2.0. Review <str<strong>on</strong>g>of</str<strong>on</strong>g> literature<br />

2.1. Historical review<br />

<str<strong>on</strong>g>Aflatoxin</str<strong>on</strong>g>s were first identified as etiological agents for animal disease<br />

in the early 1960's, following an outbreak <str<strong>on</strong>g>of</str<strong>on</strong>g> deaths <str<strong>on</strong>g>of</str<strong>on</strong>g> turkey in Engl<str<strong>on</strong>g>and</str<strong>on</strong>g> <str<strong>on</strong>g>and</str<strong>on</strong>g><br />

elsewhere. The disease, termed Turkey X disease because <str<strong>on</strong>g>of</str<strong>on</strong>g> the then<br />

unknown etiology was characterized by acute hepatic necrosis with bile duct<br />

hyperplasia, lethargy, loss <str<strong>on</strong>g>of</str<strong>on</strong>g> appetite <str<strong>on</strong>g>and</str<strong>on</strong>g> death. The cause <str<strong>on</strong>g>of</str<strong>on</strong>g> the effects was<br />

traced to Brazilian peanut meal that was used in a comp<strong>on</strong>ent <str<strong>on</strong>g>of</str<strong>on</strong>g> the poultry<br />

rati<strong>on</strong> (AIlcr<str<strong>on</strong>g>of</str<strong>on</strong>g>t <str<strong>on</strong>g>and</str<strong>on</strong>g> Carnaghan, 1963a). The toxic factors in the peanut meal,<br />

namely aflatoxins were separated in to four distinct compounds: aflatoxin Bl,<br />

Bz, G1 <str<strong>on</strong>g>and</str<strong>on</strong>g> Gz (Nesbitt etal, 1962; Sargeant et al,1961).<br />

2.2. Structural diversity <str<strong>on</strong>g>of</str<strong>on</strong>g><str<strong>on</strong>g>Aflatoxin</str<strong>on</strong>g>s<br />

Nearly all <str<strong>on</strong>g>of</str<strong>on</strong>g> the interest in aflatoxins has focused <strong>on</strong> AFBl, primarily<br />

due to its extreme acute <str<strong>on</strong>g>and</str<strong>on</strong>g> chr<strong>on</strong>ic toxicity <str<strong>on</strong>g>and</str<strong>on</strong>g> its carcinogenic activity in<br />

animals, in additi<strong>on</strong> to its potential effects in humans. The toxic factors<br />

isolated from feed were separated chromatographically into four distinct<br />

compounds: aflatoxins Bl, Bz, G1 <str<strong>on</strong>g>and</str<strong>on</strong>g> Gz.The molecular formula indicated<br />

that aflatoxins Bz <str<strong>on</strong>g>and</str<strong>on</strong>g> Gz (AFBz <str<strong>on</strong>g>and</str<strong>on</strong>g> AFGz) were dihydro derivatives <str<strong>on</strong>g>of</str<strong>on</strong>g> the<br />

parent AFB1 <str<strong>on</strong>g>and</str<strong>on</strong>g> G1 respectively (Asao et al, 1963;Chang et al, 1963;<br />

Cheung <str<strong>on</strong>g>and</str<strong>on</strong>g> Sill, 1964; Van der Merve et al, 1963; Van Dorp et al, 1963;<br />

Van Soest <str<strong>on</strong>g>and</str<strong>on</strong>g> Peerdeman, 1964). AfIatoxins c<strong>on</strong>tain a coumarin nucleus<br />

fused to a bifuran <str<strong>on</strong>g>and</str<strong>on</strong>g> either a pentan<strong>on</strong>e (AFB 1 <str<strong>on</strong>g>and</str<strong>on</strong>g> Bz) or a six membered<br />

lact<strong>on</strong>e (AFG1 <str<strong>on</strong>g>and</str<strong>on</strong>g> Gz). AFB1 <str<strong>on</strong>g>and</str<strong>on</strong>g> G1 were more toxic to ducklings, rats, <str<strong>on</strong>g>and</str<strong>on</strong>g><br />

fish than either AFBz or AFG2 with AFB 1 being the most toxic (Wogan et al,<br />

1971; Abedi <str<strong>on</strong>g>and</str<strong>on</strong>g> Scott, 1969). A similar pattern holds for its carcinogenic<br />

potency, AFB 1 > AFG 1 > AFBz (Wogan et al, 1971; Ayes et al, 1971).<br />
