RGUHS Journal of Pharmaceutical Sciences A New Reverse Phase High Performance Liquid Chromatographic Method for Determination of Lornoxicam in Bulk and Tablet Dosage Form N.C.Arjun, J.Pavitha and Nagaraj* Original Research Article Department of Pharmaceutical Analysis, PES College of Pharmacy, Hanumanthanagar, Bangalore - 560050, India A B S T R A C T A rapid, accurate, precise and sensitive RP-HPLC method for the determination of the non-steroidal anti-inflammatory drug lornoxicam (LORN) in bulk and pharmaceutical tablet dosage form has been developed. The analyte is resolved by 0 using a reverse phase phenomenex-luna 5µ C-8 (2) 100A , 250 × 4.3mm, column at room temperature. The mobile phase -1 used was acetonitrile: 1% triethylamine in methanol (60:40 v/v) at a flow rate of 1.0 mL min on an isocratic HPLC system (shimadzu) consisting of LC 10AT VP liquid pump, SPD 10AVP UV-Visible detector at a wavelength of 289nm. The -1 retention time of LORN is 1.91 min. The linear dynamic range for LORN was 2-24 µg mL with the correlation -1 coefficient was 0.9992. The limits of detection and quantification were 0.3362 and 1.1206 µg mL , respectively. The developed method was validated in terms of linearity, accuracy, precision, sensitivity, ruggedness and robustness. The % relative standard deviations of peak areas from all the studies were always less than 2%. The proposed method can be used for routine estimation of LORN in bulk and tablet dosage form. INTRODUCTION Lornoxicam (chlortenoxicam) is 6-chloro-4-hydroxy-2methyl- N-2-pyridyl-2H-thieno [2, 3-e]-1, 2-thiazine-3carbox- amide-1, 1-dioxide (Fig.1); is a novel non-steroidal anti-inflammatory drug (NSAID) with marked analgesic [1] properties . LORN belongs to the chemical class oxicams, which includes lornoxicam, tenoxicam and meloxicam. [2] LORN is a Yellow or slightly yellow powder . It works by blocking the action of cyclooxygenase, an enzyme involved in the production of chemicals, including some [3] prostaglandins in the body . It is used in musculoskeletal and joint disorders such as osteoarthritis and rheumatoid arthritis; it is also used in the treatment of other painful [4] conditions including postoperative pain . Literature survey revealed that various analytical methods [5] [6] such as TLC , liquid chromatography , polarographic [7] method , LC-ESI-MS (liquid chromatography- [8] Electrospray ionization- tandem- mass spectrometric) , [9, 10, 11, 12] spectrophotometric method , stability-indicating [13] [14, 15] HPLC and RP-HPLC method are used for estimation of LORN from its formulations individually and in combination with other drugs. The proposed RP-HPLC method has an advantage over reported RP-HPLC method, that the total run time was less than four minutes, more precise and more accurate. The method was validated as per ICH [16] guidelines . MATERIALS AND METHODS Instrumentation The Shimadzu HPLC system consisting of gradient pump (LC-10AT VP pump), mixer (SUS vp Assy (new)), rheodyne injector (20 µL), UV-VIS dual wavelength detector SPD - 10A VP) and Hamilton syringe (all from Shimadzu, Kyoto Japan) was used. The separations were achieved on a 0 Phenomenex - Luna 5µ C-8(2) 100 A , 250X4.3mm, column with UV detection at 289 nm. Analytical weighing balance (Shimadzu AUX 200) was used for weighing, sonicator (SONICA 2200MH), vaccum pump (model XI 5522050 of Millipore), Millipore filtration kit for solvents and sample filtration were used throughout the experiment. The Spinchrom CFR software-single channel was used for acquisition, evaluation and storage of chromatographic data. LORN was obtained gift sample from M/s. Micro Labs, Hosur. HPLC grade acetonitrile (ACN) and AR grade triethylamine (TEA) were obtained from Spectrochem Pvt.Ltd, Mumbai, (India). HPLC grade methanol (MeOH) was obtained from CDH (P) Ltd. Sarigam, (India).
