Doç. Dr. Fatma İnci ÖZDEMİR Eş-Danışman / Co-Advisor ÖZET Hem prokaryotik hem de ökaryotik hücrelerde protein yıkımı büyük oranda ATPbağımlı proteazlar tarafından gerçekleştirilmektedir. FtsH proteaz, bütün canlı organizmalarda bulunan bir ATP-bağımlı proteaz olup birçok hücre içi kontrol noktasında önemli rol oynamaktadır. Bununla beraber FtsH proteazın proteolitik aktivitesi ve çeşitli substratları nasıl tanıyabildiği hala tam olarak açık değildir. Farklı organizmalardan yeni FtsH proteazların izole edilmesi ve bu enzimlerin biyokimyasal ve genetik nitelendirilmelerinin yapılması, hem bu özgün proteaz sisteminin hücre içinde oynadığı biyolojik rolün, hem de protein hidroliz mekanizmasının anlaşılmasına yarar sağlayacaktır. Bu çalışmada, G. kaustophilus'a ait transmembran segmenti içermeyen yaklaşık 1530 bç uzunluğunda ftsH geni pET- 14b vektörüne aktarılmıştır. Gk-FtsH proteaz, N-terminal bölgesinde 6xHis kuyruk ile birlikte E. coli BL21 (DE3) hücrelerinde eksprese edilmeye çalışılmış ancak SDS- PAGE'de belirgin ekpresyon gözlemlenmemiştir. Gk-ftsH geninin transkripsiyonel olarak ifade edildiğini göstermek amacıyla total RNA izolasyonu ve cDNA sentezi yapılmış ve vektöre aktarımı yapılan genin transkribe olduğu gösterilmiştir. E. coli BL21 (DE3) hücresinde transkribe olan ancak protein ekspresyonu gözlenmeyen bu genin hücrede inklüzyon yapısı oluşturacağı düşüncesiyle inklüzyon yapılarından protein elde edilmiştir. Ayrıca olası 6xHis-FtsH füzyon proteini için Ni-NTA IMAC yöntemi kullanılarak saflaştırılma işlemi yapılmıştır. Her iki çalışmada elde edilen örnekler Western Blot ile analiz edilmiş ve His-tag antikoru ile bağlanan yaklaşık 70 kDa moleküler büyüklüğünde olası bant gözlemlenmiştir. Ancak elde edilen Gk-FtsH ABSTRACT In both eukaryotic and prokaryotic cells, protein degredation is largely conducted by ATP-dependent proteases. FtsH protease, an ATP-dependent protease found in all living organisms and plays a crucial role in many intracellular control points. However, the proteolytic activity of FtsH protease and how it can recognize various substrates are still not exactly clear. The isolation of new FtsH proteases from different organisms and biochemically and genetically clasification of these enzymes would help understanding the biologic role of this peculiar protease system in cell and the protein hydrolysis mechanism. In this study, approximately 1530 bp ftsH gene which do not include transmembran segment of G. kaustophilus was transferred into pET-14b vector. Gk-FtsH protease with 6xHis-tag at its N-terminus was tried to be expressed in E. coli BL21 (DE3) cells however no protein band was observed by SDS-PAGE analysis. In order to show the transcriptional expression of Gk-ftsH gene, total RNA isolation and cDNA synthesis were done and the transformed gene was shown to be transcribed. Since Gk-ftsH gene was transcribed but protein expression was not observed in E. coli BL21 (DE3) cells, it indicated an inclusion structure. Therefore we obtained protein from inclusion bodies. We also purified the fusion protein using Ni-NTA IMAC method. The samples obtained from both studies were analyzed with Western Blot and a approximately 70 kDa molecular weight contingent band was detected against His-tag antibody.
