Preparation of an autologous rescuing construct fusing the daf-16promoter to the S. stercoralis irk-1 coding sequence and a homologous rescuing construct fusing the daf-16 promoter and thedaf-2 coding sequence are nearing completion. Transgenic lines of daf-2 mutants and wild-type (N2 strain) transformed withrol-6, a co-transformation marker, have been established as negative and positive controls. Dauer switching assays revealedthat percentages of dauer and non-dauer development in N2 worms transformed with rol-6 are 0% (0/18) and 100% (18/18)respectively. Percentages of dauer, non-dauer, and pre-dauer development in daf-2 mutants transformed with rol-6 is 96%(45/47), 0% (0/47), and 4% (2/47). These baseline data will be compared to the proportions of dauer and non-dauerdevelopment in both N2 worms and daf-2 mutants transformed with the parasite gene irk-1. The ovine follicle wave is defined as ≥1 folliclegrowing from 2-3 to ≥5 mm in diameter. Every 4-5 d, a peak in serum follicle stimulating hormone (FSH) concentrationsstimulates the emergence of a follicle wave in the ewe. Previous studies in our lab have shown that exogenous FSH given inthe middle of the interwave interval will induce a follicle wave without disrupting endogenous waves. This surprisingdiscovery suggested that the follicular dominance seen in cattle does not exist in sheep. The objective of this experiment wasto give daily FSH injections to see if the ovary could respond with daily follicular waves.Transrectal ultrasonography was performed on 12 Western White Face ewes. Six ewes were given 2 injections of ovine FSH(oFSH; 0.35 µg/kg; sc; 8 h apart) daily for 4 d starting 84 h after the detection of a growing 4 mm follicle. The oFSH wasprepared in saline with 50% polyvinyl pyrrolidone and 0.05% BSA. The start of treatment coincides with the day after theemergence of the next endogenous wave and results in serum FSH concentrations similar in magnitude to an endogenousFSH peak. The exogenous oFSH was given between 2 consecutive endogenous FSH peaks. Six ewes only received vehicle.Blood samples were collected daily throughout the experimental period and every 6 h from 24 h before to 5 d after the firstinjection.During the 4 d treatment period, mean serum FSH concentrations were higher in the oFSH treated ewes than incontrol ewes (3.35±0.08 ng/mL, 1.58±0.03 ng/mL; P

Twenty-five fecal samples from adult bearded dragons from four different breeding colonies(n=100) will be used to validate the PCR assay. Bayesian analysis will be performed to estimate the prevalence and testsensitivity and specificity for the PCR and EM assays. This information will then be used as a basis for a large scale study toestimate the prevalence of adenovirus in the U.S. population of captive bearded dragons.93
