From Spieth<strong>of</strong>f et al. (1992)<br />
Squamous cell<br />
carc<strong>in</strong>oma<br />
lung tumours <strong>in</strong> <strong>the</strong> untreated rats (0/45), n<strong>or</strong> <strong>in</strong><br />
those exposed to benzo[a]pyrene alone (0/19). In<br />
<strong>the</strong> comb<strong>in</strong>ed exposures to benzo[a]pyrene and<br />
<strong>quartz</strong>, an <strong>in</strong>creased <strong>in</strong>cidence <strong>in</strong> lung tumours<br />
was observed (Group A, 14/31, 11 squamous cell<br />
carc<strong>in</strong>omas and three papillomas; Group B, 4/18,<br />
two papillomas and two carc<strong>in</strong>omas) (Pylev,<br />
1980). [The W<strong>or</strong>k<strong>in</strong>g Group noted <strong>the</strong> absence <strong>of</strong><br />
a group exposed to <strong>quartz</strong> alone.]<br />
(b) Hamster<br />
Groups <strong>of</strong> 50 male Syrian golden hamsters<br />
received weekly <strong>in</strong>tratracheal <strong>in</strong>stillations f<strong>or</strong><br />
15 weeks <strong>in</strong> sal<strong>in</strong>e <strong>of</strong> 3 mg benzo[a]pyrene <strong>or</strong><br />
3 mg ferric oxide <strong>or</strong> 3 mg ferric oxide plus 3 mg<br />

M<strong>in</strong>-U-Sil <strong>or</strong> 0.7 mg M<strong>in</strong>-U-Sil plus 3 mg benzo[a]<br />
pyrene <strong>or</strong> 7 mg M<strong>in</strong>-U-Sil <strong>or</strong> 7 mg M<strong>in</strong>-U-Sil<br />
plus 3 mg benzo[a]pyrene. Fifty male controls<br />
received sal<strong>in</strong>e alone. Surviv<strong>or</strong>s were killed at 92<br />
weeks after exposure. Co-exposures with <strong>silica</strong><br />
caused an enhancement <strong>of</strong> <strong>the</strong> number <strong>of</strong> respirat<strong>or</strong>y<br />
tract tumours <strong>in</strong>duced by benzo[a]pyrene<br />
(ma<strong>in</strong>ly <strong>in</strong> <strong>the</strong> bronchus and lung) (Niemeier<br />
et al., 1986; Table 3.7).<br />
3.9 Syn<strong>the</strong>sis<br />
Studies <strong>of</strong> <strong>the</strong> carc<strong>in</strong>ogenicity <strong>of</strong> <strong>crystall<strong>in</strong>e</strong><br />
<strong>silica</strong> <strong>in</strong> experimental animal models have<br />
shown <strong>quartz</strong> <strong>dust</strong> to be a lung carc<strong>in</strong>ogen <strong>in</strong> rats<br />
follow<strong>in</strong>g <strong>in</strong>halation and <strong>in</strong>tratracheal <strong>in</strong>stillation,<br />
but not <strong>in</strong> mice <strong>or</strong> hamsters. Rats are known<br />
to be m<strong>or</strong>e sensitive than are mice <strong>or</strong> hamsters<br />
to <strong>the</strong> <strong>in</strong>duction <strong>of</strong> lung tumours due to o<strong>the</strong>r<br />
<strong>in</strong>haled po<strong>or</strong>ly soluble particles, such as carbon<br />
black (Mauderly et al., 1994).<br />
Quartz-<strong>in</strong>duced lymphoma <strong>in</strong>cidence was<br />
also <strong>in</strong>creased <strong>in</strong> several experiments <strong>in</strong> rats after<br />
<strong>in</strong>trapleural adm<strong>in</strong>istration, and <strong>in</strong> one study<br />
<strong>in</strong> mice after subcutaneous adm<strong>in</strong>istration.<br />
Tridymite- and <strong>cristobalite</strong>-<strong>in</strong>duced lymphomas<br />
were observed <strong>in</strong> only a s<strong>in</strong>gle experiment.<br />
