Section génétique (gen) Chef Dr Paul Brennan Il est important d’identifier les gènes spécifiques et les variants génétiques qui contribuent au développement du cancer pour plusieurs raisons, notamment pour comprendre en profondeur les voies biologiques impliquées dans le cancer, élucider la façon dont les facteurs environnementaux peuvent exercer leurs effets sur le développement du cancer en interaction avec les gènes, et identifier les individus ayant un risque suffisamment élevé pour être susceptibles de bénéficier des stratégies de réduction du risque. La Section Génétique réunit deux groupes ayant pour mission générale d’identifier les gènes impliqués dans le cancer, de caractériser le spectre des variants de séquence pathogènes qu’ils portent, et de comprendre comment ils interagissent avec les facteurs non génétiques. Il s’agit des Groupes Epidémiologie génétique (GEP) et Prédispositions génétiques au cancer (<strong>GC</strong>S). Le Groupe GEP participe essentiellement à la coordination de vastes études épidémiologiques au sein de la population et à l’analyse de multiples variants génétiques fréquents, afin d’identifier de nouveaux locus de prédisposition au cancer, en s’intéressant tout particulièrement aux cancers du poumon, des voies aérodigestives supérieures (VADS, notamment du rhinopharynx), du rein et aux cancers plus rares de l’enfant. De son côté, le Groupe <strong>GC</strong>S travaille principalement à l’identification de variants ou de mutations rares, dans des locus de prédisposition au cancer connus ou fortement candidats, qui confèrent un risque de cancer important. L’accent est mis sur le cancer du sein, en particulier sur les tumeurs mammaires de type basal, avec également un intérêt grandissant pour les mélanomes. Les résultats du Groupe <strong>GC</strong>S peuvent avoir des implications directes en matière de prévention, dans la mesure où ils apportent une analyse plus précise des données de criblage mutationnel dans des gènes de prédisposition conférant un risque élevé de cancer, tels que BRCA1, BRCA2, MLH1 et MSH2. Le Groupe <strong>GC</strong>S assure également des services de génotypage aux deux Groupes, grâce à sa plate-forme génétique. section génétique 93
