[CULSPU] Sputum Cult & Gram Stain<br />

SPECIMEN REQUIRED: Sputum<br />

EXPECTORATED SPUTUM:<br />

1. Have the patient rinse and gargle with water prior to<br />

specimen collection. Instruct the patient not to<br />

expectorate saliva or post-nasal discharge. Early<br />

morning, freshly expectorated sputum is preferred<br />

2. Collect specimen resulting from a deep cough in<br />

a sterile screw-capped container<br />

INDUCED SPUTUM (performed by respiratory therapy personnel)<br />

1. Using an ultrasonic nebulizer, have the patient inhale<br />

approximately 20 to 30 ml of 3 to 10% 0.85% NaCl.<br />

2. Collect induced sputum in sterile screw-capped cup<br />

TUBE OR CONTAINER: Sterile container<br />

SPECIAL HANDLING: Indicate if sputum is induced or not induced.<br />

If specimen is from tracheal aspirate, please<br />

order CULTRA (Culture, trach sputum)<br />

Specimens with >10 squamous epithelial cells/<br />

LPF are considered heavily contaminated with<br />

saliva and are unsatisfactory for culture.<br />

Recollection will be requested<br />
