Alisal St, Salinas, CA, USAThe method of suppression subtractive hybridization (SSH) was employed toidentify lettuce (Lactuca sativa) genes that are differentially expressed insymptomatic leaves during a lettuce-V. dahliae interaction. A subset of cDNAs fromthe forward-(symptomatic) and reverse-subtracted (asymptomatic) SSH libraries weresequenced and subjected to homology searches using several databases. Genesidentified as expressed only in symptomatic leaves included those with homology topathogenesis-related (PR) protein genes such as PR-5 (thaumatin-like protein), PR-3(endochitinase), PR-1 (unknown function), PR-12 (defensin), and a putative lettucecysteine protease (LsCP1). In contrast, of those clones identified as differentiallyexpressed in the asymptomatic leaf tissue, only cDNAs sharing homology with PR-14(lipid transfer protein) were identified. Increased expression of PR-3 and PR-5 insymptomatic leaves was confirmed by quantitative RTPCR, and LsCP1 was onlyexpressed in symptomatic leaves. LsCP1 shares homology with plant cysteineproteases that are expressed in senescing leaves and may contribute to the leafsymptoms observed in this lettuce-V. dahliae interaction.38
