<strong>IARC</strong> MONOGRAPHS – 100C<br />

and 80 female SPF Wistar rats after 120 weeks.<br />

In ano<strong>the</strong>r experiment, 16 male and 16 female<br />

SPF Wistar rats dosed similarly with M<strong>in</strong>-U-Sil<br />

<strong>quartz</strong> were observed until <strong>the</strong>y were m<strong>or</strong>ibund.<br />

Eight <strong>of</strong> <strong>the</strong> 32 rats developed MLHT and three<br />

developed thymomas/lymphosarcomas. In a<br />

last experiment with <strong>the</strong> same experimental<br />

design, 18 <strong>of</strong> 32 SPF Wistar rats that had been<br />

<strong>in</strong>jected with <strong>cristobalite</strong> developed malignant<br />

lymphomas (13 MLHT and five thymomas/<br />

lymphosarcomas). No MLHT and one thymoma/<br />

lymphosarcoma tumours were observed <strong>in</strong> 15<br />

sal<strong>in</strong>e-<strong>in</strong>jected control rats. (Wagner, 1976). [The<br />

W<strong>or</strong>k<strong>in</strong>g Group noted that <strong>the</strong> distribution <strong>of</strong><br />

tumours over sexes was unspecified, and that no<br />

statistics were provided.]<br />

In one experiment, groups <strong>of</strong> 16 male and 16<br />

female Wistar rats were given <strong>in</strong>trapleural <strong>in</strong>jections<br />

<strong>of</strong> 20 mg <strong>of</strong> four types <strong>of</strong> <strong>quartz</strong> (M<strong>in</strong>-U-Sil,<br />

D&D, Snowit, and DQ12). The animals were<br />

observed f<strong>or</strong> <strong>the</strong>ir lifespan. F<strong>or</strong> all but <strong>the</strong> group<br />

treated with DQ12 <strong>quartz</strong>, <strong>the</strong>re was a statistically<br />

significant <strong>in</strong>crease <strong>in</strong> MLHT over sal<strong>in</strong>e<br />

controls (Table 3.5). In a second experiment with<br />

<strong>the</strong> same experimental design, two o<strong>the</strong>r stra<strong>in</strong>s<br />

<strong>of</strong> rats were <strong>in</strong>jected M<strong>in</strong>-U-Sil (12 male and<br />

12 female PVG rats and 20 male and 20 female<br />

Agus rats). A non-significant <strong>in</strong>crease <strong>in</strong> MLHT<br />

was observed <strong>in</strong> both stra<strong>in</strong>s, and <strong>the</strong>re was no<br />

MLHT <strong>in</strong> <strong>the</strong> sal<strong>in</strong>e controls. In a third experiment<br />

with <strong>the</strong> same experimental design, <strong>cristobalite</strong><br />

was <strong>in</strong>jected, and 4/32 treated Wistar rats<br />

developed MLHT [not significant], but none <strong>of</strong><br />

<strong>the</strong> 32 sal<strong>in</strong>e controls did. In a f<strong>in</strong>al experiment,<br />

16 male and 16 female Wistar rats were <strong>in</strong>jected<br />

triolymite (size, < 0.5 μm; 0.35x10 6 particle/μg),<br />

and observed f<strong>or</strong> <strong>the</strong>ir lifespan. A total <strong>of</strong> 16/32<br />

Wistar rats developed MLHT, whereas no such<br />

tumours were observed <strong>in</strong> <strong>the</strong> 32 sal<strong>in</strong>e controls<br />

(Wagner et al., 1980). [The W<strong>or</strong>k<strong>in</strong>g Group noted<br />

that <strong>the</strong> distribution <strong>of</strong> tumours over sexes was<br />

unspecified.]<br />

 Two groups <strong>of</strong> 36 2-month-old male<br />

Sprague-Dawley rats, received a s<strong>in</strong>gle<br />

<strong>in</strong>trapleural <strong>in</strong>jection <strong>of</strong> 20 mg DQ12 <strong>quartz</strong> <strong>in</strong><br />

sal<strong>in</strong>e <strong>or</strong> sal<strong>in</strong>e alone, and were observed f<strong>or</strong><br />

<strong>the</strong>ir lifespan. Twenty-seven male rats served<br />

as untreated controls. Six malignant histiocytic<br />

lymphomas and two malignant Schwannomas<br />

were observed <strong>in</strong> <strong>the</strong> <strong>quartz</strong>-treated group [not<br />

significant], and one chronic lymphoid leukaemia<br />

and one fibrosarcoma were observed <strong>in</strong> <strong>the</strong><br />

sal<strong>in</strong>e group and untreated controls, respectively<br />

(Jaurand et al., 1987).<br />

3.5 Intraperitoneal adm<strong>in</strong>istration<br />

3.5.1 Rat<br />

Two groups <strong>of</strong> 16 male and 16 female SPF<br />

Wistar rats received a s<strong>in</strong>gle <strong>in</strong>traperitoneal<br />

<strong>in</strong>jection <strong>of</strong> 20 mg <strong>of</strong> M<strong>in</strong>-U-Sil <strong>quartz</strong> <strong>in</strong> sal<strong>in</strong>e,<br />

and were observed f<strong>or</strong> <strong>the</strong>ir lifespan. There were<br />

12 sal<strong>in</strong>e controls [sex unspecified]. A total <strong>of</strong><br />

9/64 <strong>quartz</strong>-exposed rats developed malignant<br />

lymphomas (two MLHT and seven thymoma/<br />

lymphosarcomas). None <strong>of</strong> <strong>the</strong> sal<strong>in</strong>e controls<br />

developed MLHT, but one thymoma/lymphosarcoma<br />

was noted (Wagner, 1976). [The W<strong>or</strong>k<strong>in</strong>g<br />

3.6 Subcutaneous adm<strong>in</strong>istration<br />

3.6.1 Mouse<br />

Two groups <strong>of</strong> 40 female (C57xBALB/c) F 1<br />

mice received a s<strong>in</strong>gle subcutaneous <strong>in</strong>jection<br />

<strong>of</strong> 4 mg <strong>of</strong> d- <strong>or</strong> l-<strong>quartz</strong>. A group <strong>of</strong> 60 female<br />

mice served as sal<strong>in</strong>e controls. At 18 months after<br />

<strong>in</strong>jection, <strong>the</strong>re was an <strong>in</strong>cidence <strong>of</strong> lymphomas/<br />

leukemias <strong>of</strong> 0/60, 1/40 and 12/40 (P < 0.001), and<br />

<strong>of</strong> liver adenomas <strong>of</strong> 0/60, 1/40 and 3/40 f<strong>or</strong> <strong>the</strong><br />

sal<strong>in</strong>e controls, d-<strong>quartz</strong> and l-<strong>quartz</strong> groups,<br />

respectively. No <strong>in</strong>jection-site tumours were<br />

rep<strong>or</strong>ted (Ebbesen, 1991).<br />
