soluble in the same solvents as aflatoxin, therefore, selective extracti<strong>on</strong> or<br />

extensive purificati<strong>on</strong> methods are required to produce pure extracts.<br />

Therefore, the nature <str<strong>on</strong>g>of</str<strong>on</strong>g> the sample <str<strong>on</strong>g>and</str<strong>on</strong>g> properties <str<strong>on</strong>g>of</str<strong>on</strong>g> aflatoxin reflect the<br />

type <str<strong>on</strong>g>of</str<strong>on</strong>g> extracti<strong>on</strong> procedure. <str<strong>on</strong>g>Aflatoxin</str<strong>on</strong>g>s are soluble in slightly polar solvents<br />

<str<strong>on</strong>g>and</str<strong>on</strong>g> insoluble in completely n<strong>on</strong> polar solvents. Practically all aflatoxin are<br />

extracted using mixture <str<strong>on</strong>g>of</str<strong>on</strong>g> organic solvents such as acet<strong>on</strong>e, chlor<str<strong>on</strong>g>of</str<strong>on</strong>g>orm, or<br />

methanol in combinati<strong>on</strong> with small amounts <str<strong>on</strong>g>of</str<strong>on</strong>g> water (Bullerman, 1979).<br />

Aqueous solvents more easily penetrate hydrophilic tissues <str<strong>on</strong>g>and</str<strong>on</strong>g> enhance<br />

aflatoxin extracti<strong>on</strong> (Moss <str<strong>on</strong>g>and</str<strong>on</strong>g> Smith, 1985). Characteristic fluorescence<br />

(Sargeant et at) 1961a) <str<strong>on</strong>g>and</str<strong>on</strong>g> absorpti<strong>on</strong> under l<strong>on</strong>g wave ultraviolet light<br />

(V<str<strong>on</strong>g>and</str<strong>on</strong>g>er Merwe <str<strong>on</strong>g>and</str<strong>on</strong>g> Fourie,1963) aid detecti<strong>on</strong> <str<strong>on</strong>g>and</str<strong>on</strong>g> estimati<strong>on</strong>.<br />

3.2 Detecti<strong>on</strong> <str<strong>on</strong>g>of</str<strong>on</strong>g><str<strong>on</strong>g>Aflatoxin</str<strong>on</strong>g><br />

Analytical methods followed for the qualitative <str<strong>on</strong>g>and</str<strong>on</strong>g> quantitative<br />

estimati<strong>on</strong> are TLC (Visual <str<strong>on</strong>g>and</str<strong>on</strong>g> fluorodensitometric), HPLC, ELISA <str<strong>on</strong>g>and</str<strong>on</strong>g><br />

RIA. Visual TLC is the method <str<strong>on</strong>g>of</str<strong>on</strong>g> choice in the countries where other<br />

expensive instruments <str<strong>on</strong>g>and</str<strong>on</strong>g> the infrastructure for immunoassays are not<br />

available, though it is criticized for high degree <str<strong>on</strong>g>of</str<strong>on</strong>g> variati<strong>on</strong> due to<br />

individual's acuity. Visual TLC estimati<strong>on</strong> is simple <str<strong>on</strong>g>and</str<strong>on</strong>g> reliable, so l<strong>on</strong>g as<br />

the analyst ensures the validity <str<strong>on</strong>g>of</str<strong>on</strong>g> the method by acceptable recovery<br />

experiment. Background fluorescence should be c<strong>on</strong>sidered, whenever<br />

fluorodensitometer is used. Immunoassays <str<strong>on</strong>g>and</str<strong>on</strong>g> HPLC methods, although<br />

sensitive are not readily applicable (Shantha, 1994). As Egm<strong>on</strong>d <str<strong>on</strong>g>and</str<strong>on</strong>g><br />

Wagstaffe, 1989 opined, it is more important to apply rigorous quality<br />

assurance to the measurement procedure than to rely blindly up<strong>on</strong><br />

st<str<strong>on</strong>g>and</str<strong>on</strong>g>ardized <str<strong>on</strong>g>and</str<strong>on</strong>g> <str<strong>on</strong>g>of</str<strong>on</strong>g>ten archaic methods. Bearing this in mind, it became<br />
