It was constructed by digesting the vector pTrcHis/lacZ withNco I and Nsi I to remove the lacI q gene and the trc promoter and replacing with anNco I-Nsi I fragment containing the araC gene and the araBAD promoter. The β-galactosidaseportion of the fusion may be released by digestion with BamH I and Hind III.
