4.5 The Antioxidant Defense Mechanism<br />

To counter the harmful effects <str<strong>on</strong>g>of</str<strong>on</strong>g> ROS, antioxidant defense<br />

mechanism operate to detoxify or scavenge these reactive oxygen species.<br />

The antioxidant system comprises <str<strong>on</strong>g>of</str<strong>on</strong>g> different types <str<strong>on</strong>g>of</str<strong>on</strong>g> functi<strong>on</strong>al<br />

comp<strong>on</strong>ents classified as first line, sec<strong>on</strong>d line <str<strong>on</strong>g>and</str<strong>on</strong>g> third line defenses.<br />

4.6 Preventive Antioxidants - First Line Defense<br />

The first line defense comprises preventive antioxidants that act by<br />

quenching <str<strong>on</strong>g>of</str<strong>on</strong>g> Oz, decompositi<strong>on</strong> <str<strong>on</strong>g>of</str<strong>on</strong>g> HzOz <str<strong>on</strong>g>and</str<strong>on</strong>g> sequestrati<strong>on</strong> <str<strong>on</strong>g>of</str<strong>on</strong>g> metal i<strong>on</strong>s. The<br />

antioxidants bel<strong>on</strong>ging to this category are enzymes like superoxide<br />

dismutase (SOD), catalase, glutathi<strong>on</strong>e peroxidase <str<strong>on</strong>g>and</str<strong>on</strong>g> glutathi<strong>on</strong>e reductase<br />

<str<strong>on</strong>g>and</str<strong>on</strong>g> n<strong>on</strong>-enzymatic molecules like minerals <str<strong>on</strong>g>and</str<strong>on</strong>g> some proteins.<br />

(a) Superoxide dismutase (SOD) (E.C.l.15.1.1)<br />

SOD mainly act by quenching <str<strong>on</strong>g>of</str<strong>on</strong>g> superoxide (02.') , an active oxygen<br />

radical, produced in different stages <str<strong>on</strong>g>of</str<strong>on</strong>g> aerobic metabolism (Meier et al,<br />

1998; MacMillan-Crow et al, 1998; Yamakura et al, 1998; LiY et al, 1995;<br />

Kizaki et ai, 1997)<br />

Different enzymes <str<strong>on</strong>g>of</str<strong>on</strong>g> SOD are described. The <strong>on</strong>e present m the<br />

mitoch<strong>on</strong>dria is Mn z + dependent, that present in the cytoplasm is<br />

independent. An extra cellular Cu-Zn dependent enzyme is also reported<br />

(Halliwell <str<strong>on</strong>g>and</str<strong>on</strong>g> Gatteridge, 1985; Fridovich, 1989).<br />
