[CULSPU] Sputum Cult & Gram Stain
SPECIMEN REQUIRED: Sputum
EXPECTORATED SPUTUM:
1. Have the patient rinse and gargle with water prior to
specimen collection. Instruct the patient not to
expectorate saliva or post-nasal discharge. Early
morning, freshly expectorated sputum is preferred
2. Collect specimen resulting from a deep cough in
a sterile screw-capped container
INDUCED SPUTUM (performed by respiratory therapy personnel)
1. Using an ultrasonic nebulizer, have the patient inhale
approximately 20 to 30 ml of 3 to 10% 0.85% NaCl.
2. Collect induced sputum in sterile screw-capped cup
TUBE OR CONTAINER: Sterile container
SPECIAL HANDLING: Indicate if sputum is induced or not induced.
If specimen is from tracheal aspirate, please
order CULTRA (Culture, trach sputum)
Specimens with >10 squamous epithelial cells/
LPF are considered heavily contaminated with
saliva and are unsatisfactory for culture.
Recollection will be requested
