NATIONAL AGRI-FOOD BIOTECHNOLOGY INSTITUTE4.1.3 Characterization of arabinoxylans in regulating their biological activities will becereal grainsevaluated using in-vitro and in-vivo model, whichwill further allow us to understand the structurefunctionrelationship of the AXs in Indian milletPrincipal Investigator:Koushik Mazumdervarieties with respect to their biological activities.Introduction:Compositional sugar analysis and structuralcharacterization of the arabinoxylanpolysaccharides and oligosaccharides usingvarious chemical and modern analyticalmethods (GC-MS, HPLC, MALDI-TOFand ESI-MS/MS) to establish structure-function relationship.Cereals, the staple foods for millions of peopleacross the world, are the chief source of solubledietary fibers. Several epidemiological studieshave clearly demonstrated that increasedconsumption of whole grain cereals and solubledietary fibers has been associated with a reducedrisk of many life style disorders such as type-2diabetes and obesity. In cereal grains, AXs are themajor non-starchy polysaccharides whichconstitute the cell walls residues in cereal grainsand make up an important physicochemical andphysiological property beneficial to the health ofconsumers and for this reason, their use as foodingredients has increased rapidly. Although AXsshare the same basic chemical structure, but thepattern and degree of substitution of arabinosealong the xylan backbone vary with cerealsources. Therefore, the AXs exhibits a great dealof structural heterogeneity and variability withrespect to molecular weight mass, xylose toarabinose ratio and branching pattern,distribution of arabinose residues and substitutionwith glucuronic acid/4-O-methyl glucuronicacids.There is no documented report about thecomparative study of arabinoxylan poly andoligo-saccharides from Indian millet varietiessuch as finger millet, kodo millet, branyard milletand foxtail millet. Hence in the present study, thevariability in the structures of the AXs fromvarious Indian millet varieties and their role inResearch Objectives:The present study aims to elucidate comparativestatement profile of AXs from Indian milletvarieties and its effect on their biologicalactivities.llIsolation and purification of the arabinoxylanpolysaccharides and oligosaccharides fromthe bran of Indian variety millets.Long Term Objective:Development of nutraceutical health foods basedon dietary fibers enriched with Axs.Research in Progress:In our preliminary studies, we havestandardized the extraction protocol for theisolation of AXs from Finger millet bran and thecompositional analysis of the polysaccharideshave been carried out (GC, GC-MS analysis) asmonomeric alditol acetate derivative with inositolas internal standard (Figure 2). The compositionalanalysis showed the presence of arabinoxylan asmajor constituent (~90%) together with starch andgalactomannan as minor component.Figure 2: GC chromatogram of the monomeric sugar residues of extracted arbinoxylan fraction from fingermillet bran.
