In this chapter, the discussion is focused on these improved DNAvector-based shRNA-expression systems used in this laboratory. The protocolsdescribed in this chapter provide a comprehensive procedure for constructingthe simple and efficient shRNA-expression systems for specificgene silencing in mammalian cells.
