a radiation<br />

dose to the tissue. To allow further clinical studies with this technique, it would<br />

be necessary that the <strong>90</strong> Y r-BHD be prepared in a pharmaceutically controlled<br />

environment or, alternatively, be prepared by means of an automatic module<br />

that may be considered a closed system, thus requiring less strict environmental<br />

constraints. There<strong>for</strong>e, a study was done on the labelling of r-BHD with <strong>90</strong> Y in an<br />

automatic module of synthesis (PharmTracer) <strong>and</strong> a disposable sterile single use<br />

kit employed in the department <strong>for</strong> the labelling of peptides with 177 Lu <strong>and</strong> <strong>90</strong> Y.<br />

7.2. MATERIALS AND METHODS<br />

7.2.1. Reagents<br />

<strong>Yttrium</strong>-<strong>90</strong> chloride ( <strong>90</strong> YCl 3 ) in 0.05M HCl was purchased from<br />

PerkinElmer. The r-BHD was provided by Sigma Tau S.p.A. <strong>and</strong> dissolved<br />

in saline at a concentration of 2 mg/mL. Pure hen egg avidin was provided<br />

by Tecnogen S.p.A. Sodium acetate anhydrous DTPA, HABA, D-biotin<br />

(vitamin H) <strong>and</strong> non-radioactive 89 YCl 3 at the highest analytical grade available<br />

were purchased from Sigma-Aldrich. Pharmaceutical grade ascorbic acid was<br />

purchased from Carlo Erba. Unless otherwise stated, all solutions were prepared<br />

using ultrapure water (18 MΩ·cm resistivity).<br />

7.2.2. Synthesis of r-BHD<br />

Details of the synthetic pathway <strong>and</strong> chemical characterization of<br />

10-[2-[6-[5-[(3aS,4S,6aR)-hexahydro-2-oxo-1H-thieno[3,4]imidazol-4-yl]-<br />

1-pentylamino]-1-hexylamino]-2-oxoethyl][1,4,7,10-tetraazacyclododecane-<br />

1,4,7,10-tetraacetic acid] DOTA have been described elsewhere [7.10].<br />

7.2.3. Effect of pH on radiolabelling<br />

To determine the optimal pH <strong>for</strong> the labelling of r-BHD with <strong>90</strong> Y, the<br />

experiments were per<strong>for</strong>med at a pH ranging between 3.6 <strong>and</strong> 5.6 using 2.0M<br />

sodium acetate buffer. Because of the high buffering capacity, the final pH of<br />

the reaction mixtures was equal to that of the 2.0M sodium acetate buffer used,<br />

as confirmed by electronic pH meter measurements (Hanna Instruments). All<br />

experiments were carried out in triplicate.<br />
