ACE & Cellulosic Butanol C. thermocellum Cellulosome C. acetobutylicum Cellulosic clostridia make ethanol, not butanol They produce a ‘Cellulosome’:- a molecular nanomachine – very efficient in degrading lignocellulose The core of the complex is a modular, non-catalytic scaffoldin protein which serves to bring enzymes into close proximity via cohesin-dockerin interactions, enhance enzyme activity We seek to introduce a functional cellulosome into the genome of a noncellulolytic butanol producer using ACE technology
ACE & Cellulosic Butanol Cellulosome Scaffold Assembly • As proof of principle we tested the expression of a native hydrolase Cel5A in C. acetobutylicum in the presence and absence of the scaffoldin subunit • 1 st minicellulosome integrated into the genome of C. acetobutylicum • Functional benefit currently being tested
