Description: The homogenous Caspase-Glo ® 6 Assay measures caspase-6<br />

acitivity. Caspase-6 is a member of the cysteine aspartic acid-specific protease<br />

family and has a key effector role in the cleavage of specific target proteins<br />

during apoptosis. The Caspase-Glo ® 6 Assay provides a luminogenic substrate,<br />

Z-VEID-aminoluciferin, in a buffer optimized for caspase-6 and luciferase<br />

activity. The addition of a single Caspase-Glo ® 6 Reagent results in cleavage of<br />

the substrate, releasing aminoluciferin, and generating a glow-type luminescent<br />

signal in the presence of Ultra-Glo Recombinant Luciferase. The luminescent<br />

signal is proportional to the amount of caspase-6 activity. The Caspase-Glo ®<br />

6 Assay is designed for use with purified enzyme preparations in multiwell<br />

plate formats, making it ideal for automated high-throughput screening for<br />

caspase-6 activity and inhibitors of caspase-6 activity.<br />

Description: The homogeneous Caspase-Glo ® 8 Assay measures caspase-8<br />

activity using a proluminogenic caspase-8 substrate in a buffer system<br />

optimized for caspase activity, luciferase activity and cell lysis. The addition of a<br />

single Caspase-Glo ® 8 Reagent in an “add-mix-measure” format results in cell<br />

lysis, followed by caspase cleavage of the substrate and generation of a “glowtype”<br />

luminescent signal. The signal generated is proportional to the amount of<br />

caspase activity present. The Caspase-Glo ® Reagent relies on the properties<br />

of a proprietary thermostable luciferase (Ultra-Glo Recombinant Luciferase),<br />

which generates the stable “glow-type” luminescent signal and improves<br />

performance across a wide range of assay conditions.<br />

The system now includes a separate vial of MG-132 protease inhibitor,<br />

which may be used to reduce background, improving the performance of the<br />

Caspase-Glo ® 8 Assay in cell-based applications.<br />

Reagent significantly reduces nonspecific background in cell-based assays.<br />
